Cisplatin sensitivity is related to late DNA damage processing and checkpoint control rather than to the early DNA damage response

The present study aimed at elucidating mechanisms dictating cell death triggered by cisplatin-induced DNA damage. We show that CL-V5B hamster mutant cells, a derivative of V79B, are hypersensitive to cisplatin-induced apoptotic death. CL-V5B cells are characterized by attenuated cisplatin-induced early (2-6 h) stress response, such as phosphorylation of stress-activated protein kinases (SAPK/JNK), ATM and Rad3-related (ATR) protein kinase, histone H2AX and checkpoint kinase-1 (Chk-1). Human FANCC cells also showed a reduced phosphorylation of H2AX and SAPK/JNK at early time point after cisplatin treatment. In vitro fusion of phagosomes with different endocytic organelles from J774 macrophages.

We describe novel biochemical and electron microscopy assays to investigate in vitro fusion of latex bead phagosomes with three different endocytic organelle fractions from J774 macrophages. After formation, early phagosomes fuse avidly with early and late endosomes and for a longer period of time with lysosomes, but they subsequently become fusion-incompetent. The fusion of early, but not late, phagosomes with all three endocytic fractions could be significantly stimulated by Rab5. In contrast to other cell types investigated, this Rab is uniquely enriched on both early and late endosomes in J774 macrophages. Previous studies have shown that certain strains of herpes simplex viruses type 1 (HSV-1) are able to induce “fusion from without” (FFWO) which means no transcription or translation of the viral genome happens. The main determinants for FFWO in BHK cells are mutations in the C-terminal part of gB-1. But single mutations in this part of the genome are not sufficient to transfer the FFWO phenotype also to Vero cells. Here, we report that FFWO of HSV strains indeed need additional mutations in the N-terminal part of gD in order to produce the FFWO phenotype in BHK and Vero cells. AbstractUpon receptor-mediated activation, the gp41 hydrophobic, conserved fusion peptide inserts into the target membrane and promotes the kind of perturbations required for the progression of the HIV-cell fusion reaction. Using a synthetic combinatorial library we have identified all d-amino acid hexapeptide sequences that inhibited the fusion peptide capacity of perturbing model membranes. Two hexapeptides that effectively inhibited the fusion peptide in these systems were subsequently shown to inhibit cell–cell fusion promoted by gp41 expressed at cell surfaces. The structural behaviour of reinforced concrete columns strengthened with a system made up of fibre nets embedded in an inorganic stabilized cementitious matrix was investigated. Specimens with circular and square cross-section were cast and subjected to monotonic uniaxial compression to investigate the efficiency of a Polypara-phenylene-benzo-bisthiazole (PBO) Fibre Reinforced Cementitious Mortars (FRCM) system in increasing both carrying capacity and ductility. International audience; Aging is frequently associated with a decreased postural stability, essentially after 60 years, leading to an increased risk of falling. In this article we propose to highlight the influence of the aging of the neuromuscular system on postural stability when standing upright. To maintain balance while standing upright, human needs to control the activity of ankle muscles and particularly the plantar flexors. Generalized centro-invertible matrices with applications

Centro-invertible matrices are introduced by R.S. Wikramaratna in 2008. For an involutory matrix R, we define the generalized centro-invertible matrices with respect to R to be those matrices A such that RAR = A^−1. We apply these matrices to a problem in modular arithmetic. Specifically, algorithms for image blurring/deblurring are designed by means of generalized centro-invertible matrices. In addition, if R1 and R2 are n × n involutory matrices, then there is a simple bijection between the set of all centro-invertible matrices with respect to R1 and the set with respect to R2.

Sono stati analizzati in modo approfondito, fornendone un catalogo dettagliato, i frammenti ceramici provenienti dagli strati di fondazione delle mura di età arcaica, studiati nell'ambito del loro contesto di scavo.
