Fusarium proliferatum: induction and intracellular location of a lipoxygenase

The fungus Fusarium proliferatum was grown in a soya oil- and glucose-supplemented medium. Induction of lipoxygenase activity (13-fold) in an extract of Fusarium biomass has been observed at day four on soya oil culture medium. A band at 232 kDa was detected using a specific lipoxygenase stain combined with native PAGE. The method of fungal homogenate obtained has been checked via subcellular marker enzymes activities: succinate dehydrogenase (SDH) for mitochondrial fraction and NADH-cytochrome C reductase (NADH-cyt. Celem przeprowadzonych badań była ocena właściwości przeciwgrzybowych ekstraktów z liści i korzenia pokrzywy zwyczajnej wobec Fusarium proliferatum. Ocenę właściwości antagonistycznych ekstraktów w stężeniach 2,5, 5,0, 10, 20 i 40% przeprowadzono metodą hodowlano-płytkową z zastosowaniem podłoży PDA pomidorowego. Hodowlę prowadzono w temp. 26ºC przez 9 dni, dokonując pomiarów co 1-2 dni. Kontrolę pozytywną stanowił Topsin M 500 S.C, a kontrolę negatywną woda destylowana. To produce enniatin (ENs), Fusarium tricinctum CECT 20150 was grown in a liquid medium of potato (PDB), being mycotoxin purified by high performance liquid chromatography (HPLC) with a reverse phase semipreparative column using a mobile phase of acetonitrile/water using gradient condition. Fumonisins are mycotoxins produced by Fusarium verticillioides that commonly contaminate maize and maize products. The present work shows the results of a comparative study of three different fermentation's techniques (solid and liquid medium of corn and a solid agarized medium) for the production of fumonisins B-1, B-2 and B-3 with strains of F. verticillioides. Occurrence of fumonisins B1 and B2 in Portuguese maize and maize-based foods intended for human consumption.

Fumonisin B(1) (FB(1)) and fumonisin B(2) (FB(2)) are mycotoxins mainly produced by Fusarium verticillioides and Fusarium proliferatum, which are field pathogens of maize. A survey was conducted on the incidences of FB(1) and FB(2) in both maize and derived products purchased in Portugal. The analytical method involved extraction with methanol-water, clean-up by immunoaffinity column and derivatization with naphthalene-2,3-dicarboxaldehyde.
