Ancistrocyclinones A and B, unprecedented pentacyclic N,C-coupled naphthylisoquinoline alkaloids, from the Chinese liana Ancistrocladus tectorius
2018
Two unique pentacyclic N,C-coupled naphthylisoquinolines, the ancistrocyclinones A (5) and B (6), were discovered in the Chinese liana Ancistrocladus tectorius. Furthermore, six known, likewise N,C-coupled alkaloids, viz., ancistrocladinium A (7a) and its mono- and bisphenolic analogs 8a and 9a were isolated, along with their atropo-diastereomers 7b, 8b, and 9b. The stereostructures of 5 and 6 were determined by HRESIMS, 1D and 2D NMR, oxidative degradation, and ECD calculations. The pentacyclic ancistrocyclinones A (5) and B (6) are structurally similar to berberine alkaloids - yet arising from a most different biosynthetic pathway: they are apparently formed by N,C-coupling of their polyk…
Ubiquitin-independent function of optineurin in autophagic clearance of protein aggregates.
2013
Summary Aggregation of misfolded proteins and the associated loss of neurons are considered a hallmark of numerous neurodegenerative diseases. Optineurin is present in protein inclusions observed in various neurodegenerative diseases including amyotrophic lateral sclerosis (ALS), Huntington's disease, Alzheimer's disease, Parkinson's disease, Creutzfeld-Jacob disease and Pick's disease. Optineurin deletion mutations have also been described in ALS patients. However, the role of optineurin in mechanisms of protein aggregation remains unclear. In this report, we demonstrate that optineurin recognizes various protein aggregates via its C-terminal coiled-coil domain in a ubiquitin-independent m…
New folate-functionalized biocompatible block copolymer micelles as potential anti-cancer drug delivery systems
2006
Abstract The main objective of this study was to synthesize novel folic acid-functionalized diblock copolymer micelles and evaluate their solubilization of two poorly water-soluble anti-tumor drugs, tamoxifen and paclitaxel, which suffer from low water solubility and/or poor hydrolytic stability. The diblock copolymer consisted of a permanently hydrophilic block comprising 2-(methacryloyloxy)ethyl phosphorylcholine (MPC) residues and a pH-sensitive hydrophobic block comprising 2-(diisopropylamino)ethyl methacrylate (DPA) residues. Folic acid (FA) was conjugated to the end of the MPC block so that this group was located on the micelle periphery. Tamoxifen- and paclitaxel-loaded micelles were…
4-Nitrophenyl phosphoric acid and its four different potassium salts: a solid state structure and kinetic study
2006
The structures of 4-nitrophenyl phosphoric acid, H2NPP (1), and its four different potassium salts, K(H2NPP)(HNPP) (2), K(HNPP)·CH3OH (3), K(HNPP)·2H2O (4), and K2(NPP)·4H2O (5), were determined by X-ray diffraction methods. These investigations, together with the study of the role of potassium ions in the hydrolysis of 4-nitrophenyl phosphate, provided information about interactions between the acid, monoanionic and dianionic forms of 4-nitrophenyl phosphate and the potassium cations. In the crystalline state, the H2NPP molecules form one-dimensional chains of bifurcated O–H⋯O hydrogen bonds between phosphate groups. A different network, derived from ladder-like chains of the O–H⋯O hydroge…
Fumarate respiration of Wolinella succinogenes: enzymology, energetics and coupling mechanism.
2002
Wolinella succinogenes performs oxidative phosphorylation with fumarate instead of O2 as terminal electron acceptor and H2 or formate as electron donors. Fumarate reduction by these donors ('fumarate respiration') is catalyzed by an electron transport chain in the bacterial membrane, and is coupled to the generation of an electrochemical proton potential (Deltap) across the bacterial membrane. The experimental evidence concerning the electron transport and its coupling to Deltap generation is reviewed in this article. The electron transport chain consists of fumarate reductase, menaquinone (MK) and either hydrogenase or formate dehydrogenase. Measurements indicate that the Deltap is generat…
Long-term dynamics of water-borne nitrogen, phosphorus and suspended solids in the lower Don River basin (Russian Federation)
2011
A long-term study (1986–2002) of water-borne nutrient and suspended solids dynamics was undertaken on the lower Don River, which plays an extremely important role in the water supply of the Black Sea and Azov Sea basin. Suspended solids were greatest in spring and summer and were correlated to river discharge. Mean annual nitrogen concentrations increased from 1986 to 1995 and then decreased from 1996 to 2002. Unlike nitrogen, phosphorus concentrations (both phosphates and total phosphorus) gradually increased throughout the study period changing the river from an oligotrophic to upper mesotrophic status. If this trend continues phytoplankton could become nitrogen-limited leading to the dev…
Linking nutrient limitation and water chemistry in upland lakes to catchment characteristics
2003
The relationship between catchment characteristics and lake water chemistry, phytoplankton and periphyton biomass, and phytoplankton and periphyton nutrient limitation was investigated for 30 upland lakes in the U.K. These catchment characteristics included the proportion of different land cover categories in the catchment and some hydrological information. Multiple regression models could predict alkalinity, pH, total dissolved phosphorus, dissolved inorganic nitrogen, dissolved organic nitrogen, dissolved organic carbon and phytoplankton chlorophyll a from the proportional contribution of between two and six land cover categories within the catchment and explain between 42 and 73% of the …
Nutrient dependence of phytoplankton production in brown-water lakes with special reference to Lake Päijänne
1982
A method for predicting the mean seasonal chlorophyll a concentration, the mean seasonal in vitro phytoplankton primary productivity per unit volume, the maximum daily production per unit volume and the seasonal integral production in brown-water lakes is presented. The production values can be calculated when the mean annual concentration of total phosphorus and the mean annual colour of the water are known. This method has been developed especially for practical water pollution studies to permit rapid and inexpensive estimates of major biological consequences of changes in effluent loads. The method can be applied for brown-water lakes where phosphorus is the limiting nutrient for primary…
Mapping Sediment Accumulation Rate by using Volume magnetic Susceptibility Core Correlation in a contaminated Bay (Lake Geneva, Switzerland)
2003
The Bay of Vidy is the most contaminated area of Lake Geneva: contamination is caused by the effluent of the sewage treatment plant (STP) of the City of Lausanne. The implementation of a chemical stage in the treatment plant to remove phosphorus using FeC13 in 1971 is indirectly recorded in the lake sediments by a strong and sharp increase in the volume magnetic susceptibility (VMS) signal. A total of 43 sediment cores have been retrieved and measured for VMS. The synchronism of the VMS signal increase and its relation to the implementation of the P-removal stage in the STP has been shown in seven 137Cs-dated sediment cores. The VMS signal has been used to date by stratigraphie correlation …
Trends in nutrient concentrations in Latvian rivers and the response to the dramatic change in agriculture
2003
In recent years, the use of fertilisers in the Baltic countries (Estonia, Latvia, and Lithuania) has decreased at an unprecedented rate. The import of mineral fertilisers and feed stuff became almost non-existent, and extensive slaughtering of livestock reduced the amount of manure. In Latvia, the purchase of mineral fertilisers decreased by a factor of 15 between 1987 and 1996 and the number of livestock decreased with a factor of almost 4 during the same time period. Such abrupt and comprehensive changes in land use have never before occurred in the history of modern European agriculture. Here, the impact that this dramatic reduction has had on concentrations of nutrients in Latvian river…
