A Multicenter Phase I/II Trial of the Combination of Imatinib Mesylate with Mitoxantrone/Etoposide and Cytarabine in Patients with CML in Myeloid Bla…

2004

Abstract The combination of imatinib with mitoxantrone, etoposide or cytarabine were shown to be additive to highly synergistic on BCR-ABL-positive leukemias in vitro by several investigators, including our group. Therefore, we initiated a phase I/II trial for patients with myeloid blast crisis of chronic myelogenous leukemia. Patients were treated in four cohorts starting from mitoxantrone 10 mg/m2/d and etoposide 100 mg/m2/d for 2 or 3 consecutive days and start of imatinib 600 mg/d from day 15 (cohorts #1 and #2) or from day 1 (cohorts #3 and #4). Cytarabine was given at a dose of 10 mg/m2/d s.c. as maintenance treatment. Seventeen patients were included in the study: 8 pts in cohort 1, …

Preliminary Phase II Study Results of BBR2778 in Combination with Cyclophosphamide, Vincristine, and Prednisone in Patients with Relapsed Aggressive …

2004

Abstract Background: Pixantrone (BBR 2778) is a novel aza-anthracenedione with superior activity compared to doxorubicin and mitoxantrone in various tumor models including hematological malignancies. Pixantrone single agent therapy led to major responses in patients (pts) with aggressive lymphoma including diffuse large B-cell lymphoma (DLCL). The safety of a CHOP-like regimen with pixantrone replacing doxorubicin (CPOP) was assessed in a dose-ranging study indicating a recommended dose of 150 mg/m² for pixantrone. Method: In this phase II study the primary objective was to assess the efficacy of the CPOP regimen (cyclophosphamide 750 mg/m² d1, pixantrone 150mg/m² d1, vincristine 1.4mg/m² d…

Chronical haloperidol and clozapine treatment in rats: Differential RNA display analysis, behavioral studies and serum level determination

1998

1. Adult, female rats were treated orally for 23 days with 1.6 mg/kg haloperidol or 36 mg/kg clozapine per day, to study chronic effects of the two neuroleptics. 2. At five time points during the neuroleptic treatment, animal behavior was recorded in an open field and locomotive activity was analysed. At the end of the experiment, rats were decapitated, blood samples were collected and serum concentrations of haloperidol and clozapine were determined by a radioreceptor or HPLC assay, respectively. RNA was isolated from each brain, without cerebellum, and subjected to differential RNA display. 3. Mean serum concentrations were 8 ng/ml for haloperidol and 21 ng/ml for clozapine. Analysis of o…

Diagnosis of Heparin-Induced Thrombocytopenia: Development and Validation of a Predictive Clinical Score Based on Objective Features Identified by a …

2019

Background: Diagnosis of heparin-induced thrombocytopenia (HIT) is based on a composite of clinical likelihood and laboratory testing. We aimed to develop a diagnostic score derived from multivariate analysis of clinical features, including platelet count changes, prospectively recorded in patients with suspected HIT. Methods: This multinational observational study (ClinicalTrials.gov no. NCT00748839) included 2280 adult patients with suspected HIT: 1597 (derivation cohort) formed the basis for developing the scoring system, subsequently validated in 683 additional randomly selected patients (validation cohort). HIT was diagnosed by two independent adjudicators based on clinical features, l…

Sarcopenia and Covid-19: A New Entity?

2021

As the world comes into terms with the persistent existence of the coronavirus disease 19 (COVID-19) pandemic, the scientific and medical communities have started shifting their attention to the long-term side effects of the disease. Specialists have acknowledged that numerous COVID-19 survivors, especially those who suffered severe complications and endured prolonged bedrest, may require continuous monitoring and rehabilitative care. In this chapter, the authors discuss factors that may predispose individuals to acute sarcopenia secondary to COVID-19 infection or sarcopenic obesity secondary to altered diet and physical activity from prolonged confinement. Challenges in nutrition are highl…

Engraftment kinetics of human CD34+ cells from cord blood and mobilized peripheral blood co-transplanted into NOD/SCID mice

2004

We have reported short periods of post transplant neutropenia in human patients co-transplanted with cord blood (CB) and low numbers of haploidentical mobilized peripheral blood (MPB) CD34+ cells. To investigate the effect that the proportion of MPB to CB cells may have on engraftment kinetics, we have co-transplanted fixed numbers of human CB CD34+ cells mixed with different numbers of MPB CD34+ cells into NOD/SCID mice. We periodically quantified the proportion of human cells and the relative contribution of MPB and CB cells to the human engraftment on marrow aspirates. At the lowest MPB/CB ratios (5 : 1, 10 : 1), the contribution of CB cells predominated at all time points analyzed, and …

Magnesium and muscle performance in older persons: the InCHIANTI study1–3

2006

BACKGROUND The role of magnesium in maintaining muscle integrity and function in older adults is largely unknown. OBJECTIVE We aimed to investigate the relation between serum magnesium concentrations and muscle performance in older subjects. DESIGN Data are from the baseline examination conducted between September 1998 and March 2000 of the InCHIANTI (aging in the Chianti area) study, a prospective epidemiologic survey of risk factors for late-life disability. From among 1453 randomly selected community residents completing a home interview, 1138 men (46%) and women (aged 66.7 +/- 15.2 y; x +/- SD) with complete data on muscle performance and serum magnesium who were not severely cognitivel…

Nutrition and physical activity in the prevention and treatment of sarcopenia: systematic review

2017

Summary This systematic review summarizes the effect of\ud combined exercise and nutrition intervention on muscle mass\ud and muscle function. A total of 37 RCTs were identified.\ud Results indicate that physical exercise has a positive impact\ud on muscle mass and muscle function in subjects aged 65 years\ud and older. However, any interactive effect of dietary supplementation\ud appears to be limited.\ud Introduction In 2013, Denison et al. conducted a systematic\ud review including 17 randomized controlled trials\ud (RCTs) to explore the effect of combined exercise and\ud nutrition intervention to improve muscle mass, muscle\ud strength, or physical performance in older people.\ud They c…

Sleep apnea and its role in transportation safety

2017

Obstructive sleep apnea (OSA) is a main cause of excessive daytime sleepiness and increases the risk for driving accidents, which can be normalized by treatment with continuous positive airway pressure ventilation. Since it is estimated that OSA is not diagnosed in about 80% of cases, recognition of patients at risk for driving accidents is a problem from both medical and societal points of view. Strategies to screen and identify subjects at high risk for driving accidents are under study in order to improve safety on the road, especially for commercial drivers, who show a high prevalence of OSA.

Changes in ocular signs and symptoms when switching from preserved latanoprost 0.005% to preservative-free tafluprost 0.0015%: phase IIIb study in pa…

2008

Purpose This open-label, multinational, phase IIIb study investigated the changes in ocular signs and symptoms when patients were switched from preserved latanoprost 0.005% to preservative-free tafluprost 0.0015% (q.d. at 20:00) for 12 weeks. Methods Patients with open-angle glaucoma or ocular hypertension who had received latanoprost for at least 6 months and had at least 2 ocular symptoms or one symptom and one sign were enrolled (n=158). Change from screening in ocular symptoms and signs were the main endpoints. Data from the 6-week analysis are reported here. Results After switching to preservative-free tafluprost, the proportion of patients with at least mild symptoms decreased: irrita…
