Soluble GARP has potent antiinflammatory and immunomodulatory impact on human CD4+ T cells

2013

Glycoprotein A repetitions predominant (GARP) is expressed on the surface of activated human regulatory T cells (Treg) and regulates the bioavailability of transforming growth factor-β (TGF-β). GARP has been assumed to require membrane anchoring. To investigate the function of GARP in more detail, we generated a soluble GARP protein (sGARP) and analyzed its impact on differentiation and activation of human CD4⁺ T cells. We demonstrate that sGARP efficiently represses proliferation and differentiation of naïve CD4⁺ T cells into T effector cells. Exposure to sGARP induces Foxp3, decreases proliferation and represses interleukin (IL)-2 and interferon-γ production, resulting in differentiation …

Antibodies against tumor necrosis factor (TNF) induce T-cell apoptosis in patients with inflammatory bowel diseases via TNF receptor 2 and intestinal…

2011

Background & Aims The anti–tumor necrosis factor (TNF) antibodies infliximab, adalimumab, and certolizumab pegol have proven clinical efficacy in Crohn's disease. Here, we assessed the effects of anti-TNF antibodies on apoptosis in inflammatory bowel disease (IBD). Methods CD14 + macrophages and CD4 + T cells were isolated from peripheral blood and lamina propria mononuclear cells from patients with IBD and control patients. Cell surface markers and apoptosis were assessed by immunohistology and fluorescence-activated cell sorting techniques. Results Lamina propria CD14 + macrophages showed significantly more frequent and higher membrane-bound TNF (mTNF) expression than CD4 + T cells in IBD…

IL-28A Is a Key Regulator of T-Cell–Mediated Liver Injury via the T-Box Transcription Factor T-Bet

2006

Background & Aims: T-cell–mediated fulminant hepatitis is a potentially life-threatening event for which the underlying pathogenic mechanisms are not fully understood. Here, we demonstrate a key regulatory role of IL-28A in T-cell–mediated hepatitis. Methods: We cloned the murine IL-28A gene by reverse-transcription polymerase chain reaction, assessed the effects of recombinant IL-28A, and generated IL-28A–transgenic mice. Results: IL-28A induced TH1 cytokine production by CD4+ T lymphocytes in a T-bet–dependent manner and was up-regulated in a murine model of T-cell–mediated hepatitis upon Con A administration. In vivo, CD4+ T cells from newly created IL-28A–transgenic animals revealed an …

Embedded neural network system for microorganisms growth analysis

2020

This study presents autonomous system for microorganisms’ growth analysis in laboratory environment. As shown in previous research, laser speckle analysis allows detecting submicron changes of substrate with growing bacteria. By using neural networks for speckle analysis, it is possible to develop autonomous system, that can evaluate microorganisms’ growth by using cheap optics and electronics elements. System includes embedded processing module, CMOS camera, 670nm laser diode and optionally WiFi module for connecting to external image storage system. Due to small size, system could be fully placed in laboratory incubator with constant humidity and temperature. By using laser diode, Petri d…

Extracorporeal shock waves increase markers of cellular proliferation in primary bronchial fibroblasts of COPD patients

2019

COPD is due to a remodeling of small airways and destruction of the lung parenchyma with loss of the alveolar attachment as a result of pulmonary emphysema. Treatment of connective tissue fibroblasts with extracorporeal shock waves (ESW) increases their cellular proliferation and differentiation. To date no studies are available on ESW treatment of human primary bronchial fibroblasts from COPD and control subjects. Primary bronchial fibroblasts were obtained from bronchial biopsies from three patients with mild/moderate COPD and 3 control smokers with normal lung function. After stabilization for 24h, 1 ml of cell suspension (106 cells) from COPD and controls was treated in 2 ml tubes with …

The frequent exacerbator in real-life: an analysis of the DACCORD study

2019

Introduction: Exacerbation prevention is a major treatment goal in COPD. Hence, exacerbations are a decisive diagnostic and prognostic factor when it comes to therapy. The non-interventional prospective DACCORD study was following COPD patients for 2 years. Here, we compare patients with frequent exacerbations, non-frequent exacerbations and no exacerbations in the 6 months prior to study entry. Methods: DACCORD recruited COPD patients following a change or initiation of COPD maintenance. Data collected at baseline and every 3 months include exacerbations and COPD medication use. COPD Assessment Test (CAT) data were collected anually. Results: Out of 6527 patients, 7.1% were frequent exacer…

Examination of robust D-stability of TCP-DCR protocol

2012

This paper presents non-linear mathematical model of a computer network with a part of wireless network. The article contains an analysis of the stability of the network based on TCP-DCR, which is a modification of the traditional TCP. Block diagram of the network model was converted to a form in order to investigate the D-stability using the method of the space of uncertain parameters. Robust D-stability is calculated for constant delays values.

Adaptive Low Priority Packet Marking for Better TCP Performance

2003

This paper proposes a packet marking scheme for TCP traffic. Unlike previous literature work, in our scheme the majority of TCP packets are transmitted as high priority. The role of a low priority packet appears that of a probe, whose goal is to early discover network congestion conditions. Low priority packets are marked according to an adaptive marking algorithm. Numerical results show that our scheme provides improved throughput/delay performance.

Wages, prices, and technology in early Catalan industrialization

2015

Catalonia was the only Mediterranean region among the early followers of the British industrial revolution. The roots of this process can be traced back to the seventeenth and eighteenth centuries when the Catalan economy became integrated into international trade, and a successful printed calico industry concentrated in the city of Barcelona. Although the factory system was largely adopted by the cotton industry in the 1840s, the diffusion of the spinning jenny in Catalonia had occurred earlier, in the 1790s. In line with Allen, this article explores whether relative factor prices played a role in the widespread adoption of the spinning jenny in Catalonia. First, series of real wages in Ba…
