Interpatient variability in drug response can be widely explained by genetically determined differences in metabolizing enzymes, drug transporters, and drug targets, leading to different pharmacokinetic and/or pharmacodynamic behaviors of drugs. Genetic variations affect or do not affect drug responses depending on their influence on protein activity and the relevance of such proteins in the pathway of the drug. Previous studies have suggested that polymorphisms in CASP8 on chromosome 2 are associated with breast cancer risk. To clarify the role of CASP8 in breast cancer susceptibility, we carried out dense genotyping of this region in the Breast Cancer Association Consortium (BCAC). Single-nucleotide polymorphisms (SNPs) spanning a 1 Mb region around CASP8 were genotyped in 46 450 breast cancer cases and 42 600 controls of European origin from 41 studies participating in the BCAC as part of a custom genotyping array experiment (iCOGS). Sarcopenia is a common condition in older people. Increasing literature is showing its importance for negative health outcomes, also beyond the perimeter of geriatric medicine. In this chapter, we will discuss the consequences of sarcopenia. Overall, epidemiological research suggests that sarcopenia is significantly associated with falls and fractures also suggesting a strong association between muscle and bone loss conditions, as in the case of osteosarcopenia. Moreover, sarcopenia is associated with disability, hospitalization (particularly in older people already hospitalized), and finally mortality. Interaction between clients and physiotherapists in group exercise classes in geriatric rehabilitation

The aim of this paper is to explore how older people construct their interaction in group exercise classes in geriatric rehabilitation and what is their contribution to the interaction. Discourse analysis was employed and data, consisting of seven videotaped group-based exercise sessions, were collected from 52 older people (aged 66–93 years) and nine rehabilitation professionals in seven rehabilitation centres.
