Combined Targeting of the Menin-MLL1 Chromatin Complex and FLT3 As a Novel Therapeutic Concept Against NPM1 Mutant or MLL-Rearranged AML with Mutated…

2019

NPM1mutant (NPM1mut) and MLL1-rearranged (MLL-r) acute myeloid leukemias (AMLs) exhibit aberrant expression of HOX and MEIS1 transcription factors and commonly harbor an activating mutation in the receptor tyrosine kinase FLT3. Pharmacologic inhibition of the menin-MLL1 complex reverses leukemogenic gene expression including MEIS1 and FLT3 and represents a therapeutic opportunity for the treatment of these leukemias. Here, we investigate the contribution of the menin-MLL1 complex to leukemic FLT3 signaling and assess the therapeutic potential of dual menin-MLL1 and FLT3 targeting. First, we performed RNA sequencing to delineate transcriptional changes associated with menin-MLL1 inhibition (…

2015

This paper deals with the judicial identification of corpses in nineteenth-century France. The case of the widow Houet (1833) is particularly interesting for this purpose because two contrasting techniques of identification were employed: forensic medicine and phrenology. Mateu Orfila, dean of the Paris Faculty of Medicine, had recently developed a quantitative method for the identification of corpses, which was later regarded as a landmark in nineteenth-century forensic medicine. Pierre-Marie Dumoutier, an outstanding member of the Society of Phrenology, analysed the skull and offered surprising data about the personality of the widow. The episode stirred up controversies on the evidentiar…

Effects of ouabain on human bronchial muscle in vitro

2003

The effects of ouabain, an inhibitor of the plasmalemmal Na(+)/K(+)-ATPase activity, were examined in human isolated bronchus. Ouabain produced concentration-dependent contraction with -logEC(50)=7.16+/-0.11 and maximal effect of 67+/-4% of the response to acetylcholine (1 mM). Ouabain (10 microM)-induced contraction was epithelium-independent and was not depressed by inhibitors of cyclooxygenase and lipoxygenase, antagonists of muscarinic, histamine H(1)-receptors and alpha-adrenoceptors, or neuronal Na(+) channel blockade. The inhibition of ouabain contraction in tissues bathed in K(+)-free medium, and the inhibition by ouabain of the K(+)-induced relaxation confirm that the contractile a…

Effects of fenspiride on human bronchial cyclic nucleotide phosphodiesterase isoenzymes: functional and biochemical study.

1998

We have investigated the role of human bronchial cyclic nucleotide phosphodiesterases in the effects of fenspiride, a drug endowed with bronchodilator and anti-inflammatory properties. Functional studies on human isolated bronchi showed that fenspiride (10(-6)-3 x 10(-3) M, 30 min) induced a shift to the left of the concentration-response curves for isoprenaline and sodium nitroprusside with -logEC50 values of 4.1+/-0.1 (n = 7) and 3.5+/-0.2 (n = 8), respectively. Biochemical studies were carried out on three human bronchi in which separation of cyclic nucleotide phosphodiesterase isoenzymes was performed by ion exchange chromatography followed by determination of phosphodiesterase activity…

Multiscaling and the classification of continuous phase transitions

1992

Multiscaling of the free energy is obtained by generalizing the classification of phase transitions proposed by Ehrenfest. The free energy is found to obey a new generalized scaling form which contains as special cases standard and multiscaling forms. The results are obtained by analytic continuation from the classification scheme of Ehrenfest.

Critical point Higgs inflation in the Palatini formulation

2021

We study Higgs inflation in the Palatini formulation with the renormalisation group improved potential in the case when loop corrections generate a feature similar to an inflection point. Assuming that there is a threshold correction for the Higgs quartic coupling $\lambda$ and the top Yukawa coupling $y_t$, we scan the three-dimensional parameter space formed by the two jumps and the non-minimal coupling $\xi$. The spectral index $n_s$ can take any value in the observationally allowed range. The lower limit for the running is $\alpha_s>-3.5\times10^{-3}$, and $\alpha_s$ can be as large as the observational upper limit. Running of the running is small. The tensor-to-scalar ratio is $2.2\tim…

Pinning down the strength function for ordinary muon capture on 100Mo

2019

Ordinary muon capture (OMC) on 100Mo is studied both experimentally and theoretically in order to access the weak responses in wide energy and momentum regions. The OMC populates states in 100Nb up to some 50 MeV in excitation energy. For the first time the associated OMC strength function has been computed and compared with the obtained data. The present computations are performed using the Morita-Fujii formalism of OMC by extending the original formalism beyond the leading order. The participant nuclear wave functions are obtained in extended no-core single-particle model space using the spherical version of proton-neutron quasiparticle random-phase approximation (pnQRPA) with two-nucleon…
