Identification of N- and C-terminal Amino Acids of Lhca1 and Lhca4 Required for Formation of the Heterodimeric Peripheral Photosystem I Antenna LHCI-…

2002

Apoproteins of higher plant light-harvesting complexes (LHC) share considerable amino acid sequence identity/similarity. Despite this fact, they occur in different oligomeric states (i.e., monomeric, dimeric, and trimeric). As a step toward understanding the underlying structure requirements for different oligomerization behavior, we analyzed whether amino acids at the N- and C-termini of Lhca1 and Lhca4 are involved in the formation of the heterodimeric LHCI-730. Using altered proteins produced by deletion or site-directed mutagenesis for reconstitution, we were able to identify amino acids required for the assembly of LHCI-730. At the N-terminus of Lhca1, W4 is involved in heterodimerizat…

Do dynamic effects play a significant role in enzymatic catalysis? A theoretical analysis of formate dehydrogenase.

2010

A theoretical study of the protein dynamic effects on the hydride transfer between the formate anion and nicotinamide adenine dinucleotide (NAD + ), catalyzed by formate dehydrogenase (FDH), is presented in this paper. The analysis of free downhill molecular dynamic trajectories, performed in the enzyme and compared with the reaction in aqueous solution, has allowed the study of the dynamic coupling between the reacting fragments and the protein or the solvent water molecules, as well as an estimation of the dynamic effect contribution to the catalytic effect from calculation of the transmission coefficient in the enzyme and in solution. The obtained transmission coefficients for the enzyme…

Straightforward Regio- and Diastereoselective Synthesis, Molecular Structure, Intermolecular Interactions and Mechanistic Study of Spirooxindole-Engr…

2021

Straightforward regio- and diastereoselective synthesis of bi-spirooxindole-engrafted rhodanine analogs 5a–d were achieved by one-pot multicomponent [3 + 2] cycloaddition (32CA) reaction of stabilized azomethine ylide (AYs 3a–d) generated in situ by condensation of L-thioproline and 6-chloro-isatin with (E)-2-(5-(4-chlorobenzylidene)-2,4-dioxothiazolidin-3-yl)-N-(2-morpholinoethyl)acetamide. The bi-spirooxindole-engrafted rhodanine analogs were constructed with excellent diastereo- and regioselectivity along with high chemical yield. X-ray crystallographic investigations for hybrid 5a revealed the presence of four contiguous stereocenters related to C11, C12, C19 and C22 of the spiro struct…

DNA interaction of CuII, NiII and ZnII functionalized salphen complexes: studies by linear dichroism, gel electrophoresis and PCR.

2013

The interaction of salphen-type NiII, CuII and ZnII complexes with native DNA was investigated by exploiting linear dichroism experiments. The NiII complex behaves as a typical intercalator, binding strongly and stiffening and unwinding the DNA. The strength of the DNA interaction is slightly weaker for the copper complex and much weaker for the zinc complex. Plasmid-DNA gel electrophoresis experiments indicated that while CuII and ZnII complexes do not induce the unwinding of supercoiled DNA, the NiII complex has a nuclease activity without the addition of external agents. On the other hand, as shown in the PCR assays, we demonstrate that, at the used concentrations, only the CuII complex …

Solid-state cis–trans isomerism in bis(oxamato)palladate(ii) complexes: synthesis, structural studies and catalytic activity

2014

A new generation of bis(oxamato)palladate(II) monomeric complexes has been prepared by using N-2,6-dimethylphenyloxamate (2,6-Me2pma) as the ligand. Four alkaline salts of the complex, namely {[Na(H2O)]2trans-[PdII(2,6-Me2pma)2]}n (1a), {[Na4(H2O)2]cis-[PdII(2,6-Me2pma)2]2}n (1b), {[K4(H2O)3]cis-[PdII(2,6-Me2pma)2]2}n (2), {[Rb4(H2O)3]cis-[PdII(2,6-Me2pma)2]2}n (3) and {[Cs6(H2O)7]trans-[PdII(2,6-Me2pma)2]2cis-[PdII(2,6-Me2pma)2]}n·3nH2O (4), were obtained and structurally characterized by single crystal X-ray diffraction. Both the cis and trans stereoisomers of the [PdII(2,6-Me2pma)2]2− complex anion were isolated in the solid state, in a cation-dependent manner. The trans-isomer as the so…

N-sulfinyl amines as a nitrogen source in the asymmetric intramolecular aza-Michael reaction: total synthesis of (-)-pinidinol.

2010

N-Sulfinyl amines have been successfully employed as nitrogen nucleophiles for the asymmetric intramolecular aza-Michael reaction. The synthetic strategy involves a cross-metathesis reaction followed by the Michael-type cyclization, either in a base-catalyzed two-step procedure or in a tandem fashion. The developed methodology allows access to chiral substituted pyrrolidines and piperidines bearing one or two stereocenters and it has been applied to the synthesis of the piperidine alkaloid (-)-pinidinol.

Diastereoselective Michael Addition of (S)-Mandelic Acid Enolate to Nitroalkenes. Enantioselective Synthesis of α-Hydroxy-α,β-diaryl-γ-lactams.

2004

Abstract The reaction of the lithium enolate of the ( S , S )- cis -1,3-dioxolan-4-one derived from optically active ( S )-mandelic acid and pivalaldehyde with several aromatic nitroalkenes in the presence of HMPA proceeds readily to give the corresponding Michael adducts in good yields and diastereoselectivities. Reduction of the nitro group with Zn/HCl/EtOH/H 2 O with concomitant intramolecular aminolysis of the acetal moiety leads directly to enantiomerically pure α-hydroxy-α,β-diaryl-γ-lactams.

Molecular structure of the compound [Rh2(O2CCH3)3{(C6H4)P(BrC6H4-1,2)(C6H5)} · (HO2CCH3)2].Kinetic study of the exchange reaction of acetate groups w…

1993

Abstract The compound [Rh 2 (O 2 CCH 3 ) 3 {(C 6 H 4 )P(BrC 6 H 4 -1,2) (C 6 H 5 )} · (HO 2 CCH 3 ) 2 ] has been isolated in high yield from the thermal reaction of dirhodium tetraacetate and the phosphine P(BrC 6 H 4 -1,2) (C 6 H 5 ) 2 in acetic acid. The structure of this compound has been determined by X-ray diffraction; it crystallizes in the P 1 (triclinic) space group and contains three acetate groups bridging a Rh 2 4+ unit the RhRh distance being 2.432(1)A; the fourth bridging ligand is an ortho -bromophenyldiphenylphosphine metallated at one of the ortho positions of the unsubstituted phenyl rings. Two molecules of acetic acid occupy the axial coordination positions. Stepwise exch…

Lewis Acid Mediated Domino Reaction between 2-Cyclohexenone and Methyl Azide - A DFT Study

2005

We have studied the Lewis acid mediated domino reaction between 2-cyclohexenone and methyl azide at the B3LYP/6-31G* and B3LYP/6-31+G**//B3LYP/6-31G* levels of theory. A complete survey of the potential energy surface is presented, and the structures of the stationary points and the corresponding molecular mechanism are discussed. The calculations suggest that the product, a cyclopentanone derivative, is formed by means of a domino process that comprises three consecutive reactions. The first and rate-limiting step is a concerted 1,3-dipolar cycloaddition between the Lewis acid coordinated cyclohexenone and methyl azide that yields the corresponding bicyclic 1,2,3-triazoline. The second rea…
