Étude des mécanismes moléculaires associés aux effets de l'ODN sur des cellules astrogliales et microgliales soumises à un stress oxydant : impact su…

2019

Neurodegenerative diseases are characterized by oxidative stress associated with mitochondrial damages leading to neuronal cell death. To mitigate these damages and promote neuronal cytoprotection and neurogenesis, endogenous (Neuropeptide: octadecaneuropeptide (ODN)) or exogenous (Polyphenols: resveratrol (RSV) and apigenin (API)) natural neurotrophic factors could be used as therapeutic agents to promote neuronal differentiation of immature and pluripotent stem cells. ODN is a peptide produced by astrocytes and known as a powerful neuroprotective agent. It is therefore of interest of studying its effects on the mobilization of calcium, its ability to protect neuronal cells against apoptos…

Genetic and biochemical characterization by random and site-directed mutagenesis, repressors involved in stress response phenolic acids in Bacillus s…

2009

The aim of this work was to characterize the genetic and biochemical mechanisms involved in the PASR, the phenolic acid stress response induced by phenolic acids Bacillus subtilis. By genetics and molecular biology strategies, like reverse genetic, PCR or transposon random mutagenesis, gene deletion, complementation of strains, and gene expression, the main components of the PASR were identified and quantified. Site-directed and random mutagenesis of respectively the promoter of padC encoding the phenolic acid decaroxylase, and PadR, the repressor of the padC expression, allowed us to identify the site of binding of PadR with this promoter, as well as the amino-acid residues involved in the…

Effets des polyphénols de vin rouge sur la prolifération cellulaire et sur le métabolisme du resvératrol

2011

It is known that moderate consumption of red wine may protect against many diseases, in particular because of its polyphenols. We studied in particular the effect of these polyphenols on colorectal cancer, one of the most common cancers in industrialized countries. Some purified polyphenols from red wine, like resveratrol and quercetin, showed antiproliferative effects against colorectal cancer cells. Our first approach was to use polyphenolic extracts from Burgundy red wine. We then studied their in vitro effects on human colorectal tumor cell lines SW480’s proliferation and the interactions between the polyphenols and the transport and metabolism of resveratrol; and secondly, the effects o…
