Three 2-polyprenyl-1,4-hydroquinone derivatives (2-heptaprenyl-1,4-hydroquinone: IS1, 2-octaprenyl-1,4-hydroquinone: IS2 and 2-[24-hydroxy]-octaprenyl-1,4-hydroquinone: IS3) isolated from the Mediterranean sponge Ircinia spinosula, were evaluated for effects on phospholipase A2 activity of different origin (Naja naja venom, human recombinant synovial fluid and bee venom), as well as on human neutrophil function and mouse ear oedema induced by 12-O-tetradecanoylphorbol 13-acetate (TPA). IS1 interacted minimally with these responses. The ganglioside GD3 has a variety of biological functions. These include stimulatory effects on proliferation, natural killer activity and cytokine production by freshly isolated peripheral T cells. In this study we have characterized anti-GD3 antibody (MoAb Z21) mediated effects on T cell clones. Our data indicate that alpha/beta TCR CD4+ and CD8+ as well as gamma/delta TCR positive T cells can be stimulated resulting in proliferation and cytokine production. This effect could be blocked by cyclosporin A and did not involve the LFA-3 or CD4 molecule. Sponges (Porifera), represent the phylogenetically oldest metazoan phylum still extant today. Recently, molecular biological studies provided compelling evidence that these animals share basic receptor/ligand systems, especially those involved in bodyplan formation and in immune recognition, with the higher metazoan phyla. An in vitro cell/organ-like culture system, the primmorphs, has been established that consists of proliferating and differentiating cells, but no canals of the aquiferous system. We investigated the possibility of transfer immunity from Cryptosporidium parvum-infected interferon-gamma (GKO) and interleukin-12p40 (IL-12KO) deficient C57BL/6 mice to naive mice by transfer of intraepithelial lymphocytes (IELs) and CD4(+) T cells from spleen and mesenteric lymph nodes (MLNs). Three days after the transfer recipients were infected with C. parvum. IELs isolated from GKO donor mice after resolution of infection (day 15) but not at the peak of infection (day 8) significantly reduced the parasite load in recipient mice. The depositional and diagenetic processes involved in the formation of carbonates in the evaporitic environment of the Messinian Salinity Crisis are investigated in Southern Italy (Sicily and Calabria). Strong differences are observed between the studied sections that reflect specific depositional and diagenetic evolution in the interconnected sub-basins resulting from the syn-sedimentary tectonic fragmentation of the Central Sicilian and Calabrian domains. Abstract A multi-scale analysis of sedimentary carbonate facies and post-sedimentary diagenetic features of the Calcare di Base Formation, the precursor to evaporites in Upper Messinian successions of Northern Calabria and Central Sicily, has revealed their microbial bio-mediated origin. Massive to laminated microbial boundstones represent the most common sedimentary facies forming flat to low relief cm to m scale stromatolitic and thrombolitic bodies. The fabric of the micrite varies from peloidal to aphanitic, and almost always preserves filamentous bacteria which characterized the original microbial mat. Abstract Three sections of the early Messinian Tripoli Formation from the northern and southern margins of the central Sicilian Basin (Serra Pirciata, Torrente Vaccarizzo, and Marianopoli) have been studied with the aim to reconstruct the sedimentary and environmental changes which occurred during the transition between marine conditions and the evaporitic events of the Salinity Crisis recorded in the overlying Calcare di Base Formation.
