A block copolymer (6) with both hydrophilic and hydrophobic regions was synthesized, in order to examine its interaction with model membranes and its uptake by living cells. The copolymer comprised poly(ethylene oxide) and poly(L-lysine) with 50 mol-% substitution of the e-amino groups with palmitoyl groups. To permit 125I-labelling, p-methoxyphenyl residues (1–4 mol-%) were incorporated into the block copolymer and into a poly(ethylene oxide) used for comparison. Membrane fission is an essential process in membrane trafficking and other cellular functions. While many fissioning and trafficking steps are mediated by the large GTPase dynamin, some fission events are dynamin independent and involve C-terminal-binding protein-1/brefeldinA-ADP ribosylated substrate (CtBP1/BARS). Even though it is known for more than one decade that antigen-encoding RNA can deliver antigenic information to induce antigen-specific immunity against cancer, the nature and mechanism of RNA uptake have remained enigmatic. In this study, we investigated the pharmacokinetics of naked RNA administered into the lymph node. We observed that RNA is rapidly and selectively uptaken by lymph node dendritic cells (DCs). Furthermore, in vitro and in vivo studies revealed that the efficient internalization of RNA by human and murine DCs is primarily driven by macropinocytosis.
