Experimental evolution of an oncolytic vesicular stomatitis virus with increased selectivity for p53-deficient cells

Experimental evolution has been used for various biotechnological applications including protein and microbial cell engineering, but less commonly in the field of oncolytic virotherapy. Here, we sought to adapt a rapidly evolving RNA virus to cells deficient for the tumor suppressor gene p53, a hallmark of cancer cells. To achieve this goal, we established four independent evolution lines of the vesicular stomatitis virus (VSV) in p53-knockout mouse embryonic fibroblasts (p53-/- MEFs) under conditions favoring the action of natural selection. This chapter summarizes some basic notions of thermodynamics and defines the empirical variables which are needed for the description of thermodynamic systems in equilibrium. Empirical temperature and several scales used to measure temperature are defined. The so-called “zeroth law of thermodynamics” is formulated which says that systems which are in mutual equilibrium have the same temperature. European eel (Anguilla anguilla) were exposed to sublethal fenitrothion concentrations in a continuous flow-through system for 4 days. Brain acetylcholinesterase (AChE) activity was evaluated after 2, 8, 12, 24, 32, 48, 56, 72, and 96 h pesticide exposure. Results indicated that AChE activity in eel brains decreased as the concentration of fenitrothion increased. The pesticide induced significant inhibitory effects on the AChE activity ofA. anguilla,ranging from >40% inhibition at a sublethal concentration of 0.02 ppm to >60% inhibition at a sublethal concentration of 0.04 ppm. The formation of zeolites by hydrothermal alteration has been investigated by taking trass from the Laach volcanic area as a sample. Zeolites to be found are chabazite, phillipsite and analcime, all of which originated from the same phonolitic glass. This paper aims at explaining the formation of zeolites by means of experimental alteration of the pumice with various solutions. NaOH and KOH solutions were used in the experiments, these limited the formation conditions of chabazite, phillipsite, analcime in alkaline environments.
