Deficient p27 Phosphorylation at Serine 10 Increases Macrophage Foam Cell Formation and Aggravates Atherosclerosis Through a Proliferation-Independen…

2011

OBJECTIVE: Genetic ablation of the growth suppressor p27(Kip1) (p27) in the mouse aggravates atherosclerosis coinciding with enhanced arterial cell proliferation. However, it is unknown whether molecular mechanisms that limit p27's protective function contribute to atherosclerosis development and whether p27 exerts proliferation-independent activities in the arterial wall. This study aims to provide insight into both questions by investigating the role in atherosclerosis of p27 phosphorylation at serine 10 (p27-phospho-Ser10), a major posttranslational modification of this protein. METHODS AND RESULTS: Immunoblotting studies revealed a marked reduction in p27-phospho-Ser10 in atheroscleroti…

Thermo-oxidative stabilization of poly(lactic acid) with antioxidant intercalated layered double hydroxides

2016

Two antioxidant modified layered double hydroxides (AO-LDHs) were successfully prepared by the intercalation of 3-(3,5-di-tert-butyl-4-hydroxyphenyl)propionic acid (IrganoxCOOH) and 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox) in the layered structure of LDH. It was found that by anchoring the phenolic moieties to the LDH layers the antioxidant power is retained in the case of Trolox, and even amplified in the case of IrganoxCOOH. A small amount of the two AO-LDHs was incorporated into poly(lactic acid), PLA, by solution mixing and melt extrusion. The thermo-oxidative stability of the composites was compared with that of the neat PLA and PLA containing free AOs. SEC analy…

Oxidative dehydrogenation of ethane on Te-containing MoVNbO catalysts

2004

Abstract Ethylene has selectively been obtained during the oxidative dehydrogenation of ethane on a Mo–V–Te–Nb–O mixed oxide catalyst. The catalyst was prepared hydrothermally and heat-treated at 600 °C for 2 h in a N2-stream. For comparison, undoped and Te-doped Mo–V–Nb–O catalysts and Mo–V–Te–O and Mo–Te–Nb–O mixed oxides have also been prepared. The catalytic performance of the Mo–V–Nb–Te–O mixed oxide catalyst cannot be explained by the incorporation of Te on the surface of a Mo–V–Nb–O mixed oxide but by the presence of an orthorhombic Te2M20O57 (M=Mo, V, Nb) crystalline phase. The role of tellurium in these catalysts is also discussed.

The Effect of Calcium Carbonate on the Photo-Oxidative Behavior of Poly(butylene adipate-co-terephthalate)

2020

The aim of this work is to evaluate the influence of nanosized CaCO3 on the photo-oxidation of poly(butylene adipate-co-terephthalate) (PBAT)-based nanocomposites. The PBAT/CaCO3 nanocomposites are prepared by using a corotating twin-screw extruder with 0, 2, and 5 wt% of CaCO3. The films are obtained by film blowing process. Specimens of the nanocomposites and that of the pure polymer are subjected to accelerated aging with a cycle of 8 h of light at a temperature of 55 °C followed by 4 h condensation at 45 °C to evaluate the effects of the CaCO3 addition on the photo-oxidation of PBAT. The results indicate that the modulus and tensile strength of the nanocomposites are increased significa…

Manganese interferes with calcium, perturbs ERK signaling, and produces embryos with no skeleton.

2011

Manganese (Mn) has been associated with embryo toxicity as it impairs differentiation of neural and skeletogenic cells in vertebrates. Nevertheless, information on the mechanisms operating at the cellular level remains scant. We took advantage of an amenable embryonic model to investigate the effects of Mn in biomineral formation. Sea urchin (Paracentrotus lividus) embryos were exposed to Mn from fertilization, harvested at different developmental stages, and analyzed for their content in calcium (Ca), expression of skeletogenic genes, localization of germ layer markers, and activation of the extracellular signal-regulated kinase (ERK). By optical and immunofluorescence microscopy, we found…

Pancreatic T cell protein-tyrosine phosphatase deficiency ameliorates cerulein-induced acute pancreatitis.

2014

Background Acute pancreatitis (AP) is a common clinical problem whose incidence has been progressively increasing in recent years. Onset of the disease is trigged by intra-acinar cell activation of digestive enzyme zymogens that induce autodigestion, release of pro-inflammatory cytokines and acinar cell injury. T-cell protein tyrosine phosphatase (TCPTP) is implicated in inflammatory signaling but its significance in AP remains unclear. Results In this study we assessed the role of pancreatic TCPTP in cerulein-induced AP. TCPTP expression was increased at the protein and messenger RNA levels in the early phase of AP in mice and rats. To directly determine whether TCPTP may have a causal rol…

The Low Energy-Coupling Respiration in Zymomonas mobilis Accelerates Flux in the Entner-Doudoroff Pathway.

2015

Performing oxidative phosphorylation is the primary role of respiratory chain both in bacteria and eukaryotes. Yet, the branched respiratory chains of prokaryotes contain alternative, low energy-coupling electron pathways, which serve for functions other than oxidative ATP generation (like those of respiratory protection, adaptation to low-oxygen media, redox balancing, etc.), some of which are still poorly understood. We here demonstrate that withdrawal of reducing equivalents by the energetically uncoupled respiratory chain of the bacterium Zymomonas mobilis accelerates its fermentative catabolism, increasing the glucose consumption rate. This is in contrast to what has been observed in o…

Flow cytometric analysis of peroxidative activity in granulocytes from coronary and peripheral blood in acute myocardial ischemia and reperfusion in …

1999

BACKGROUND Methionine has shown protective effects in experimental models of myocardial infarction and is highly reactive to oxidative compounds produced by polymorphonuclear leukocytes (PMN), which in turn have been associated with myocardial damage. We have investigated the effect of methionine administration on spontaneous leukocyte peroxidative activity in myocardial ischemia and reperfusion. METHODS In anesthetized dogs, with coronary occlusion (90 min) and reperfusion (90 min), PMN activation was measured by flow cytometric determination of H(2)O(2) with dihydrorhodamine 123, and correlated to hemodynamic parameters and infarct presence. To assess a possible direct effect of methionin…

The thiol switch C684 in Mitofusin-2 mediates redox-induced alterations of mitochondrial shape and respiration

2017

Mitofusin-2 (MFN2) is a GTPase in the outer mitochondrial membrane involved in the regulation of mitochondrial fusion and bioenergetics. MFN2 also plays a role in mitochondrial fusion induced by changes in the intracellular redox state. Adding oxidized glutathione (GSSG), the core cellular stress indicator, to mitochondrial preparations stimulates mitochondrial fusion by inducing disulphide bond-mediated oligomer formation of MFN2 and its homolog MFN1 which involve cysteine 684 (C684) of MFN2. Mitochondrial hyperfusion represents an adaptive stress response that confers transient protection by increasing mitochondrial ATP production but how this depends on the thiol switch C684 in MFN2 has …
