The effect of biological sealants and adhesive treatments on matrix metalloproteinase expression during renal injury healing

2017

Background Renal injuries are relatively common in cases of abdominal trauma. Adhesives and sealants can be used to repair and preserve damaged organs. Using a rat model, this study explores the activity of different matrix metalloproteinases (MMP) during the healing of renal injuries treated by two biological adhesives (TachoSil and GelitaSpon) and a new synthetic elastic cyanoacrylate (Adhflex). Methods Renal traumatic injuries were experimentally induced in 90 male Wistar rats by a Stiefel Biopsy Punch in the anterior aspect of the left kidney. Animals were divided into five groups: 1, sham non-injured (n = 3); 2, non-treated standard punch injury (n = 6); 3, punch injury treated with Ta…

Physiologically based metformin pharmacokinetics model of mice and scale-up to humans for the estimation of concentrations in various tissues

2020

Metformin is the primary drug for type 2 diabetes treatment and a promising candidate for other disease treatment. It has significant deviations between individuals in therapy efficiency and pharmacokinetics, leading to the administration of an unnecessary overdose or an insufficient dose. There is a lack of data regarding the concentration-time profiles in various human tissues that limits the understanding of pharmacokinetics and hinders the development of precision therapies for individual patients. The physiologically based pharmacokinetic (PBPK) model developed in this study is based on humans’ known physiological parameters (blood flow, tissue volume, and others). The missing tissue-s…

Cardiac and renal dysfunction is associated with progressive hearing loss in patients with Fabry disease.

2017

Background Fabry disease (FD) is an X-linked recessive hereditary lysosomal storage disorder which results in the accumulation of globotriaosylceramid (Gb3) in tissues of kidney and heart as well as central and peripheral nervous system. Besides prominent renal and cardiac organ involvement, cochlear symptoms like high-frequency hearing loss and tinnitus are frequently found with yet no comprehensive data available in the literature. Objective To examine hearing loss in patients with FD depending on cardiac and renal function. Material and methods Single-center study with 68 FD patients enrolled between 2012 and 2016 at the Department of Oto-Rhino-Laryngology, Plastic, Aesthetic and Reconst…

High blood pressure responders show largest increase in heartbeat perception accuracy after post-learning stress following a cardiac interoceptive le…

2020

Mental disorders with physical symptoms, e.g. somatic symptom disorder, are characterized by altered interoceptive accuracy (IAc), which can be explained by individual differences in interoceptive learning (IL). We investigated if stress facilitates IL. Seventy-three healthy participants performed a heartbeat counting task (HCT: T1) and a heartbeat perception training (HBPT). After exposure to a socially-evaluated cold pressor stress test (SECPT; n = 48) or a control condition (n = 25), two more HCTs were performed (T2: 30 min after SECPT; T3: 24 h later). After the HBPT, all participants showed an increase in IAc. We separated the stress group into high vs. low systolic blood pressures (SB…

Integrated and diurnal indices of maternal pregnancy cortisol in relation to sex-specific parasympathetic responsivity to stress in infants.

2020

Maternal hypothalamic-pituitary-adrenal (HPA) axis activity may prenatally program sex-specific stress response pathways. We investigated associations between maternal cortisol during pregnancy and infant parasympathetic responsivity to stress among 204 mother-infant pairs. Cortisol indices included 3(rd) trimester hair cortisol, as well as diurnal slope and area under the curve, derived from saliva samples collected during pregnancy. Mother-infant dyads participated in the Repeated Still-Face Paradigm (SFP-R) at age 6 months. We calculated respiration-adjusted respiratory sinus arrhythmia (RSA(c)), an indicator of parasympathetic activation, from infant respiration and cardiac activity mea…

Automatic classification of tissues on pelvic MRI based on relaxation times and support vector machine

2019

International audience; Tissue segmentation and classification in MRI is a challenging task due to a lack of signal intensity standardization. MRI signal is dependent on the acquisition protocol, the coil profile, the scanner type, etc. While we can compute quantitative physical tissue properties independent of the hardware and the sequence parameters, it is still difficult to leverage these physical properties to segment and classify pelvic tissues. The proposed method integrates quantitative MRI values (T1 and T2 relaxation times and pure synthetic weighted images) and machine learning (Support Vector Machine (SVM)) to segment and classify tissues in the pelvic region, i.e.: fat, muscle, …

Effect of mountain ultramarathon distance competition on biochemical variables, respiratory and lower-limb fatigue

2020

The study aimed at assessing the acute physiological effects of running a 65-km vs a 107- km mountain ultramarathon. Nineteen athletes (15 males and 4 females) from the shorter race and forty three athletes (26 males and 17 females) from the longer race were enrolled. Body weight, respiratory and lower limb strength were assessed before and after the race. Blood samples were obtained before, after and 24-h post-race. Body weight loss did not differ between races. A decrease in squat jump height (p<0.01; d = 1.4), forced vital capacity (p<0.01; d = 0.5), forced expiratory volume in 1 s (p<0.01; d = 0.6), peak inspiratory flow (p<0.01; d = 0.6) and maximal inspiratory pressure (p<0.01; d = 0.…

Socio-demographic, health-related, and individual correlates of diagnostic self-testing by lay people: Results from a representative survey in Germany

2017

Introduction A broad range of self-tests (testing for e.g. HIV, cancer, hepatitis B/C) have become available and can be conducted by lay consumers without the help of a health professional. The aims of this study were to (a) investigate the prevalence of self-testing, (b) identify the most frequently used self-tests, and (c) explore the associations between socio-demographic, health-related and individual factors with self-testing. Methods A face-to-face plus paper-pencil cross-sectional survey was conducted. The sample consisted of 2.527 respondents who were representative of the German population in terms of the age, sex, and residence. Basic descriptive statistics and univariate logistic…

The cortisol awakening response and memory performance in older men and women.

2012

Summary The activity and regulation of the hypothalamus-pituitary-adrenal axis has been related to cognitive decline during aging. This study investigated whether the cortisol awakening response (CAR) is related to memory performance among older adults. The sample was composed of 88 participants (44 men and 44 women) from 55 to 77 years old. The memory assessment consisted of two tests measuring declarative memory (a paragraph recall test and a word list learning test) and two tests measuring working memory (a spatial span test and a spatial working memory test). Among those participants who showed the CAR on two consecutive days, we found that a greater CAR was related to poorer declarativ…

Chronic psychosocial stress and antidepressant treatment in tree shrews: time-dependent behavioral and endocrine effects.

1999

Abstract Social defeat has been shown to cause a number of behavioral, physiological, and central nervous changes in male tree shrews. The present study was designed to assess: (i) a potential time lag in the occurrence of behavioral alterations (locomotor activity, self-grooming, marking behavior, food and water intake, and avoidance behavior) after stress and long-term antidepressant treatment; and (ii) to investigate potential interactions between behavioral and endocrine variables (urinary cortisol and norepinephrine). Male tree shrews were submitted to chronic psychosocial stress for 39 days. In this paradigm, the stress-induced behavioral and endocrine alterations in subordinate anima…
