Synthesis of mesoporous silica nanoparticles (MSNs) and encapsulation of cisplatin for targeted cancer therapies

The aim of this PhD thesis is to elaborate mesoporous silica nanoparticles (MSNs) able to sustain the release of cisplatin into the intracellular compartments of solid tumors. The parametric study shows that morphological, structural and textural properties of MSNs-MCM-41, synthesized by sol-gel reaction by using TEOS as a silica source and CTAB as a structure-directing agent, depend on pH, stirring speed, temperature and extraction process of CTAB. Testing Soil Encasing Materials for Measuring Hydraulic Conductivity of a Sandy-Loam Soil by the Cube Methods

The Cube Method (CM) and the Modified Cube Method (MCM) were developed for measuring vertical (K v ) and horizontal (K h ) saturated hydraulic conductivity of a single soil sample. By these methods, a cube of soil is carved out in situ and a suitable material is applied to enclose the cube in a tightly fitting casing before moving the sample to the laboratory. Problems may be associated with the use of gypsum, originally used to encase soil. The suitability of molten wax and expandable polyurethane foam to encase a soil cube was tested for a sandy-loam soil. This thesis was carried out at the Laboratory of Pharmacognosy and the Laboratory of Pharmaceutical technology, at the UFR Sciences de Santé, circonscription Pharmacy, in the University of Burgundy Franche-Comté, to find new natural molecules to encapsulate. First of all, we focused on the natural saponins from plants of the Vietnam biodiversity and the french horticulture, belonging to the three genera Dracaena, Cordyline (Asparagaceae) and Weigela (Caprifoliaceae). The work led to the successful isolation and elucidation of 42 natural saponins using various chromatographic techniques. The structures were determined by ESI mass spectrometry and NMR spectroscopy. Encapsulation of Xenon by a Self-Assembled Fe4L6 Metallosupramolecular Cage

We report (129)Xe NMR experiments showing that a Fe4L6 metallosupramolecular cage can encapsulate xenon in water with a binding constant of 16 M(-1). The observations pave the way for exploiting metallosupramolecular cages as economical means to extract rare gases as well as (129)Xe NMR-based bio-, pH, and temperature sensors. Xe in the Fe4L6 cage has an unusual chemical shift downfield from free Xe in water. The exchange rate between the encapsulated and free Xe was determined to be about 10 Hz, potentially allowing signal amplification via chemical exchange saturation transfer. At higher RH, additional water uptake can only add-up to an adsorbed water layer. The present work provides evidence that a small amount of the adsorbent dissolves in the adsorbed layer and gypsum starts precipitating at 97-98 %RH even though condensation of the vapour phase does not take place. Irreversible modifications of the porous microstructure of soluble anhydrite β-CaSO4 induced by hydration-dehydration cycles

26 pages; The internal surface area of hexagonal (soluble) primary anhydrite CaSO4 produced by dehydration of gypsum decreases by about 70 % when the anhydrite is successively subjected to rehydration and dehydration at room temperature in humid air and in vacuum respectively. In the rehydration step, the hemihydrate is formed; its dehydration yields secondary anhydrite. Additional hydration-dehydration cycles in the same conditions have a much smaller effect. The first cycle also brings about a modification of the t-plot, which reveals that the micropores of primary anhydrite are irreversibly healed.
