Inhibition of gluconeogenesis by extracellular ATP in isolated rat hepatocytes.

1991

The aim of this study was to determine the effect of externally added ATP on gluconeogenesis by isolated hepatocytes from starved rats. High concentrations of extracellular ATP inhibited gluconeogenesis from lactate and pyruvate but not from glycerol or fructose. This inhibition was associated with an increase in intracellular adenosine contents. ADP, AMP, or adenosine but not guanosine 5'triphosphate, inosine 5' triphosphate, or adenine also inhibited gluconeogenesis. alpha, beta-Methylene-ATP, a nonmetabolizable structural analogue of ATP, did not affect the rate of gluconeogenesis. Intracellular ATP levels were increased by externally added ATP or adenosine, but ATP-to-ADP ratios in the…

A tectonic inversion by steps during the Cenozoic : the Dover Strait

2004

Abstract The Boulonnais is a dome incised by a former marine gulf inset into a zone of tectonic inversion from the Middle Eocene, which was already partly excavated at least at the Upper Eocene. New sedimentological and paleopedological data obtained within the Boulonnais, completed with old seismic profiles, allow a better understanding of the inversion process which developed step by step. The initial breaching probably took place in the late Eocene. The Dover Strait was probably opened during the Lutetian, a part of the Oligocene and the late Neogene. Oligocene and Pliocene faunal assemblages are identical on both sides of the Strait. It was closed again for tectonic and eustatic reasons…

New insular taxa from the oldest Terre Rosse fissure filling (Gargano, Southeastern Italy).

2012

A rich amount of fossil remains of a highly diversified vertebrate fauna, known as “Mikrotia fauna”, has been retrieved from the red soil deposits (Terre Rosse) which fill the extensive palaeokarst network that affects the Mesozoic limestone along the north-western slopes of Mount Gargano (Southern Italy). The faunal assemblages reveal a rather complex history of bioevents such as dispersals and extinctions, which occurred when the area was isolated. These reconstructions were based on the materials collected during the seventies and the eighties of the last century. Forty years after its discovery, the Gargano Terre Rosse finally yielded evidence of an older faunal settlement. The peculiar…

The desert gerbil Psammomys obesus as a model for metformin-sensitive nutritional type 2 diabetes to protect hepatocellular metabolic damage: Impact …

2017

Introduction While metformin (MET) is the most widely prescribed antidiabetic drug worldwide, its beneficial effects in Psammomys obesus (P. obesus), a rodent model that mimics most of the metabolic features of human diabetes, have not been explored thoroughly. Here, we sought to investigate whether MET might improve insulin sensitivity, glucose homeostasis, lipid profile as well as cellular redox and energy balance in P. obesus maintained on a high energy diet (HED). Materials and methods P. obesus gerbils were randomly assigned to receive either a natural diet (ND) consisting of halophytic plants (control group) or a HED (diabetic group) for a period of 24 weeks. MET (50 mg/kg per os) was…

Neurogenin 3+ cells contribute to β-cell neogenesis and proliferation in injured adult mouse pancreas

2013

Abstract: We previously showed that injury by partial duct ligation (PDL) in adult mouse pancreas activates Neurogenin 3 (Ngn3)(+) progenitor cells that can differentiate to beta cells ex vivo. Here we evaluate the role of Ngn3(+) cells in beta cell expansion in situ. PDL not only induced doubling of the beta cell volume but also increased the total number of islets. beta cells proliferated without extended delay (the so-called 'refractory' period), their proliferation potential was highest in small islets, and 86% of the beta cell expansion was attributable to proliferation of pre-existing beta cells. At sufficiently high Ngn3 expression level, upto 14% of all beta cells and 40% of small i…

Effect of metal ion catalyzed oxidation of rifamycin SV on cell viability and metabolic performance of isolated rat hepatocytes.

1991

The effect of rifamycin SV on metabolic performance and cell viability was studied using isolated hepatocytes from fed, starved and glutathione (GSH) depleted rats. The relationships between GSH depletion, nutritional status of the cells, glucose metabolism, lactate dehydrogenase (LDH) leakage and malondialdehyde (MDA) production in the presence of rifamycin SV and transition metal ions was investigated. Glucose metabolism was impaired in isolated hepatocytes from both fed and starved animals, the effect is dependent on the rifamycin SV concentration and is enhanced by copper (II). Oxygen consumption by isolated hepatocytes from starved rats was also increased by copper (II) and a partial i…

Dependence of hepatic gluconeogenesis on PO2: inhibitory effects of halothane

1987

The dependence of gluconeogenesis and O2 uptake on PO2 in isolated rat hepatocytes is presented. Maintenance of steady-state PO2 was achieved with an oxystat system (Biochem. J. 236: 765–769, 1986). O2 uptake showed a half-maximal (K0.5) value of 0.5 Torr PO2, whereas the glucose synthesis rate was half-maximal at 1.2 Torr PO2. Halothane at concentrations greater than 1 mM exerted a parallel inhibition of O2 uptake and glucose synthesis at all PO2 levels studied. In contrast, at halothane concentrations less than 1 mM, inhibition of glucose synthesis occurred only at less than 20 Torr PO2. At these low concentrations, halothane was without significant effects on cellular O2 uptake. In isol…

Ages and stratigraphical architecture of late Miocene deposits in the Lorca Basin (Betics, SE Spain): New insights for the salinity crisis in margina…

2020

International audience; Unlike most Neogene basins of the Betic Cordillera where the Salinity Crisis is dated to the Messinian, a contradictory Tortonian dating was proposed for evaporites of the Lorca Basin. As a consequence, complex structural models have been proposed in the literature to explain this discrepancy in the timing of evaporites. In order to integrate the Lorca Basin into the geological context of the western Mediterranean domain during the Late Miocene, new sedimentological and stratigraphical studies coupled with new dating were performed, which allow us to propose a Messinian age for both diatomite-bearing deposits and evaporites of the Lorca Basin. These new ages challeng…

The onset of the Messinian salinity crisis: Insights from Cyprus sections

2009

Abstract The Neogene basins of Southern Cyprus provide a good opportunity to improve the knowledge of the paleoenvironmental changes involved in the triggering of the Messinian evaporite deposition in the Mediterranean, and of their chronology, which is still questionable with regards to the parameters responsible for the triggering of the salinity crisis. It is still difficult to discriminate the individual effects of tectonics, climate, global sea-level changes. In Cyprus, considerable progress has been made on the events leading to the MSC, since the 70's, in high-resolution microfossil biostratigraphy, astrochronology, cyclostratigraphy and magnetostratigraphy. A study of the Tochni sec…
