Activating mutations in the FLT3 tyrosine kinase (TK) occur in approximately 35% of patients with acute myeloid leukemia (AML). Therefore, targeting mutated FLT3 is an attractive therapeutic strategy, and early clinical trials testing FLT3 TK inhibitors (TKI) showed measurable clinical responses. Most of these responses were transient; however, in a subset of patients blast recurrence was preceded by an interval of prolonged remission. The etiology of clinical resistance to FLT3-TKI in AML is unclear but is of major significance for the development of future therapeutic strategies. This interview with Micheál Ó Nualláin, Brian O’Nolan’s brother and the only surviving member of his generation of the family, was conducted on 10 June 2014 in Monkstown, County Dublin. Its purpose was to obtain information about Brian O’Nolan’s private library at the John J. Burns Library, Boston College; the archive being the focal point of this issue of The Parish Review. It was Micheál who sold the last of his brother’s effects to Boston College in May 1997 and therefore he is in a position to answer questions about the purchase of the library, its contents and their provenance, and the reading practices of Brian and Evelyn O’Nolan.To read the article, click Download or View PDF.

NPM1mutant (NPM1mut) and MLL1-rearranged (MLL-r) acute myeloid leukemias (AMLs) exhibit aberrant expression of HOX and MEIS1 transcription factors and commonly harbor an activating mutation in the receptor tyrosine kinase FLT3. Pharmacologic inhibition of the menin-MLL1 complex reverses leukemogenic gene expression including MEIS1 and FLT3 and represents a therapeutic opportunity for the treatment of these leukemias. Bakalaura darba mērķis ir noskaidrot kā notiek kinematogrāfiskā maldināšana Kristofera Nolana filmās, kā arī noteikt kādi kino valodas elementi tiek izmantoti, lai radītu naratīva metalepsi. Darbā tika aplūkoti laika secības, maldināšanas, naratīva un semiotikas teorētiskie aspekti, kā arī tika veikta naratīva un semiotiskā analīze. Veiktā pētījuma rezultāti parādīja, ka maldināšanu veicina filmu naratīva struktūra, informācijas slēpšana, personāži-viltotāji un laika secības izmaiņas. Structural characterization of 2,2-di-n-butyl-4-methyl-1,3,2-dioxastannolane isolated from supercritical CO2 conditions

The title compound has been isolated as single crystals from the synthesis of propylene carbonate from racemic 1,2-propanediol and carbon dioxide using n-Bu2SnO as a catalyst precursor. The X-ray crystallographic structure analysis revealed the self-assembly of di-n-butyltin(IV) 1,2-propanediolate units, linked together through long-distance Sn-O interactions leading to a one-dimensional polymeric architecture organized in a syndiotactic arrangement. The coordination geometry around the tin atoms can be described as an unusual faced-capped trigonal bipyramidal environment.
FLT3 as a therapeutic target in AML: still challenging after all these years

Abstract Mutations within the FMS-like tyrosine kinase 3 (FLT3) gene on chromosome 13q12 have been detected in up to 35% of acute myeloid leukemia (AML) patients and represent one of the most frequently identified genetic alterations in AML. Over the last years, FLT3 has emerged as a promising molecular target in therapy of AML. Here, we review results of clinical trials and of correlative laboratory studies using small molecule FLT3 tyrosine kinase inhibitors (TKIs) in AML patients. We also review mechanisms of primary and secondary drug resistance to FLT3-TKI, and from the data currently available we summarize lessons learned from FLT3-TKI monotherapy.
