Use of reversed-phase C18 Sep-Pak cartridges for the purification and concentration of sepiapterin and other pteridines

Abstract Several pteridines have been tested for their ability to bind to C 18 Sep-Pak. Riboflavin, sepiapterin, deoxysepiapterin, 6-acetyl-7,8-dihydropterin, 6-acetyldihydrohomopterin and 3′-hydroxysepiapterin were strongly retained and all but 6-acetyldihydrohomopterin quantitatively recovered upon elution with 2 ml of methanol. The effect of the concentration and volume of the sample, pH and salt concentration on the retention of sepiapterin have been studied. The procedure was very useful for the purification, desalting, solvent exchange and concentration of pteridines having high affinity for the cartridge. Protein and amino acid composition of the tergal gland secretions ofBlatta orientalis andEurycotis floridana (Dictyoptera: Blattidae)

Summary Nymphs and adult females ofBlatta orientalis and nymphs ofEurycotis floridana produce a proteinaceous sticky secretion which accumulates on the last abdominal tergites. The proteic patterns do not differ between individuals of the same species. HPLC analyses show that all the common amino acids are found in both species, aspartic and glutamic acids representing 24 to 37% of the total amount of amino acids. InB. orientalis, glutamic acid is the more abundant amino acid whereas inE. floridana it is the aspartic acid. The secretion appears and accumulates rapidly on isolated insects. Behavioural assays revealed that these secretions have a defensive role.

Target Analogue Imprinted Polymers with Affinity for Folic Acid and Related Compounds

Two approaches to synthesize molecularly imprinted polymers with affinity for folic acid and other substituted pteridines have been compared. In the first approach, the folic acid analogue methotrexate was used as template and functional monomers capable of generating selective binding sites were searched in a miniaturized screening system based on binding assessment in the batch mode. Highest selectivity was seen using 2-vinylpyridine as functional monomer, which was confirmed in the chromatographic mode for a batch synthesized on a gram scale. Regulatory Changes in Pterin and Carotenoid Genes Underlie Balanced Color Polymorphisms in the Wall Lizard

ABSTRACTReptiles use pterin and carotenoid pigments to produce yellow, orange, and red colors. These conspicuous colors serve a diversity of signaling functions, but their molecular basis remains unresolved. Here, we show that the genomes of sympatric color morphs of the European common wall lizard, which differ in orange and yellow pigmentation and in their ecology and behavior, are virtually undifferentiated. Implication of eNOS Uncoupling in Cardiovascular Disease

Under physiological conditions, nitric oxide (NO) is produced in the vasculature mainly by the endothelial nitric oxide synthase (eNOS). Endothelial NO relaxes blood vessels, inhibits platelet activity, and protects against atherosclerosis. Under pathological conditions such as hypertension, diabetes, and hypercholesterolemia, eNOS may become uncoupled. Uncoupled eNOS generates superoxide at the expense of NO and contributes substantially to oxidative stress and endothelial dysfunction. Major mechanisms of eNOS uncoupling include deficiency of the eNOS cofactor tetrahydrobiopterin, deficiency of the eNOS substrate L-arginine, and eNOS S-glutathionylation. δ18O values of coexisting brachiopods and fish: Temperature differences and estimates of paleo–water depths

To estimate vertical thermal gradients and paleo–water depths to marine platforms we present a new method based on the difference between δ 18 O values of contemporaneous brachiopod carbonate and fish phosphate. Present-day marine fauna of well-known ecology from the surface to the sea floor record isotopic temperatures that agree with measured temperatures. We predict distributions of isotopic data that result from sampling strategy, basin morphology, and fauna ecology and discuss limitations. Application of the method to the Jurassic Paris-London basin gives vertical thermal variations of up to 14 °C associated with depths varying from a few meters to 170 ± 30 m. Influence of bacteria on the metabolite pattern of phenol in zebra fish (Brachydanio rerio)

Summary An investigation was performed on the potential influence of bacteria in the medium on the metabolism of phenol in zebra fish. The results obtained in vitro and in vivo show that bacteria may influence biotransformation studies of xenobiotics by fish in different manner. First, it was shown that phenol conjugates were deconjugated. Hydrolysis of phenyl glucuronide was observed both in vitro and in vivo. Deconjugation of phenyl sulphate was found to a minor extent merely under in vitro conditions. Second, the xenobiotic itself, in this case phenol, was degradated to CO2. Climate change may influence the phenology of organisms unequally across trophic levels and thus lead to phenological mismatches between predators and prey. In cases where prey availability peaks before reproducing predators reach maximal prey demand, any negative fitness consequences would selectively favor resynchronization by earlier starts of the reproductive activities of the predators.
