Abstract The flow-injection spectrophotometric determination of nalidixic (I) and pipemidic (II) acids were carried out by reaction with Fe(III) ions entrapped in a polymeric material and filling a packed-bed reactor; the released Fe(III) complexed with the quinolone is monitored at 426 nm. The method worked with a high reproducibility, even when using different reactors, days and solutions. Selectivity of the procedure was tested with different foreign compounds found in human urine as well as in pharmaceutical formulations containing both quinolones; no relevant interferences were observed. The method of chromatin photo-stabilization by the action of visible light in the presence of ethidium bromide was used for investigation of higher-level chromatin structures in isolated nuclei. As a model we used rat hepatocyte nuclei isolated in buffers which stabilized or destabilized nuclear matrix. Several higher-level chromatin structures were visualized: 100 nm globules—chromomeres, chains of chromomeres—chromonemata, aggregates of chromomeres—blocks of condensed chromatin. All these structures were completely destroyed by 2 M NaCl extraction independent of the matrix state, and DNA was extruded from the residual nuclei (nuclear matrices) into a halo. Furosemide assay in pharmaceuticals by Micellar liquid chromatography: study of the stability of the drug.

A simplified high-performance liquid chromatographic procedure is described for the determination of furosemide (4-chloro-N-furfuryl-5-sulphamoylanthranillic acid), which makes use of UV detection, a C18, reversed-phase column, and micellar mobile phases of sodium dodecyl sulphate (SDS) and 1-propanol at pH 3 buffered with phosphate system. The most adequate experimental conditions to handle furosemide solutions in the analytical laboratory are studied. The mixture of furosemide and its degradation products which are formed upon light exposition was resolved with a mobile phase of 0.04 M SDS-2% propanol.
