Determination of Aflatoxins and Ochratoxins in Sicilian Sweet Wines by High-Performance Liquid Chromatography with Fluorometric Detection and Immunoa…

2015

Ochratoxin A is the only mycotoxins with legal limits in the European Union for wine, by contrast, there is no EU legislation imposed for Aflatoxins and Ochratoxin B. A reproducible and sensitive analytical method for six mycotoxins: Ochratoxin A, Ochratoxin B, Aflatoxins B1, Aflatoxin B2, Aflatoxin G1 and Aflatoxin G2 was developed by using immunoaffinity column cleanup and HPLC-FLD method in 30 sweet wines produced in Sicily. This typical type of wine is manufactured from grapes grown in warmer climates, which have a higher sugar content; also, the over-ripening and the drying process of the grapes before fermentation conditions are responsible for the particular susceptibility of these b…

Mycotoxins and mycotoxigenic moulds in nuts and sunflower seeds for human consumption

1991

A survey was carried out to obtain data on the occurrence of mycotoxins and the mycotoxin-producing potential of fungi isolated from nuts (almonds, peanuts, hazelnuts, pistachio nuts) and sunflower seeds in Spain. Thin-layer chromatography was used to separate the toxins. Aflatoxins were detected in one sample of almonds (95 ppb aflatoxin B1 and 15 ppb aflaxtoxin B2) and in one sample of peanuts at a level below 10 ppb of aflatoxin B1. 100% of samples showed variable incidence of fungal contamination. The predominant fungi present in samples were Penicillium spp, Aspergillus niger, A. flavus, A. glaucus and Rhizopus spp. The results showed that isolates of different species were able to pro…

Predominant mycotoxins, mycotoxigenic fungi and climate change related to wine

2017

Wine is a significant contributor to the economies of many countries. However, the commodity can become contaminated with mycotoxins produced by certain fungi. Most information on mycotoxins in wine is from Spain, Italy and France. Grapes can be infected by mycotoxigenic fungi, of which Aspergillus carbonarius producing ochratoxin A (OTA) is of highest concern. Climate is the most important factor in determining contamination once the fungi are established, with high temperatures being a major factor for OTA contamination: OTA in wine is at higher concentrations in warmer southern Europe than northern. Contamination by fumonisins is a particular concern, related to Aspergillus niger produci…

Determination of mycotoxins in fruit berry by-products using QuEChERS extraction method

2017

Abstract Fruit berries by-products are mainly elaborated with berry but most of them mixed with other fruits, grapes, plums or apples. These present favourable properties for the growth of a great range of mycotoxins mould producers. Consequently, alternariol (AOH), alternariol monomethyl ether (AME), tentoxin (TEN), aflatoxins (AFB1, AFB2, AFG1, AFG2) and ochratoxin A (OTA) are the main mycotoxins that should be present in berries by-products. To establish a reliable analytical method of these mycotoxins on two fruit berries by-products (jam and juice), four different QuEChERS extraction method were evaluated. Recoveries obtained were higher than 74% and 76% in berries juice and jam berrie…

Potential impact of engineered silver nanoparticles in the control of aflatoxins, ochratoxin A and the main aflatoxigenic and ochratoxigenic species …

2019

Abstract The potential use of nanotechnology in the control of toxigenic fungi and mycotoxin production has been little explored. In this report, engineered silver nanoparticles (AgNPs) were synthesized and characterized by single particle Inductively Coupled Plasma Mass Spectrometry. Then, their effectiveness in the control of the growth of the main aflatoxigenic and ochratoxigenic species affecting foods and aflatoxins (AFs) and ochratoxin A (OTA) production was studied. The target species and their associated mycotoxins were Aspergillus flavus (AFB1 and AFB2), A. parasiticus (AFB1, AFB2, AFG1 and AFG2), A. carbonarius, A. niger, A. ochraceus, A. steynii, A. westerdijkiae and Penicillium …

Effect of Gamma Irradiation on Aflatoxins and Ochratoxin A Reduction in Almond Samples

2014

<p>The widespread contamination of food by mycotoxins may present a serious hazard to human and animal health. The gamma rays were applied to reduce ochratoxin A (OTA) and aflatoxins B1, B2, G1 and G2 (AFB1, AFB2, AFG1 and AFG2) in almonds artificially contaminated. In the present study we investigated the effect of gamma irradiation dosages, ranging from 0 to 15 kGy and the reduction of mycotoxins concentration in almond samples. In order to determine the efficiency of the method, a high-performance liq…

Microbiological and safety evaluation of green table olives marketed in Italy

2011

The microbiological and safety conditions of green table olives sold on the Italian market were evaluated on 40 samples, 20 loose and 20 packed in containers, purchased at street markets and supermarkets. The olives were analyzed for microflora and food safety indices, and for aflatoxin B1 and ochratoxin A occurrence, and the results showed acceptable security. There was wide heterogeneity in the microflora, the numerical values being in relation to the olive type. The microbial population was dominated by the yeasts and lactic bacteria responsible for the fermentation process, and their numbers, together with the metabolic activity, led to conditions unfavorable for the development of path…

Novel haptens and monoclonal antibodies with subnanomolar affinity for a classical analytical target, ochratoxin A

2018

Ochratoxin A is a potent toxic fungal metabolite whose undesirable presence in food commodities constitutes a problem of public health, so it is strictly regulated and controlled. For the first time, two derivatives of ochratoxin A (OTAb and OTAd) functionalized through positions other than the native carboxyl group of the mycotoxin, have been synthesized in order to better mimic, during the immunization process, the steric and conformational properties of the target analyte. Additionally, two conventional haptens making use of that native carboxyl group for protein coupling (OTAe and OTAf) were also prepared as controls for the purpose of comparison. The immunological performance in rabbit…

Risk management of ochratoxigenic fungi and ochratoxin A in maize grains by bioactive EVOH films containing individual components of some essential o…

2018

Abstract Aspergillus steynii and Aspergillus tubingensis are possibly the main ochratoxin A (OTA) producing species in Aspergillus section Circumdati and section Nigri, respectively. OTA is a potent nephrotoxic, teratogenic, embryotoxic, genotoxic, neurotoxic, carcinogenic and immunosuppressive compound being cereals the first source of OTA in the diet. In this study bioactive ethylene-vinyl alcohol copolymer (EVOH) films containing cinnamaldehyde (CINHO), linalool (LIN), isoeugenol (IEG) or citral (CIT) which are major components of some plant essential oils (EOs) were produced and tested against A. steynii and A. tubingensis growth and OTA production in partly milled maize grains. Due to …

Influence of different coffee drinks preparations on ochratoxin A content and evaluation of the antioxidant activity and caffeine variations.

2011

Coffee is one of the most frequently consumed beverages in North America and Europe. It is well known that coffee contains caffeine and that coffee beans can be contaminated by Ochratoxin A (OTA). The operating conditions however affect OTA and caffeine extraction from the roasted coffee. OTA content found in the beverages can be greater than that found in the roasted coffee used to prepare it, representing a potential OTA related risk factor for the human health. Moreover the coffee beans and coffee based beverages have an anti oxidant activity. This study investigates the OTA content, the amount of caffeine, and the antioxidant activity in five different preparations: American coffee, Mok…
