An organelle-specific protein landscape identifies novel diseases and molecular mechanisms.

Cellular organelles provide opportunities to relate biological mechanisms to disease. Here we use affinity proteomics, genetics and cell biology to interrogate cilia: poorly understood organelles, where defects cause genetic diseases. Two hundred and seventeen tagged human ciliary proteins create a final landscape of 1,319 proteins, 4,905 interactions and 52 complexes. Reverse tagging, repetition of purifications and statistical analyses, produce a high-resolution network that reveals organelle-specific interactions and complexes not apparent in larger studies, and links vesicle transport, the cytoskeleton, signalling and ubiquitination to ciliary signalling and proteostasis. Proteomics evaluation of enniatins acute toxicity in rat liver

Abstract Enniatins (ENs) are emerging mycotoxins produced by Fusarium fungi which are cytotoxic also at low concentrations due to its ionophoric properties. The aim of this study was to evaluate the hepatic toxicity of ENs exposure at different concentrations in Wistar rats through a proteomic approach. Animals were intoxicated by oral gavage with medium (EN A 256, ENA1 353, ENB 540, ENB1 296 μg/mL) and high concentrations (ENA 513, ENA1 706, ENB 1021, ENB1 593 μg/mL) of an ENs mixture and sacrificed after 8 h. Progress in ISOL target–ion source systems

2008

The heart of every ISOL (isotope separation on-line) facility is its target and ion source system. Its efficiency, selectivity and rapidity is decisive for the production of intense and pure ion beams of short-lived isotopes. Recent progress in ISOL target and ion source technology is discussed at the examples of radioactive ion beams of exotic zinc and tin isotopes that were purified by isothermal chromatography and molecular sideband separation respectively. An outlook is given to which other elements these purification methods are applicable.

Radionuclides in wastewater treatment plants: monitoring of Sicilian plants.

Three Sicilian wastewater treatment plants were monitored to assess the occurrence and the behaviour of radionuclides. Two sampling campaigns (screening and long-term) were carried out during which liquid and solid samples have been analysed. It was found that 131I mostly occurred in the samples analysed during the screening campaign (43% of the analysed samples contained 131I). High 131I specific activity was found in the mixed liquor, recycled sludge and dehydrated sludge samples. This finding was mainly due to the tendency of 131I to be associated with solid particles. During the long-term sampling campaign an influence of the sludge retention time (SRT) on the 131I behaviour was found. …

Virus diversity in a winter epidemic of acute diarrhea in France.

ABSTRACT In France, an epidemic peak of acute diarrhea is observed each winter. Previous results suggested a viral etiology for these winter epidemics. We investigated the role of enteric viruses in acute diarrhea and their molecular diversity. One hundred sixty-one patients with acute diarrhea and 45 healthy patients (controls) from the general population were given a standardized questionnaire between December 1998 and May 1999. Stool specimens were screened for group A and C rotaviruses, human caliciviruses, astroviruses, and adenovirus types 40 and 41 by reverse transcription-PCR and/or enzyme immunoassay. Photochemical sample treatment for extracts clean up in PCB analysis from sediments.

Sample purification can be considered the most polluting step of the whole analytical process for PCBs determination in sediment samples. The use of photochemical sample treatment represents an alternative methodology for extracts clean up allowing for a reduction of the used amount of organic solvents. The first application of a photochemical sample treatment for the selective removal or reduction of organic substances interfering with PCBs analyses in sediments is reported. Silica Materials Containing Cyclodextrin for Pollutant Removal

This chapter reviews the use of cyclodextrin-silica hybrid systems and cyclodextrin-functionalized silica used as adsorbents or filters for the removal of inorganic and organic pollutants from aqueous solutions in solid-phase extraction and adsorption-oriented processes. Actually, there is a need to develop efficient processes for the synthesis and application of multifunctional silica-based materials for pollutant removal by adsorption or filtration, and for sample purification and concentration using solid-phase extraction.

Protein diffusion through charged nanopores with different radii at low ionic strength

[EN] The diffusion of two similar molecular weight proteins, bovine serum albumin (BSA) and bovine haemoglobin (BHb), through nanoporous charged membranes with a wide range of pore radii is studied at low ionic strength. The effects of the solution pH and the membrane pore diameter on the pore permeability allow quantifying the electrostatic interaction between the chargedpore and the protein. Because of the large screening Debye length, both surface and bulk diffusion occur simultaneously.
