A genomic epidemiology investigation of yaws re-emergence and bacterial drug resistance selection

2020

Abstract Background In a longitudinal study assessing the WHO strategy for yaws eradication using mass azithromycin treatment, we observed resurgence of yaws cases with dominance of a single JG8 sequence type and emergence of azithromycin-resistant Treponema pallidum. Here, we analyse genomic changes in the bacterial population using samples collected during the study. Methods We performed whole bacterial genome sequencing directly on DNA extracted from 37 lesion swabs collected from patients on Lihir Island, Papua New Guinea, between 2013 and 2016. We produced phylogenies and correlated these with temporo-spatial information to investigate the source of new cases and the emergence of five …

2012

ABSTRACT Klebsiella pneumoniae KpQ3 is a multidrug-resistant isolate obtained from a blood culture of a patient in a burn unit in the Hospital Universitario La Paz (Madrid, Spain) in 2008. The genome contains multiple antibiotic resistance genes, including a plasmid-mediated DHA-1 cephalosporinase gene.

Impact of Superfinish Turning on Surface Integrity of Pure Copper

2011

Abstract The presented study deals with comprehension of relationships between machining cutting conditions and physical-chemical properties linked with surface integrity. A multi-physics model is presented to calculate residual stress, temperature and dislocation fields in the machined material. This model includes both thermal-mechanical, structural evolution and dynamic recristallisation approaches. The model was applied to pure copper after superfinish turning. Good agreement was found between numerical values and experimental ones. This paper also presents a work on the impact of mechanical and microstructural changes induced by machining on the electrochemical behaviour and corrosion …

Experimental and Numerical Study on Linear Friction Welding of AA2011 Aluminum Alloy

2014

Linear Friction Welding (LFW) is a solid-state joining process used for non-axisymmetric components. LFW involves joining of materials through the relative motion of two components undergoing an axial force. In the process, the heat source is given by the frictional forces work decaying into heat and determining a local softening of the material and eventually the needed bonding conditions. In the paper, an experimental and numerical campaign is proposed for AA2011 aluminum alloys welding. Different case studies are considered with fixed oscillation frequency and varying pressure at the interface between the specimens. Constant oscillation amplitude and specimens geometry is used. The calcu…

Effect of different aqueous solutions of pure salts and salt mixtures in reverse electrodialysis systems for closed-loop applications

2018

Abstract Reverse Electrodialysis (RED) in a closed-loop arrangement is a viable way to convert low-grade heat into electric power. The present work experimentally investigates the use of pure salt- and equimolar two salts-water solutions as feeds in a lab-scale RED unit. RED performances were analysed in terms of Open Circuit Voltage (OCV), stack resistance and corrected power density. The pure salts and the mixtures employed were chosen via a computational analysis. Effect of feed solution velocity and concentration was investigated. Results concerning the pure salt-water experiments show that NH4Cl is the most performing salt in the concentration range probed, while higher power density v…

Effect of exercises with weight vests and a patient education programme for women with osteopenia and a healed wrist fracture: a randomized, controll…

2015

Background Exercise programmes have shown to be important for the prevention of fractures in patients with established osteoporosis. However, few studies have evaluated the effect of such programmes for women with low bone mineral density (BMD) (osteoporosis or osteopenia) who have already suffered a fracture. Studies have indicated that exercise programmes concentrating on muscular strength and dynamic balance have a positive effect on significant risk factors for falls such as quadriceps strength and balance. The aim of the present study was to assess the effect of a 6-month exercise programme and a patient education component (OsteoACTIVE) on quadriceps strength, BMD, dynamic balance, wa…

Time-dependent current-voltage characteristics of Al/p-CdTe/Pt x-ray detectors

2012

Current-voltage (I-V) characteristics of Schottky Al/p-CdTe/Pt detectors were investigated in dark and at different temperatures. CdTe detectors with Al rectifying contacts, very appealing for high resolution x-ray and gamma ray spectroscopy, suffer from bias-induced polarization phenomena which cause current increasing with the time and severe worsening of the spectroscopic performance. In this work, we studied the time-dependence of the I-V characteristics of the detectors, both in reverse and forward bias, taking into account the polarization effects. The I-V measurements, performed at different time intervals between the application of the bias voltage and the measurement of the current…

Cytotoxicity and modes of action of three naturally occurring xanthones (8-hydroxycudraxanthone G, morusignin I and cudraxanthone I) against sensitiv…

2014

Abstract Background Resistance of cancer to chemotherapy remains a challenging issue for scientists as well as physicians. Naturally occurring xanthones possess a variety of biological activities such as anti-inflammatory, anti-bacterial, and anti-cancer effects. The present study was aimed at investigating the cytotoxicity and the modes of action of three naturally occurring xanthones namely, morusignin I (1), 8-hydroxycudraxanthone G (2) and cudraxanthone I (3) against a panel of nine cancer cell lines, including various sensitive and drug-resistant phenotypes. Methods The cytotoxicity of the compounds was determined using a resazurin reduction assay, whereas the caspase-Glo assay was use…

Cytotoxicity of compounds from Xylopia aethiopica towards multi-factorial drug-resistant cancer cells.

2015

Abstract Introduction Multidrug resistance (MDR) in cancer represent a major hurdle in chemotherapy. Previously, the methanol extract of the medicinal spice Xylopia aethiopica displayed considerable cytotoxicity against multidrug resistant (MDR) cancer cell lines. Methods The present study was designed to assess the cytotoxicity of compounds, 16 α -hydroxy- ent -kauran-19-oic acid ( 2 ), 3,4′,5-trihydroxy-6″,6″-dimethylpyrano[2,3-g]flavone ( 3 ), isotetrandrine ( 5 ) and trans -tiliroside ( 6 ) derived from the methanol crude extract of Xylopia aethiopica against 9 drug-sensitive and -resistant cancer cell lines. The resazurin reduction assay was used to evaluate the cytotoxicity of these c…

Cytotoxicity of the crude extract and constituents of the bark of Fagara tessmannii towards multi-factorial drug resistant cancer cells.

2019

Abstract Ethnopharmacological relevance Fagara tessmannii Engl. is an African medicinal plant used in Cameroonian traditional medicine to treat various types of cancers. Aim of the study This work was designed to determine the cytotoxicity of the crude extract (FTB), fractions (FTBa-d) and compounds isolated from the bark of Fagara tessmannii, namely lupeol (1), fagaramide (2), zanthoxyline (3), hesperidin (4), nitidine chloride (5), fagaridine chloride (6), and β-sitosterol-3-O-β-D-glucopyranoside (7). The study was extended to the mode of induction of apoptosis by FTB, compounds 5 and 6. Materials and methods The resazurin reduction assay was used to evaluate the cytotoxicity of samples. …
