Oxidative DNA base damage induced by singlet oxygen and photosensitization: recognition by repair endonucleases and mutagenicity.

We have analyzed the recognition by various repair endonucleases of DNA base modifications induced by three oxidants, viz. [4-(tert-butyldioxycarbonyl)benzyl]triethylammonium chloride (BCBT), a photochemical source of tert-butoxyl radicals, disodium salt of 1,4-etheno-2,3-benzodioxin-1,4-dipropanoic acid (NDPO(2)), a chemical source of singlet oxygen, and riboflavin, a type-I photosensitizer. The base modifications induced by BCBT, which were previously shown to be mostly 7,8-dihydro-8-oxoguanine (8-oxoGua) residues, were recognized by Fpg and Ogg1 proteins, but not by endonuclease IIII, Ntg1 and Ntg2 proteins. DNA damage can significantly modulate expression of the affected genes either by direct structural interference with transcription components or as a collateral outcome of cellular repair attempts. Thus, DNA glycosylases of the base excision repair (BER) pathway have been implicated in negative transcriptional response to several spontaneously generated DNA base modifications, including a common oxidative DNA base modification 8-oxoguanine (8-oxoG). This article addresses a series of hypothesis concerning the octagonal space indicated in Guarino Guarini’s drawings of the Church of the Santissima Annunziata in Messina in his Architettura Civile. The façade of the church was completed around 1660 by Guarino Guarini and presented a «prodigiously high» bell tower at the corner, which collapsed during the 1693 earthquake and was later rebuilt shorter. The entire church was destroyed during the 1908 earthquake. Guarini’s drawings published in his treatise suggest that there must have been an octagonal chapel with columns at the corners under the tower; perhaps it was the baptistery of the church. Il contributo è dedicato all'analisi delle geometrie sottese al progetto della chiesa per i Padri Somaschi di Messina di Guarino Guarini, progetto non datato e mai realizzato, noto grazie alla pubblicazione all'intero del trattato Architettura Civile dell'architetto modenese. L'indagine si avvale del processo di ricostruzione digitale della chiesa, che ha consentito un approfondimento delle geometrie che informano ogni singolo elemento dell'architettura, nonché del constante confronto con le opere esistenti d Guarini, in particolar modo la chiesa di San Lorenzo e la cappella del SS. Sudario di Torino.

The article analyzes some passages of Euripides' Trojan Women, of Aeschylus' Agamemnon and Seven against Thebes, in which the war of Troy as well as the fratricidal war between Eteocles and Polynices is presented as a violent predation described with characteristics of ferociousness particularly shown by cannibalism. The underlying assumption is that the process of bestialization undergone by certain characters when engaged in war or winners of a war, shows, on the contrary, the poet's aim to build models of human and humanity. According to this interpretation the war myths dramatized on the tragic stage turn the tragedies into powerful and effective rites of peace.
