Spectroscopic analysis used to uncover the original paint colour of the Helsinki Government Palace tower clock faces

The paint stratigraphy of the two clock faces from the tower clock of the Government Palace in Helsinki (Finland) was analysed in order to determine their original colour before restoration works. Paint cross-section samples from both clock faces were analysed by confocal Raman microscopy and scanning electron microscopy coupled to an energy dispersive X-ray spectrometer (SEM-EDS). The results revealed the complex superimposition of paint layers applied over the original black colour. FTIR/ATR analyses proved that the original paint was prepared with linseed oil-resin media. Phylogenetic analysis of Lymnaeid snails based on 18S rDNA sequences.

The 18S rDNA sequences of the six most common European Lymnaeidae species (Mollusca:Gastropoda:Basommatophora) have been obtained by direct PCR cycle sequencing and silver staining methods. The sequence alignment and secondary structures of the 18S rRNA gene of Lymnaea stagnalis, L. auricularia, L. peregra, L. palustris, L. glabra, and L. truncatula are analyzed. This gene proves to be a good marker for both specific determination and supraspecific lymnaeid phylogeny. ABSTRACT We investigated the effects of internal primer-template mismatches on the efficiency of PCR amplification using the 16S rRNA gene as the model template DNA. We observed that the presence of a single mismatch in the second half of the primer extension sequence can result in an underestimation of up to 1,000-fold of the gene copy number, depending on the primer and position of the mismatch.

Xyloglucan xyloglucosyltransferase/endohydrolase (XTHs: EC 2.4.1.207 and/or EC 3.2.1.151) has been proposed to have a dual role integrating newly secreted xyloglucan chains into an existing wall-bound xyloglucan and restructuring existing cell wall material by catalyzing transglucosylation between previously wall bound xyloglucan molecules. Among amphibians, a number of probably related groups are known from both continents, but their phylogenies are so far largely unresolved. In the lactatingmammary gland, weaning produces mitochondrial cytochrome c release and nuclear DNA fragmentation, as determined by gel electrophoresis.
