In vitro morphogenesis from excised leaf explants of Digitalis obscura L.

The morphogenic capacity of Digitalis obscura leaf explants cultured in vitro has been studied, noting factors promoting the differentiation of roots, buds and shoots as well as those promoting callus proliferation. Complete plant regeneration was obtained only by first culturing the leaf explants in a medium with NAA and BA to induce formation of buds, and subsequently transferring them to a medium without growth regulators to achieve the further development of shoots.

Mosses of the subfamily Orthotrichoideae represent one of the main components of the cryptogam epiphytic communities in temperate areas. During the last two decades, this taxonomical group has undergone an extensive revision that has led to its rearrangement at the generic level. However, their phylogenetic relationships and inferences on the evolutionary patterns that have driven the present diversity have little advanced. Knowledge of the processes by which plants colonize old structures is a key element for nature-based design both in urban and suburban contexts. This paper analyses the natural vegetation on walls and in other microhabitats of the roadway structures of Monte Pellegrino (606 m a.s.l.) near Palermo (Sicily), built in the first half of the 1900s. The historical road has particular construction and architectural features, and its characteristics have been maintained to this day. Genetic instability in calamondin (Citrus madurensis Lour.) plants derived from somatic embryogenesis induced by diphenylurea derivative

Somatic embryos were regenerated in vitro from calamondin style-stigma explants cultured in the presence of N (6)-benzylaminopurine (BAP) cytokinin and three synthetic phenylurea derivatives, N-(2-chloro-4-pyridyl)-N-phenylurea (4-CPPU), N-phenyl-N'-benzothiazol-6-ylurea (PBU) and N,N'-bis-(2,3-methilendioxyphenyl)urea (2,3-MDPU). The phenylurea derivative compounds tested at micromolar level (12 muM) were able to induce a percentage of responsive explants significantly higher from that obtained with BAP and hormone-free (HF) conditions. Somatic embryogenesis and plant regeneration from pistil thin cell layers of Citrus

Callus induction, somatic embryogenesis and plant regeneration were obtained in six different citrus species [Citrus deliciosa Ten. (cv 'Avana'), C.limon (L.) Burm. (cv 'Berna'), C.madurensis Lour. Investigating the cryopreservation of nodal explants of Lithodora rosmarinifolia (Ten.) Johnst., a rare, endemic Mediterranean species

In this study, we investigated the possibility of using the droplet-vitrification technique for cryopreserving nodal segments of in vitro plantlets of the endangered plant species Lithodora rosmarinifolia. Among the three vitrification solutions tested, only solutions B1, containing (w/v) 50 % glycerol and 50 % sucrose, and B3, containing 40 % glycerol and 40 % sucrose, were able to induce cryotolerance in nodal explants, resulting in intermediate survival and recovery after cryopreservation. Relevant habitats neglected by the Directive 92/43 EEC: the contribution of Vegetation Science for their reappraisal in Sicily

Field investigation carried out by the Sicilian botanists in the last 20 years enabled them to identify eight habitat types of high biogeographic and conservation interest, neglected by the Directive 92/43, which deservead hocconservation measures. Summary The main objective of the present study was to test the hypothesis that increased concentrations of ethylene under conditions of limited gas exchange during in vitro cultivation of potato ( Solanum tuberosum L.) stem explants may lead to increased capaciry of antioxidative defense. By varying the tightness of closure and application of inhibitors of ethylene biosynthesis it was possible to accumulate various concentrations of ethylene in the internal atmosphere of culture flasks, which depended on excision-stress induced ethylene biosynthesis.
