Antimikrobiālo virsmu efektivitāte atkarībā no mikroorganismu fizioloģiskā stāvokļa

Esošā pētījuma uzdevums ir salīdzināt vara nanodaļiņu antimikrobiālā pārklājuma iedarbības efektivitāti uz divām baktēriju sugām, to trijos fizioloģiskajos stāvokļos, kā arī novērtēt izmantotās metodoloģijas piemērotību mērķa sasniegšanai. Production of a Recombinant Catechol 2,3-Dioxygenase for the Degradation of Micropollutants

Phenolic compounds such as catechol represent a particular type of micropollutant whose high stability prevents rapid decay and metabolization in the environment. We successfully cloned a catechol 2,3-dioxygenase (C2,3O) from Pseudomonas putida mt-2 and expressed it in Escherichia coli BER2566. The biomass isolated from shake-flask fermentations was used to partially purify the enzyme. The enzyme proved unstable in clarified liquid fractions (50 mM Tris buffer, pH 7.6) and lost more than 90% of its activity over 7 h at 25 °C. 2-butoxyethanol is one of the most used glycol ether in industrial activities and the treatment of air 2-butoxyethanol-emissions become necessary. Biotechnologies are potential treatment technologies due to their low operational costs. Numerical Taxonomy of Pseudomonads Isolated from Water, Sediment and Eels

Summary A numerical taxonomic study was performed on 129 pseudomonad isolates from aquatic environments. 67 out of 98 field strains were isolated from water and sediment samples from Albufera lake, a hypereutrophic lake near Valencia, and 31 strains came from an eel hatchery. 10 clusters were formed at 84% similarity level when the Sokal-Michener similarity coefficient was used. Five clusters were designated as Pseudomonas putida (16 strains), P. fluorescens biotype I (9 strains), P. pseudoalcaligenes (11 strains), P. stutzen (4 strains) and Shewanella putrefaciens (3 strains). Isolation and characterisation of a bacterial strain degrading the herbicide sulcotrione from an agricultural soil

BACKGROUND: The dissipation kinetics of the herbicide sulcotrione sprayed 4 times on a French soil was studied using a laboratory microcosm approach. An advanced cultivation-based method was then used to isolate the bacteria responsible for biotransformation of sulcotrione. Chromatographic methods were employed as complementary tools to define its metabolic pathway. RESULTS: Soil microflora was able quickly to biotransform the herbicide (DT50 ≈ 8 days). 2-Chloro-4-mesylbenzoic acid, one of its main metabolites, was clearly detected. Data from: Evolution of bacterial life history traits is sensitive to community structure

Very few studies have experimentally assessed the evolutionary effects of species interactions within the same trophic level. Here we show that when Serratia marcescens evolve in multispecies communities, their growth rate exceeds the growth rate of the bacteria that evolved alone, whereas the biomass yield gets lower. In addition to the community effects per se, we found that few species in the communities caused strong effects on S. marcescens evolution. The results indicate that evolutionary responses (of a focal species) are different in communities, compared to species evolving alone. Invasions pose a large threat to native species, but the question of why some species are more invasive, and some communities more prone to invasions than others, is far from solved. Using ten different three-species bacterial communities, we tested experimentally if the phylogenetic relationships between an invader and a resident community and propagule pressure affect invasion probability. We found that greater diversity in phylogenetic distances between the resident community members and the invader lowered invasion success, and higher propagule pressure increased invasion success whereas phylogenetic distance had no clear effect.
