Thermophilic anaerobic conversion of raw microalgae: Microbial community diversity in high solids retention systems

[EN] The potential of microbial communities for efficient anaerobic conversion of raw microalgae was evaluated in this work. A long-term operated thermophilic digester was fed with three different Organic Loading Rates (OLR) (0.2, 0.3 and 0.4¿g·L¿1·d¿1) reaching 32¿41% biodegradability values. The microbial community analysis revealed a remarkable presence of microorganisms that exhibit high hydrolytic capabilities such as Thermotogae (~44.5%), Firmicutes (~17.6%) and Dictyoglomi, Aminicenantes, Atribacteria and Planctomycetes (below ~5.5%) phyla. The generality of increasing diversity of fungi and bacteria across arctic sand dune succession was tested. Microbial communities were examined by high-throughput sequencing of 16S rRNA genes (bacteria) and internal transcribed spacer (ITS) regions (fungi). We studied four microbial compartments (inside leaf, inside root, rhizosphere and bulk soil) and characterized microbes associated with a single plant species (Deschampsia flexuosa) across two sand dune successional stages (early and late). Bacterial richness increased across succession in bulk soil and leaf endosphere. Significance Ephemeral diets such as carrion are high-quality resources that are susceptible to microbial spoilage. Carrion-feeding insects that breed on decaying carcasses must overcome challenges arising from competing microbes. Here we report that a carrion-feeding burying beetle preserves carcasses by regulating its microbial growth, resulting in changes in its biochemical properties including the reduction of toxic polyamines associated with putrefaction and nutrient loss. The beetle’s microbial symbionts form a biofilm-like matrix on carcasses, which is important for optimal larval development. Necrophagous beetles utilize carrion, a highly nutritious resource that is susceptible to intense microbial competition, by treating it with antimicrobial anal and oral secretions. However, how this regulates the carcass microbiota remains unclear. Here, we show that carcasses prepared by the burying beetle Nicrophorus vespilloides undergo significant changes in their microbial communities subsequent to their burial and ‘preparation’. Prepared carcasses hosted a microbial community that was more similar to that of beetles’ anal and oral secretions than to the native carcass community or the surrounding soil, indicating that the beetles regulated the carcass microbiota. Abstract Harvested microalgae Chlorella spp. and primary sludge were co-digested in a laboratory-scale anaerobic membrane bioreactor (AnMBR) under thermophilic conditions (55 °C). The system was run for 700 days divided into four experimental phases to determine the influence of the organic loading rate on the process performance and the microbial community. The rise in organic loading rate from 0.17 to 0.5 gCOD·L−1·d−1 led to a 35% improvement in methane production.
