Structural Determinants for the Mode of Action of Imidazopyridine DS2 at δ-Containing γ-Aminobutyric Acid Type A Receptors

Despite the therapeutic relevance of δ-containing γ-aminobutyric acid type A receptors (GABAARs) and the need for δ-selective compounds, the structural determinants for the mode and molecular site of action of δ-selective positive allosteric modulator imidazo[1,2-a]pyridine DS2 remain elusive. To guide the quest for insight, we synthesized a series of DS2 analogues guided by a structural receptor model. Using a fluorescence-based fluorometric imaging plate reader membrane potential assay, we found that the δ-selectivity and the pharmacological profile are severely affected by substituents in the 5-position of the imidazopyridine core scaffold. Interferon-regulatory factor 4 is essential for the developmental program of T helper 9 cells.

Summary Interferon-regulatory factor 4 (IRF4) is essential for the development of T helper 2 (Th2) and Th17 cells. Herein, we report that IRF4 is also crucial for the development and function of an interleukin-9 (IL-9)-producing CD4 + T cell subset designated Th9. IRF4-deficient CD4 + T cells failed to develop into IL-9-producing Th9 cells, and IRF4-specific siRNA inhibited IL-9 production in wild-type CD4 + T cells. Chromatin-immunoprecipitation (ChIP) analyses revealed direct IRF4 binding to the Il9 promoter in Th9 cells. Lead Nanowires for Microaccumulators Obtained Through Indirect Electrochemical Template Deposition

Metallic lead nanowires were deposited within pores of commercial anodic alumina membranes having an average pore diameter of 210 nm. "Direct" electrodeposition was attempted from 0.1 M Pb(NO 3 ) 2 aqueous solution with a variable concentration of H 3 BO 3 as a chelating agent, but it gave unsatisfactory results. An "indirect" two-step deposition procedure was then adopted, consisting of the anodic electrodeposition of α-PbO 2 nanowires, followed by their in situ reduction to metallic lead. Both these processes occurred at a high rate so that the indirect method led to a complete template pore filling with pure polycrystalline Pb in short times and with a high current efficiency.

Ultrastructure of the Interlamellar Membranes of the Nacre of the Bivalve Pteria hirundo, Determined by Immunolabelling.

The current model for the ultrastructure of the interlamellar membranes of molluscan nacre imply that they consist of a core of aligned chitin fibers surrounded on both sides by acidic proteins. This model was based on observations taken on previously demineralized shells, where the original structure had disappeared. Despite other earlier claims, no direct observations exist in which the different components can be unequivocally discriminated. We have applied different labeling protocols on non-demineralized nacreous shells of the bivalve Pteria. Synergistic Platelet Inhibitory Effect of the Phosphodiesterase Inhibitor Piroximone and Iloprost

Platelet activity is regulated through synthesis and degradation of the intracellular second messengers cAMP or cGMP. The antiplatelet effect of the phosphodiesterase (PDE) III inhibitor Piroximone (PIR) was studied in vitro in platelet rich plasma. ADP induced aggregation was inhibited by PIR with an IC50 of 67 +/- 43 microM. The inhibitory effect was time and dose dependent. The antiaggregatory effects in vivo were studied in anaesthetised rats. Reduction of platelet count following injection of 100 micrograms/kg bw collagen was measured after bolus injection of PIR and vehicle. Soil morphological, physical and chemical properties are described at four locations along an elevational transect in the northeastern part of the Moldavian Plateau (Romania). These data contribute to the knowledge of the soils of this area and to their classification according to the USDA-Soil Taxonomy, FAO-WRB and the SRTS-Romanian System. Organophosphorus moieties have been incorporated into mesoporous ALPOs through a one-pot surfactant-assisted procedure leading, for the first time, to periodic mesoporous aluminium phosphonates and diphosphonates.
