Small interfering RNAs (siRNAs) have the potential to be of therapeutic value for many human diseases. So far, however, a serious obstacle to their therapeutic use is represented by the absence of appropriate delivery systems able to protect them from degradation and to allow an efficient cellular uptake. In this work we developed a siRNA delivery system based on inulin (Inu), an abundant and natural polysaccharide. Pharmacokinetics of triamterene after i.v. administration to man: determination of bioavailability.

With a new formulation, which made intravenous infusion of triamterene (TA) possible, plasma levels and urinary excretion rates of TA and its main metabolite (OH-TA-ester) were measured in a randomized, cross-over trial in 6 healthy volunteers given triamterene 10 mg i.v. and 50 mg p.o. TA and OH-TA-ester were determined by densitometric measurement of native fluorescence after thin layer chromatography. Distribution volumes of the central compartment of TA and OH-TA-ester were 1.49 l/kg and 0.11 l/kg, respectively. Terminal half-lives were 255 min for TA and 188 min for OH-TA-ester after i.v. administration. For TA total plasma clearance was 4.5 l/min and renal plasma clearance 0.22 l/kg. …

Introduction: Ernest syndrome involves the stylomandibular ligament. It is characterised by pain in the preauricular area and mandibular angle, radiating to the neck, shoulder, and eye on the same side, and associated with pain during palpation of that ligament. Abstract Two capillary electrophoresis methods have been developed for the direct determination of triamterene and its main metabolite hydroxytriamterene sulfate in human urine. Analytes were detected using conventional UV detection as well as laser-induced fluorescence (LIF) detection with an HeCd-laser operating at a wavelength of 325 nm. The results of both detection techniques were compared. Indeed, the limit of quantification was eightfold lower using LIF detection (50 ng/ml) in comparison to UV detection (400 ng/ml). Abstract Solution equilibrium studies on the Cu(II)–polyamine–methionine ternary systems (polyamine: ethylenediamine (en), diethylenetriamine (dien), N,N,N′,N′′,N′′-pentamethyldiethylenetriamine (Me5dien)) have been performed by pH-potentiometry, UV–Vis spectrophotometry and EPR methods. The obtained results suggest the formation of the mixed-ligand complexes with [Cu(A)(Met)]+ stoichiometry in all studied systems. Our spectroscopic results indicate the tetragonal geometry for [Cu(en)(Met)]+, the geometry slightly deviated from square pyramidal for [Cu(dien)(Met)]+ and strongly deviated from square pyramidal towards trigonal bipyramidal for the [Cu(Me5dien)(Met)]+ species. Abstract The interaction of five open chain polyamines (ethylenediamine (en), diethylenetriamine (dien), trietylenetetramine (trien), tetraethylenepentamine (tetren), spermine (sper)) with the major components of seawater was studied potentiometrically at 25°C, in an artificial seawater (containing Na+, K+, Mg2+, Ca2+, Cl− and SO42−) at different salinities (5–45‰). This study was carried out to evaluate the influence of long-term treatment with doxorubicin (DXR) (4mg/kg IV for 5 weeks) on heart and liver lysosomes of mice. We evaluated the variations in both total and "sedimentable" enzyme activity of cathepsin D, which is the major endopeptidase of myocites and probably involved in physiologic and pathologic degradation of actomyosin and mitochondria, and that of acid phosphatase, which is more prominent in interstitial cells. Our results show that marked changes occur in both total and sedimentable enzyme activity of cathepsin D in the heart of treated animals and to a lesser extent in the liver.
