Biopartitioning micellar chromatography: an in vitro technique for predicting human drug absorption.

The main oral drug absorption barriers are fluid cell membranes and generally drugs are absorbed by a passive diffusion mechanism. Biopartitioning micellar chromatography (BMC) is a mode of micellar liquid chromatography that uses micellar mobile phases of Brij35 under adequate experimental conditions and can be useful to mimic the drug partitioning process in biological systems. In this paper the usefulness of BMC for predicting oral drug absorption in humans is demonstrated. A hyperbolic model has been obtained using the retention data of a heterogeneous set of 74 compounds, which shows predictive ability for drugs absorbed by passive diffusion. Abstract An automated HPLC method with column switching is described for the determination of quetiapine, clozapine, perazine, olanzapine and metabolites in blood serum. After clean-up on silica C8 material (20 μm particle size) drugs were separated on ODS Hypersil C18 material (5 μm; column size 250 mm × 4.6 mm i.d.) within 25 min and quantified by ultraviolet (UV) detection at 254 nm. The limit of quantification ranged between 10 and 50 ng/ml. At therapeutic concentrations of the drugs, the inter-assay reproducibility was below 10%. The drug permeability prediction across the ocular tissues is important in the development of new drugs and drug delivery strategies. Physicochemical characteristics of drugs, mainly acid-base character, hydrophobicity and the molecular size determine both their transport across the eye tissue barriers and their retention in biopartitioning micellar chromatography (BMC). Aripiprazole is a novel atypical antipsychotic drug for the treatment of schizophrenia and schizoaffective disorders (1)(2)(3). The drug is metabolized by the cytochrome P450 isoenzymes 3A4 and 2D6 (4). Because of high interindividual variability in the expression of these enzymes, the aripiprazole concentration varies among healthy individuals after administration of the drug (5). In patients, insufficient response or side effects, such as somnolence, akathisia, or nausea, may result from too low or too high drug concentrations. In the Common woodlouse Porcellio scaber (Crustacea: Isopoda: Oniscidea), experimental immune challenge did not induce the expression of pro-phenoloxidase that, in most other invertebrates studied thus far, can be activated into phenoloxidase via an activation cascade upon immune challenge. Instead, Porcellio hemocyanin proved to exhibit catecholoxidase activity upon activation. Comparison of various liquid chromatographic methods for the analysis of avermectin residues in citrus fruits.

Abstract Various liquid chromatographic (LC) techniques for analyzing avermectin (Abamectin) were compared after extraction of residues from citrus fruit samples by matrix solid-phase dispersion (MSPD). LC with UV and fluorescence detection were used as also was LC coupled to the mass spectrometer by an electrospray interface. The results obtained by the three methods were compared in terms of sensitivity and selectivity. The combination of MSPD extraction and LC with fluorescence detection have made it possible to quantify 0.5 μg kg −1 of Abamectin in 0.5 g of orange sample, with an overall average recovery of 94%. In Vitro Dialyzability of Zinc from Different Salts Used in the Supplementation of Infant Formulas

Seven zinc salts--acetate, chloride, lactate, sulfate, citrate, gluconate, and oxide--were added to milk--and soy-based infant formulas to estimate possible differences in zinc availability depending on the type of salt used. For this purpose, an in vitro method that estimates the dialyzability of the element (i.e., the fraction available for absorption) was applied. Zinc dialyzability is always higher in milk-based products than in soy products, even when the total zinc contents are higher in the latter. Cadmium binding proteins induced in exposed freshwater crayfish Procambarus clarkii.

This work describes results on the characterization of cadmium binding proteins (Cd-BPs) obtained from cadmium exposed freshwater crayfish Procambarus clarkii. After acclimation to laboratory conditions, induction of Cd-BPs was achieved by water exposure at a concentration of 100 micrograms Cd/L during 2, 15, and 30 d. In accordance with the method followed by Engel and Brouwer, in each case two midgut glands were minced and homogenized in Tris-HC1 buffer with PMSF to prevent protease activity and DTT to maintain reducing conditions. Multicolour LEDs in educational demonstrations of physics and optometry

LED light sources are used to design experimental setup for university courses teaching human color vision. The setup allows to demonstrate various vision characteristics and to apply for student practical exercises to study eye spectral sensitivity in different spectral range using heterochromatic flicker photometry. Technique can be used in laboratory works for students to acquire knowledge in visual perception, basics of electronics and measuring, or it can be applied as fully computer control experiment.
