Immunomodulatory and Hematopoietic Effects of Recombinant Human Interleukin-6 in Patients with Advanced Renal Cell Cancer

1996

Interleukin-6 (IL-6) is a cytokine with pleiotropic biologic activities on B cells, T cells, and hematopoietic progenitors. The present study was undertaken to assess pharmacodynamic effects of subcutaneous administration of IL-6 on blood counts, immunologic parameters, and acute-phase reactants. Blood samples were taken from patients with advanced renal cell cancer participating in a phase II trial of recombinant human IL-6. Multiparameter FACS analyses of peripheral blood mononuclear cells were performed using antibodies against CD3, CD4, CD8, HLA-DR, CD56, CD28, CD38, CD19, sIgM, and sIgG. Serum levels of IL-10, soluble CD23 (sCD23), sCD25, IL-1 receptor antagonist protein (IL-1RA), solu…

Identification of 5,6,7,8-tetrahydropterin and 5,6,7,8-tetrahydrobiopterin in Drosophila melanogaster.

1988

Summary Using reversed-phase high-performance liquid chromatography with electrochemical detection we have demonstrated the occurrence of 5,6,7,8-tetrahydropterin and 5,6,7,8-tetrahydrobiopterin in Drosophila melanogaster . The former is the first time that has been detected in vivo . The identification has been based on the retention times, hydrodinamic voltagrams and the differential concentration in three strains of Drosophila melanogaster . Compared to the wild type, the Punch 2 mutant has diminished levels of both pteridines, whereas Henna-recessive 3 lacks completely tetrahydropterin and has increased levels of tetrahydrobiopterin, as expected according to their biochemical lesions.

Paraplangia sinespeculo, a new genus and species of bush-cricket, with notes on its biology and a key to the genera of Phaneropterinae (Orthoptera: T…

2018

Madagascar is a well-known hotspot of biodiversity. However, many Orthoptera, and especially the Tettigonioidea, belong to little-studied groups. Here we describe a new genus and species of bush-cricket reared from field-collected eggs.Paraplangiasinespeculogen. nov., sp. nov. belongs to Phaneropterinae and shares diagnostic characteristics with members of the tribe Amblycoryphini and its African subtribe Plangiina stat. nov.Paraplangia, which has a chromosome number of 31 X0, differs from other African members of the tribe and subtribe such asEurycoryphaandPlangia, which both have 29 X0. In addition to morphology, we describe the male calling song, female acoustic response, and mating beha…

Nitric oxide synthase inhibition and oxidative stress in cardiovascular diseases: Possible therapeutic targets?

2013

International audience; Nitric oxide (• NO) is synthetized enzymatically from L-arginine (L-Arg) by three NO synthase isoforms, iNOS, eNOS and nNOS. The synthesis of NO is selectively inhibited by guanidino-substituted analogs of L-Arg or methylarginines such as asymmetric dimethylarginine (ADMA), which results from protein degradation in cells. Many disease states, including cardiovascular diseases and diabetes, are associated with increased plasma levels of ADMA. The N-terminal catalytic domain of these NOS isoforms binds the heme prosthetic group as well as the redox cofactor, tetrahydrobiopterin (BH 4) associated with a regulatory protein, calmodulin (CaM). The enzymatic activity of NOS…

Reptiles use pterin and carotenoid pigments to produce yellow, orange, and red colors. These conspicuous colors serve a diversity of signaling functions, but their molecular basis remains unresolved. Here, we show that the genomes of sympatric color morphs of the European common wall lizard ( Podarcis muralis ), which differ in orange and yellow pigmentation and in their ecology and behavior, are virtually undifferentiated. Genetic differences are restricted to two small regulatory regions near genes associated with pterin [ sepiapterin reductase ( SPR )] and carotenoid [ beta-carotene oxygenase 2 ( BCO2 )] metabolism, demonstrating that a core gene in the housekeeping pathway of pterin bi…

Nitric Oxide Synthesis in Vascular Physiology and Pathophysiology

2015

Nitric oxide (NO) is produced by three NO synthase (NOS) isoforms: neuronal NOS (nNOS), inducible NOS (iNOS), and endothelial NOS (eNOS). Under physiological conditions, vascular NO is produced by eNOS and nNOS, with both playing atheroprotective roles. Under pathological conditions, iNOS can be induced and eNOS may become uncoupled. iNOS produces a large amount of NO, induces vascular dysfunction, and promotes atherogenesis. Uncoupled eNOS generates superoxide instead of NO and contributes significantly to endothelial dysfunction and atherogenesis. Major mechanisms of eNOS uncoupling include depletion of tetrahydrobiopterin, an essential co-factor for the eNOS enzyme, and deficiency of L-a…

Synonymy of two recently described taxa of Phaneropterinae (Orthoptera, Tettigoniidae)

2018

Specimens of the two taxa Terpnistriella bredoi Massa, 2017 and Pseudogoetia constanti Massa, 2017 from Democratic Republic of Congo were certainly erroneously labeled, because they resulted synonymous with neotropical taxa Vellea cruenta (Burmeister, 1838) and Philophyllia guttulata Stål, 1873.

The tropical African tribes Poreuomenini, Zeuneriini and Morgeniini with the description of a new genus (Orthoptera: Tettigoniidae: Phaneropterinae).

2018

Species currently considered belonging to the tribe Poreuomenini Brunner von Wattenwyl, 1878 are here separated; only Poreuomena Brunner von Wattenwyl, 1878 and Cestromoecha Karsch, 1893 remain in the tribe Poreuomenini, while Zeuneria Karsch, 1889 and Gravenreuthia Karsch, 1892 are transferred to the new tribe Zeuneriini Karsch, 1890. Morgenia Karsch, 1890 and Mangomaloba Sjöstedt, 1902 are moved to the new tribe Morgeniini Karsch, 1890. The new genus and species Paraporeuomena signata is described from the Tri National Sangha (TNS) (Central African Republic) and tentatively considered belonging to the tribe Poreuomenini. Its general aspect is hardly similar to that of the genus Poreuomena…

Cyclic AMP-mediated upregulation of the expression of neuronal NO synthase in human A673 neuroepithelioma cells results in a decrease in the level of…

2004

The expression level of neuronal nitric oxide synthase (nNOS) can vary depending on the (patho)physiological conditions. Here we document a marked induction of nNOS mRNA, protein, and total NO production in response to dibutyryl cyclic AMP (db-cAMP) in human A673 neuroepithelial cells. However, the upregulation of nNOS was associated with a decreased level of production of bioactive NO and by an increase in the level of generation of reactive oxygen species (ROS). ROS production could be prevented by the NOS inhibitor L-NAME, suggesting nNOS itself is involved in ROS generation. Sepiapterin supplementation of db-cAMP-treated A673 cells could restore full bioactive NO production, most likely…
