Monitoring Commercial Starter Culture Development in Presence of Red Grape Pomace Powder to Produce Polyphenol-Enriched Fresh Ovine Cheeses at Indust…

2021

Red grape Nero d’Avola cultivar grape pomace powder (GPP) was applied during fresh ovine cheese production in order to increase polyphenol content. Before cheeses were produced, the bacteria of a freeze-dried commercial starter culture were isolated and tested in vitro against GPP. Two dominant strains, both resistant to GPP, were identified. Thestarter culture was inoculated in pasteurized ewe’s milk and the curd was divided into two bulks, one added with 1% (w/w) GPP and another one GPP-free. GPP did not influence the starter culture development, since lactic acid bacteria (LAB) counts were 109 CFU/g in both cheeses at 30 d. To exclude the interference of indigenous LAB, the pasteurized m…

Oxidative stress, oxidative stress-based diseases and Mediterranean diet. Raman spectroscopy monitoring of skin carotenoids in healthy individuals an…

Abstract Evaluating the level of body antioxidants may reveal a more or less severe oxidative stress condition in either healthy people or subjects with pathologies, and can be suggestive for suitable dietary interventions to possibly affect it. In this work taking advantage from the Raman spectroscopic technology to measure the level of skin carotenoids in a simple and non-invasive manner, healthy people (age 15 to 70, n=155) and women who had surgery for breast cancer (BC) (age 38 to 76, n=71) have been screened to assess to what extent an increased intake of fruit and vegetables (healthy people and patients), associated with diets based on a classical Mediterranean style and that did not…

First measurement of quarkonium polarization in nuclear collisions at the LHC

2021

The polarization of inclusive J/$\psi$ and $\Upsilon(1{\rm S})$ produced in Pb-Pb collisions at $\sqrt{s_{\rm{NN}}}=5.02$ TeV at the LHC is measured with the ALICE detector. The study is carried out by reconstructing the quarkonium through its decay to muon pairs in the rapidity region $2.5<y<4$ and measuring the polar and azimuthal angular distributions of the muons. The polarization parameters $\lambda_{\theta}$, $\lambda_{\phi}$ and $\lambda_{\theta\phi}$ are measured in the helicity and Collins-Soper reference frames, in the transverse momentum interval $2<p_{\rm T}<10$ GeV/$c$ and $p_{\rm T}<15$ GeV/$c$ for the J/$\psi$ and $\Upsilon(1{\rm S})$, respectively. The polarization parameter…

Hepatocellular cancer cell lines, Hep-3B and Hep-G2 display the pleiotropic response to resveratrol and berberine

2022

Purpose Human carcinoma cells with different p53 status exposed to a combination of bioactive substances, resveratrol and berberine, revealed different responses in cell viability via p53-dependant apoptosis pathway activation. Materials and methods Using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay, we investigated various and opposing effects in hepatocellular carcinoma cells, Hep-G2 and Hep-3B with different p53-status. Results Cells decreased in viability after treatment with dose-dependent concentrations of resveratrol and berberine. Hep-3B p53 mutants were more sensitive in comparison to the p53 wild type Hep-G2 cell line. A syne…

Apollon gene silencing induces apoptosis in breast cancer cells via p53 stabilisation and caspase-3 activation

2009

We analysed the effects of small interfering RNA (siRNA)-mediated silencing of Apollon, a member of the inhibitors of apoptosis protein family, on the proliferative potential and ability of human breast cancer cell lines to undergo apoptosis. In wild-type p53 ZR75.1 cells, Apollon knockdown resulted in a marked, time-dependent decline of cell growth and an increased rate of apoptosis, which was associated with p53 stabilisation and activation of the mitochondrial-dependent apoptotic pathway. Pre-incubation of cells with a p53-specific siRNA resulted in a partial rescue of cell growth inhibition, as well as in a marked reduction of the apoptotic response, indicating p53 as a major player in …

UV-B exposure causes DNA damage and changes in protein expression in northern pike (Esox lucius) posthatched embryos.

2012

The ongoing anthropogenically caused ozone depletion and climate change has increased the amount of biologically harmful UV-B radiation, which is detrimental to fish in embryonal stages. The effects of UV-B radiation on the levels and locations of DNA damage manifested as cyclobutane pyrimidine dimers (CPDs), heat shock protein 70 (HSP70) and p53 protein in newly hatched embryos of pike were examined. Pike larvae were exposed in the laboratory to current and enhanced doses of UV-B radiation. UV-B exposure caused the formation of CPDs in a fluence rate-dependent manner, and the CPDs were found deeper in the tissues with increasing fluence rates. UV-B radiation induced HSP70 in epidermis, and…

Digital control circuitry for the p53 dynamics in cancer cell and apoptosis

2010

Abstract Experimental work and theoretical models deduce a “digital” response of the p53 transcription factor when genomic integrity is damaged. The mutual influence of p53 and its antagonist, the Mdm2 oncogene, is closed in a feedback. This paper proposes an aerospace-based architecture for translating the p53/Mdm2/DNA damage network into a digital circuitry in which the optimal control theory is applied for obtaining the requested dynamic evolutions of some considered cell species for repairing a DNA damage. The purpose of this paper is to demonstrate the usefulness of such digital circuitry design to detect and predict the cell species dynamics for shedding light on their inner and mutua…

Molecular mechanisms of MYCN-dependent apoptosis and the MDM2-p53 pathway: an Achille’s heel to be exploited for the therapy of MYCN amplified neurob…

2012

The p53 oncosuppressor is very seldom mutated in neuroblastoma, but several mecha- nisms cooperate to its functional inactivation in this tumor. Increased MDM2 levels, due to genetic amplification or constitutive inhibition of p14ARF, significantly contribute to this event highlighting p53 reactivation as an attractive perspective for neuroblastoma treat- ment. In addition to its role in tumorigenesis, MYCN sensitizes untransformed and cancer cells to apoptosis. This is associated to a fine modulation of the MDM2-p53 pathway Indeed MYCN induces p53 and MDM2 transcription, and, by evoking a DNA damage response (DDR), it stabilizes p53 and its proapoptotic kinase Homeodomain Interacting Prote…

Measurement of the average shape of longitudinal profiles of cosmic-ray air showers at the Pierre Auger Observatory

2019

The profile of the longitudinal development of showers produced by ultra-high energy cosmic rays carries information related to the interaction properties of the primary particles with atmospheric nuclei. In this work, we present the first measurement of the average shower profile in traversed atmospheric depth at the Pierre Auger Observatory. The shapes of profiles are well reproduced by the Gaisser-Hillas parametrization within the range studied, for E>10 17.8 eV .A detailed analysis of the systematic uncertainties is performed using ten years of data and a full detector simulation. The average shape is quantified using two variables related to the width and asymmetry of the profile, and …
