Toward a Rationale for the PTC124 (Ataluren) Promoted Readthrough of Premature Stop Codons: A Computational Approach and GFP-Reporter Cell-Based Assay

2014

The presence in the mRNA of premature stop codons (PTCs) results in protein truncation responsible for several inherited (genetic) diseases. A well-known example of these diseases is cystic fibrosis (CF), where approximately 10% (worldwide) of patients have nonsense mutations in the CF transmembrane regulator (CFTR) gene. PTC124 (3-(5-(2-fluorophenyl)-1,2,4-oxadiazol-3-yl)-benzoic acid), also known as Ataluren, is a small molecule that has been suggested to allow PTC readthrough even though its target has yet to be identified. In the lack of a general consensus about its mechanism of action, we experimentally tested the ability of PTC124 to promote the readthrough of premature termination c…

Intraoperative microvascular Doppler monitoring of blood flow within a spinal dural arteriovenous fistula: a precious surgical tool. Case report.

2001

The authors describe a case of spinal arteriovenous fistula (AVF) treated by a microvauscular Doppler–assisted surgical interruption of the arterialized vein. Microvascular Doppler monitoring represents a valid, widely available, non-invasive tool that enables identification, through flow spectrum analysis, of components of this type of vascular malformation. In this case because the location of the fistula was identified prior to opening the dura only minimally invasive surgery was required. Direct recordings of the arterialized draining vein and the nidus of the fistula demonstrated a pathological spectrum caused by the arterial supply and the disturbed venous outflow in which a high-resi…

ENTREPRENEURIAL AND REGIONAL GROWTH ACTIVITY IN FINLAND

2010

Audretsch & Feldman (2004) argue that an agglomeration is a collection of localized firms with a common focus. As firms thrive, resources are attracted to the region. They state that, if entrepreneurship serves as a mechanism for knowledge spillovers, measures of entrepreneurial activity should be linked positively to regional growth performance. In Schumpeterian economics the engine of economic development is entrepreneurial innovation. Creative destruction makes way for new innovations and growth. In this study, we simultaneously examine the regional entrepreneurial activity and regional growth activity in Finland. A further aim of the study was to find out if entrepreneurial activit…

Frequency and Characterization of Movement Disorders in Anti-IgLON5 Disease

2021

Background and ObjectivesAnti-IgLON5 disease is a recently described neurologic disease that shares features of autoimmunity and neurodegeneration. Abnormal movements appear to be frequent and important but have not been characterized and are underreported. We describe the frequency and types of movement disorders in a series of consecutive patients with this disease.MethodsIn this retrospective, observational study, the presence and phenomenology of movement disorders were assessed with a standardized clinical questionnaire. Available videos were centrally reviewed by 3 experts in movement disorders.ResultsSeventy-two patients were included. In 41 (57%), the main reason for initial consult…

Urban change and regional development at the margins of Europe: an introduction

2018

Since the beginning of the nineties the urban dimension has taken a growing relevance within the EU’s regional policy. The implementation of community initiatives or other areabased projects under the structural funds has meant for several cities a tangible opportunity to start urban renewal, introduce innovative planning instruments and implement new governance relations. In the so called “less developed” regions, this process has also been accompanied by significant financial resources, giving local governments and municipalities the chance to start large infrastructure projects of metropolitan or even regional relevance. Development processes in regions and urban areas, however, have fol…

Methyl-CpG-binding domain sequencing reveals a prognostic methylation signature in neuroblastoma

2016

Accurate assessment of neuroblastoma outcome prediction remains challenging. Therefore, this study aims at establishing novel prognostic tumor DNA methylation biomarkers. In total, 396 low- and high-risk primary tumors were analyzed, of which 87 were profiled using methyl-CpG-binding domain (MBD) sequencing for differential methylation analysis between prognostic patient groups. Subsequently, methylation-specific PCR (MSP) assays were developed for 78 top-ranking differentially methylated regions and tested on two independent cohorts of 132 and 177 samples, respectively. Further, a new statistical framework was used to identify a robust set of MSP assays of which the methylation score (i.e.…

TECNICHE DI RICOSTRUZIONE DELLA SUPERFICIE POSTERIORE DEL PADIGLIONE AURICOLARE E DELLA MASTOIDE CON LEMBI LOCALI

2007

B a c k g ro u n d. Reconstruction of the posterior auricular surface and of the mastoid has never been given much attention and there are no techniques described. The authors describe two techniques for reconstruction of these areas after cancer excision with local flaps. M e t h o d s . 13 patients have been treated after cancer resection with an advancement flap from the posterior auricular surface and a bilobed flap from the mastoid. R e s u l t s. All flaps healed uneventfully with a satisfactory aesthetic outcome. No relevant complications have been o b s e r v e d . C o n c l u s i o n s.
