Risk-reducing hysterectomy and bilateral salpingo-oophorectomy in female heterozygotes of pathogenic mismatch repair variants: a Prospective Lynch Sy…
2021
Abstract Purpose To determine impact of risk-reducing hysterectomy and bilateral salpingo-oophorectomy (BSO) on gynecological cancer incidence and death in heterozygotes of pathogenic MMR ( path_MMR ) variants. Methods The Prospective Lynch Syndrome Database was used to investigate the effects of gynecological risk-reducing surgery (RRS) at different ages. Results Risk-reducing hysterectomy at 25 years of age prevents endometrial cancer before 50 years in 15%, 18%, 13%, and 0% of path_MLH1 , path_MSH2 , path_MSH6 , and path_PMS2 heterozygotes and death in 2%, 2%, 1%, and 0%, respectively. Risk-reducing BSO at 25 years of age prevents ovarian cancer before 50 years in 6%, 11%, 2%, and 0% and…
Tumor-derived immuno-modulators induce overlapping pro-tolerogenic gene expression signatures in human dendritic cells.
2016
Immature dendritic cells (iDCs) and tolerogenic DCs are essential for the induction and maintenance of peripheral tolerance. Tumors produce immuno-modulatory factors which imprint a pro-tolerogenic, maturation-resistant state in DCs. Here we asked for common markers of differentially tolerized human monocyte-derived DC populations. For this, PBMC-derived monocytes were differentiated to DCs in the presence of established immuno-modulators as released by tumors (IL-6, IL-10, TGF-β, glucocorticoid [GC], prostaglandin E2 [PGE2]). Most unstimulated pro-tolerogenic DC populations commonly over-expressed some tolerance-associated markers (ILT-4, IL-10, HO-1) as compared with iDCs. These markers m…
Full arch fixed prostheses vs. full arch telescopic-retained retrievable prostheses both supported by implants and natural tooth abutments in periodo…
2019
Background The clinical outcome of full arch fixed prostheses vs. full arch telescopic-retained retrievable prostheses supported by implants and natural tooth abutments in periodontally treated patients has been reported by few studies, with controversial results. The objective of this study was to evaluate long-term (15 years) complications of abutment teeth and dental implants in periodontally treated patients, rehabilitated with full arch telescopic-retained retrievable prostheses (TRP)s vs. full arch fixed prostheses (FP)s supported by teeth and implants. Material and methods After active periodontal therapy (non-surgical and surgical), and implant placement (replacement of hopeless tee…
A Standardized Wedelia chinensis Extract Overcomes the Feedback Activation of HER2/3 Signaling upon Androgen-Ablation in Prostate Cancer
2017
Crosstalk between the androgen receptor (AR) and other signaling pathways in prostate cancer (PCa) severely affects the therapeutic outcome of hormonal therapy. Although anti-androgen therapy prolongs overall survival in PCa patients, resistance rapidly develops and is often associated with increased AR expression and upregulation of the HER2/3-AKT signaling pathway. However, single agent therapy targeting AR, HER2/3 or AKT usually fails due to the reciprocal feedback loop. Previously, we reported that wedelolactone, apigenin, and luteolin are the active compounds in Wedelia chinensis herbal extract, and act synergistically to inhibit the AR activity in PCa. Here, we further demonstrated th…
THU0520 Assessment of cardiovascular risk in patients with fibromyalgia by carotid-femoral pulse wave velocity – results of a prospective study
2018
Background Autonomic dysfunction, a basic element of fibromyalgia (FM), has been in some cases related to increased risk of cardiovascular (CV) disease. CV risk associates with aortic stiffness, which can be reliably assessed by carotid-femoral pulse wave velocity (cfPWV). Objectives Aims of this study were to test the hypothesis of increased cfPWV in a group of patients with FM and to examine its association with FM associated parameters and selected traditional CV risk factors. Methods We performed measurements of cfPWV in 99 FM patients and 102 healthy controls. The difference between cfPWV values in the two groups after controlling for possible confounding factors was evaluated through …

Hyaluronic-based antibacterial hydrogel coating for implantable biomaterials in orthopedics and trauma. From basic research to clinical applications.
2018
Bacterial colonization of implanted biomaterials remains one of the most challenging complications in orthopedics and trauma surgery, with extremely high social and economic costs. Antibacterial coating of implants has been advocated by many experts as a possible solution to reduce the burden of implant-related infection and several different solutions have been proposed in the last decades. However, while most of the investigated technologies have shown their efficacy in vitro and/or in vivo, only few were able to reach the market, due to clinical, industrial, economic and regulatory issues. Hyaluronic acid composites have been previously shown to possess antifouling capabilities and have …
Influence of the metaphysis positioning in a new reverse shoulder prosthesis
2016
Aim of this work is to investigate the behaviour of a new reverse shoulder prosthesis, characterized by a humeral metaphysis with a variable offset, designed to increase the range of movements and to reduce the impingement. In particular, by means of virtual prototypes of the prosthesis, different offset values of the humeral metaphysis have been analysed in order to find the best positioning able to maximize the range of movements of the shoulder joint. The abduction force of the deltoid, at different offset values, has been also estimated. The study has been organized as follows. In the first step, the point clouds of the surfaces of the different components of the prosthesis have been ac…

Background: We prospectively tested technical feasibility and clinical outcome of percutaneous ultrasound-guided tenotomy of long head of biceps tendon (LHBT). Methods: We included 11 patients (6 women; age: 73 ± 8.6 years) with symptomatic full-thickness rotator cuff tear and intact LHBT, in whom surgical repair was not possible/refused. After ultrasound-guided injection of local anesthetic, the LHBT was cut with a scalpel under continuous ultrasound monitoring until it became no longer visible. Pain was recorded before and at least six months after procedure. An eight-item questionnaire was administered to patients at follow-up. Results: A median of 4 tendon cuts were needed to ensure com…
A prospective study of food preferences in childhood
2004
Abstract This study has evaluated the impact of food choices at 2–3 years old on food preferences later in life, by following up the same subjects. Early preferences were estimated through recordings of food choices conducted in a nursery canteen in children aged 2–3, from 1982 to 1999. The children were free to choose the composition of their lunch from among a varied offering of eight dishes. The same subjects ( n =341) were contacted in 2001–2002 and so their ages varied from 17–22 ( n =91), 13–16 ( n =68), 8–12 ( n =99) to 4–7 ( n =83). Their present preference for the 80 foods most frequently presented at the nursery canteen was assessed through a questionnaire. Five food categories we…
