Increased serum miR-193a-5p during non-alcoholic fatty liver disease progression: Diagnostic and mechanistic relevance
2022
Background & Aims Serum microRNA (miRNA) levels are known to change in non-alcoholic fatty liver disease (NAFLD) and may serve as useful biomarkers. This study aimed to profile miRNAs comprehensively at all NAFLD stages. Methods We profiled 2,083 serum miRNAs in a discovery cohort (183 cases with NAFLD representing the complete NAFLD spectrum and 10 population controls). miRNA libraries generated by HTG EdgeSeq were sequenced by Illumina NextSeq. Selected serum miRNAs were profiled in 372 additional cases with NAFLD and 15 population controls by quantitative reverse transcriptase PCR. Results Levels of 275 miRNAs differed between cases and population controls. Fewer differences were seen wi…

Molecular analyses on HPV infections in semen
Introduction: Human Papillomaviruses (HPVs) are non-enveloped double stranded DNA viruses classified in different genera and several different genotypes. Depending on the genotypes, HPV infections can be asymptomatic or can cause from warts to malignant tumours. Some genotypes, such as HPV18 and HPV16, are considered high risk (HR) HPVs and they are the major cause of cervical cancer. HPV is highly tissue-tropic and infects epithelial cells, but it can also binds other cell types. It is known that HPV can be found in semen but its effects on spermatozoa and male reproductive system are not completely clarified. Materials and Methods: In order to study the HPV infection in semen, we develope…
Prevalence of Mycobacterium bovis in agroecosystems : analysis of potential environmental reservoirs (soil, fresh water, soil fauna and aquatic fauna…
2016
Bovine tuberculosis is an infectious disease caused by Mycobacterium bovis. This disease affects cattle, and many species of domestic and wild mammals, and humans. The circulation of the bacteria in various multi-host systems promotes the maintenance of the disease and the contamination of cattle in the vicinity. Beside direct transmission of the bacteria through the respiratory route, indirect transmission, through inhalation or ingestion of environmental matrices contaminated by an infected animal excretory, is suspected in several countries. Environmental contamination with M. bovis appears to be a crucial factor in the persistence of the infection in multi-host systems. In Côte d'Or, a F…
RNA-seq transcriptome analysis provides candidate genes for resistance to Tomato leaf cur New Delhi virus in melon
2022
[EN] Tomato leaf curl New Delhi virus (ToLCNDV) emerged in the Mediterranean Basin in 2012 as the first DNA bipartite begomovirus (Geminiviridae family), causing severe yield and economic losses in cucurbit crops. A major resistance locus was identified in the wild melon accession WM-7 (Cucumis melo kachri group), but the mechanisms involved in the resistant response remained unknown. In this work, we used RNA-sequencing to identify disease-associated genes that are differentially expressed in the course of ToLCNDV infection and could contribute to resistance. Transcriptomes of the resistant WM-7 genotype and the susceptible cultivar Piñonet Piel de Sapo (PS)(C. melo ibericus group) in ToLC…
Conceptual Design and Modeling of Fast Discharge Unit for Quench Protection of Superconducting Toroidal Field Magnets of DTT
2020
The paper deals with the modelling and simulation of a Fast Discharge Unit (FDU) for quench protection of the Toroidal Field (TF) magnets of the Divertor Tokamak Test, an experimental facility under design and construction in Frascati (Italy). The FDU is a safety key component that protects the superconducting magnets when a quench is detected through the fast extraction of the energy stored in superconducting magnets by adding in the TF magnets a dump (or discharge) resistor. In the paper, two different configurations of dump resistors (fixed and variable respectively) have been analysed and discussed. As a first result, it is possible to underline that the configuration with variable dump…
Hypoxia Positively Regulates the Expression of pH-Sensing G-Protein–Coupled Receptor OGR1 (GPR68)
2016
Background & Aims: A novel family of proton-sensing G-proteinâcoupled receptors, including ovarian cancer G-proteinâcoupled receptor 1 (OGR1) (GPR68) has been identified to play a role in pH homeostasis. Hypoxia is known to change tissue pH as a result of anaerobic glucose metabolism through the stabilization of hypoxia-inducible factor-1α. We investigated how hypoxia regulates the expression of OGR1 in the intestinal mucosa and associated cells. Methods: OGR1 expression in murine tumors, human colonic tissue, and myeloid cells was determined by quantitative reverse-transcription polymerase chain reaction. The influence of hypoxia on OGR1 expression was studied in monocytes/macrophages and…
Inter-laboratory evaluation of the ISO standard 11063 "Soil quality -- Method to directly extract DNA from soil samples"
2011
International audience; Extracting DNA directly from micro-organisms living in soil is a crucial step for the molecular analysis of soil microbial communities. However, the use of a plethora of different soil DNA extraction protocols, each with its own bias, makes accurate data comparison difficult. To overcome this problem, a method for soil DNA extraction was proposed to the International Organization for Standardization (ISO) in 2006. This method was evaluated by 13 independent European laboratories actively participating in national and international ring tests. The reproducibility of the standardized method for molecular analyses was evaluated by comparing the amount of DNA extracted, …
