Bile acid–cysteamine conjugates: Structural properties, gelation, and toxicity evaluation

Abstract Design, synthesis, and characterization of six novel bile acid–cysteamine conjugates together with investigation of their structural studies, gelation properties, and preliminary toxicity evaluation, are reported. Solid state properties of selected compounds were studied by means of X-ray diffraction and 13C CPMAS NMR spectroscopy. N-(2-thioethyl)-3α,7α,12α-trihydroxy-5β-cholan-24-amide was shown to exhibit (pseudo)polymorphism, and a single crystal structure of its non-stoichiometric hydrate is reported herein. Twelve N-glycosyl amines were synthesised using 4,6-O–benzylidene-D-glucopyranose and different substituted aromatic amines, including some diamines that resulted in bis-glycosyl amines. Another set of six N–glycosyl amines was synthesised using different hexoses and pentoses and 2–(o–aminophenyl)benzimidazole. All compounds were isolated as solid products and purified, their elemental compositions were established, and these were characterised by NMR (1H and 13C), UV–Vis, and FTIR spectroscopy, by FAB mass spectrometry (molecular-ion peaks gave molecular weights), and by their optical rotations. Chemical and spectroscopical properties of cis- and trans-cyclopropano-anellated cycloalkynes of medium ring size are investigated and compared with results from force field calculations. IR and 13C-NMR data of the alkynes reflect the deformation of the triple bond due to ring strain. Parallel to bending the reactivity increases with decreasing ring size, the trans fused alkynes prove to be nearly as reactive as the cis fused with a two carbon smaller ring size. The bicycloalkynes are complexed with silver triflate. 13C NMR reveals that the triple bond as well as the cyclopropane serve as ligands.

Abstract Cycloaddition reactions between 3-vinylthiophen and the dienophiles maleic anhydride, methyl acrylate, dimethyl acetylene - dicarboxylate, and methyl propiolate give products including methyl benzo[b]thiophen-7-carboxylate (11) and its dihydroderivative (10), the 6,7-dicarboxylic acid (3), its dimethyl ester (7) and dihydroester (6); the naphtho[a,b]dithiophen (14), and the novel ethano bridged benzo[b]thiophen (15).

Abstract The reaction of ortho-lithiated N,N-dimethyl-1-arylethyl-amines with triphenyl- boron leads, with elimination of phenyllithium, to (R)-(−)-2,2,3-trimethyl-1,1-diphenyl -1H-2-azonia-1-borata-indene (1), 2,3-dihydro-1,1,2-trimethyl-3,3-diphenyl- 1H-2-azonia-3-borata-benz[e]indene (2) and 2,3-dihydro-2,2,3-trimethyl-1,1-diphenyl -1H-2-azonia-1-borata-benz[e]indene (3) with a strongB←N bond. 1, 2 and 3 are stable against oxygen, water and alcohols. Triarylboron compounds sensitive to the same reagents are formed by the reaction of triphenylboron with some aryllithium compounds substituted with electron-donating groups.
Diorganotin(IV) N-acetyl-L-cysteinate complexes: synthesis, solid state, solution phase, DFT and biological investigations.

Diorganotin(IV) complexes of N-acetyl-L-cysteine (H(2)NAC; (R)-2-acetamido-3-sulfanylpropanoic acid) have been synthesized and their solid and solution-phase structural configurations investigated by FTIR, Mössbauer, (1)H, (13)C and (119)Sn NMR spectroscopy. FTIR results suggested that in R(2)Sn(IV)NAC (R = Me, Bu, Ph) complexes NAC(2-) behaves as dianionic tridentate ligand coordinating the tin(IV) atom, through ester-type carboxylate, acetate carbonyl oxygen atom and the deprotonated thiolate group. From (119)Sn Mössbauer spectroscopy it could be inferred that the tin atom is pentacoordinated, with equatorial R(2)Sn(IV) trigonal bipyramidal configuration. In DMSO-d(6) solution, NMR spectr…

International audience; : Transverse and longitudinal relaxation times (T1ρ and T1 ) have been widely exploited in NMR to probe the binding of ligands and putative drugs to target proteins. We have shown recently that long-lived states (LLS) can be more sensitive to ligand binding. LLS can be excited if the ligand comprises at least two coupled spins. Herein we broaden the scope of ligand screening by LLS to arbitrary ligands by covalent attachment of a functional group, which comprises a pair of coupled protons that are isolated from neighboring magnetic nuclei. A new sulfated lupane triterpene, Gypsophilin (1), and its glucosyl ester, Gypsophilinoside (2) were isolated from the roots of Gypsophila repens whereas a new flavone C-glycoside (3) was obtained from the aerial parts. Their structures were established as (3beta)-3-O-(sulfo)lup-20(29)-en-23,28-dioic acid (1), (3beta)-3-O-(sulfo)lup-20(29)-en-23,28-dioic acid -28-O-beta-D-glucopyranosyl ester (2) and luteolin-7-O-alpha-L-arabinopyranosyl-6-C-beta-glucopyranoside (3) by spectroscopic methods such as 1D and 2D NMR, HR-ESI-MS and FAB-MS.
