Citrus rootstock breeding: response of four allotetraploid somatic hybrids to Citrus tristeza virus induced infections
2018
Four allotetraploid somatic hybrids of citrus, with potential for rootstock improvement, have been evaluated for their response to Citrus tristeza virus (CTV) infection. CTV is the most important viral pathogen affecting citrus production worldwide. Somatic combinations of ‘Milam’ lemon (Citrus jambhiri Lush.) + Sour orange (C. aurantium L Osb.), Calamondin (C. madurensis Lour.) + ‘Keen’ sour orange (C. aurantium L.), Calamondin + ‘Femminello‘ lemon (C. limon L. Burm. F.) and Cleopatra mandarin (C. reshni Hort. ex Tan.) + ‘Femminello’ lemon, were studied. Plants were grafted with CTV-infected “Valencia” sweet orange budwood. Two different CTV strains collected in Sicily, considered as “mild…
Exploring species-level taxonomy in the Cryptocephalus flavipes species complex (Coleoptera: Chrysomelidae)
2016
In insects, morphological species identification is often challenging. The discrimination of closely related species may be hampered when only subtle differences in phenotypic characters or a continuum in their variability are present. This is exemplified in the Cryptocephalus flavipes species complex (Coleoptera; Chrysomelidae) where, until now, the species have been discriminated only by the yellow pattern on frons, pronotum, and epipleurae. In the present study, the phylogeny of the C. flavipes species complex was resolved through a multi-locus sequence approach, and the inclusion in the group of the phenotypically similar Cryptocephalus quadripustulatus Gyllenhal, 1813 was evaluated. Su…
Thioredoxin (Trxo1) interacts with proliferating cell nuclear antigen (PCNA) and its overexpression affects the growth of tobacco cell culture.
2017
Thioredoxins (Trxs), key components of cellular redox regulation, act by controlling the redox status of many target proteins, and have been shown to play an essential role in cell survival and growth. The presence of a Trx system in the nucleus has received little attention in plants, and the nuclear targets of plant Trxs have not been conclusively identified. Thus, very little is known about the function of Trxs in this cellular compartment. Previously, we studied the intracellular localization of PsTrxo1 and confirmed its presence in mitochondria and, interestingly, in the nucleus under standard growth conditions. In investigating the nuclear function of PsTrxo1 we identified proliferati…
A comparative study of viral infectivity, accumulation and symptoms induced by broad bean wilt virus 1 isolates
2018
Broad bean wilt virus 1 (BBWV-1, genus Fabavirus, family Secoviridae) is a bipartite positive-sense single-stranded RNA virus distributed worldwide infecting many herbaceous species. Until now, scarce information regarding biological properties of BBWV-1 isolates is available. This work shows a comparative study on virus infectivity (proportion of infected plants over inoculated plants), virus accumulation and symptoms induced by four genetically different BBWV-1 isolates (Ben, B41/99, NSRV and PV0548) which were mechanically inoculated on several herbaceous hosts. The four BBWV-1 isolates infected broad bean, tomato, pepper and Nicotiana benthamiana plants, whereas none of them infected cu…
Phylogenomics Identifies an Ancestral Burst of Gene Duplications Predating the Diversification of Aphidomorpha
2019
Aphids (Aphidoidea) are a diverse group of hemipteran insects that feed on plant phloem sap. A common finding in studies of aphid genomes is the presence of a large number of duplicated genes. However, when these duplications occurred remains unclear, partly due to the high relatedness of sequenced species. To better understand the origin of aphid duplications we sequenced and assembled the genome of Cinara cedri, an early branching lineage (Lachninae) of the Aphididae family. We performed a phylogenomic comparison of this genome with 20 other sequenced genomes, including the available genomes of five other aphids, along with the transcriptomes of two species belonging to Adelgidae (a close…
DNA-Based Identification of Larvae Offers Insights into the Elusive Lifestyles of Native Olive Seed Wasps in South Africa
2020
Wild and cultivated olives in the Western Cape of South Africa are direct or indirect hosts to a high diversity of Braconidae and Chalcidoidea wasps. Olive-associated Braconidae are known to parasitise the larvae of Bactrocera oleae (Rossi), and probably also Bactrocera biguttula (Bezzi). The lifestyle of olive-associated Chalcidoidea is not fully understood, and may include phytophagous, parasitoid and hyperparasitoid species. Some chalcids could represent olive seed wasps (OSW), a generic term that designates the seed feeders responsible for losses in commercial olive production. In order to obtain direct DNA-based evidence for the lifestyle of four putative OSW – Eupelmus spermophilus Si…
Differential Effect of Plant Lipids on Membrane Organization
2015
SPE IPM; International audience; The high diversity of the plant lipid mixture raises the question of their respective involvement in the definition of membrane organization. This is particularly the case for plant plasma membrane, which is enriched in specific lipids, such as free and conjugated forms of phytosterols and typical phytosphingolipids, such as glycosylinositolphosphoceramides. This question was here addressed extensively by characterizing the order level of membrane from vesicles prepared using various plant lipid mixtures and labeled with an environment-sensitive probe. Fluorescence spectroscopy experiments showed that among major phytosterols, campesterol exhibits a stronger…
Fig virus-free production and survival rate improvement using meristem tip culture techinique associated with the encapsulation technology
2018
Three Mediterranean F. carica genotypes, i.e. cultivars Palazzo, Bifera nera and Catalanisca, initially infected by Fig leaf mottle-associated virus 1 (FLMaV-1), Fig leaf mottle-associated virus 2 (FLMaV-2), Fig mild mottling-associated virus (FMMaV), Fig mosaic virus (FMV), Fig latent virus 1 (FLV-1), Fig Badnavirus 1 (FBV-1) and Fig fleck-associated virus (FFkaV), were subjected to the sanitation technique via Meristem Tip (0.3-0.5 mm in size) Culture (MTC), also associated with the encapsulation technique (MTC-SS), in order to produce virus-free plant material. Encapsulation was tested to overcome the very low survival and regeneration rates, due to the small propagule size. Encouraging …
Biotrickling filter modeling for styrene abatement. Part 1: Model development, calibration and validation on an industrial scale
2017
Abstract A three-phase dynamic mathematical model based on mass balances describing the main processes in biotrickling filtration: convection, mass transfer, diffusion, and biodegradation was calibrated and validated for the simulation of an industrial styrene-degrading biotrickling filter. The model considered the key features of the industrial operation of biotrickling filters: variable conditions of loading and intermittent irrigation. These features were included in the model switching from the mathematical description of periods with and without irrigation. Model equations were based on the mass balances describing the main processes in biotrickling filtration: convection, mass transfe…
The cost of open access publication of papers by spanish authors in five areas of the social sciences
2021
