Crystallization and preliminary X-ray analysis of native and selenomethionyl vinorine synthase from Rauvolfia serpentina.

Vinorine synthase (VS) is a central enzyme of the biosynthesis of the antiarrhythmic drug ajmaline and is a member of the BAHD superfamily of acyltransferases. So far, no three-dimensional structure with significant sequence homology with VS is known. Crystals of VS and selenomethionyl-labelled VS from the medicinal plant Rauvolfia serpentina have been obtained by the hanging-drop technique at 305 K with ammonium sulfate and PEG 400 as precipitants. VS crystals diffract to 2.8 Å and belong to space group P212121, with unit-cell parameters a = 82.3, b = 89.6, c = 136.2 Å. The selenomethionyl VS crystal was nearly isomorphous with the VS crystal.
Selenium Imides: 77Se NMR Investigations of the SeCl2−tBuNH2 Reaction and X-ray Structures of Se3(NtBu)3, tBuNSe(μ-NtBu)2SO2, and tBuNSe(μ-NtBu)2SeO

The reaction of SeCl2 with tert-butylamine in various molar ratios in THF at −78 °C has been investigated by 77Se NMR spectroscopy. In addition to the known Se−N heterocycles Se6(NtBu)2 (1) and Se9(NtBu)6 (2), the acyclic imidoselenium(II) dichlorides ClSe[N(tBu)Se]nCl (4, n = 1; 5, n = 2) and two new cyclic selenium imides [Se3(NtBu)2]n (3, n = 1 or 2) and Se3(NtBu)3 (6) have been isolated and identified. An X-ray analysis shows that 6 is a six-membered ring in a chair conformation with |d(Se−N)| = 1.833 A. Crystal data: 6, trigonal, P3c1, a = 9.8660(3) A, c = 20.8427(7) A, V = 1757.0(1) A3, Z = 6. Recognition of Li+ by a Salophen−UO2 Homodimeric Complex

Self-assembly via mutual U-coordination of the salophen-UO(2) complex 1 creates a dimeric species which is shown to be useful for metal binding. Indeed, the 1 dimer has affinity for alkali metal cations and, interestingly, a marked selectivity for Li(+), determined by electrospray ionization mass spectrometry and (1)H NMR techniques. X-ray diffraction helped in the elucidation of the dimeric complex structure, which presents a crown-ether-type coordination site, in analogy to the more familiar 12-crown-4, responsible for the metal interaction. Comparison with isomer 2, and the salen derivative 3, increases the understanding of the behavior of such systems in solution and in the solid state.
Star-shaped conjugated compounds forming nematic discotic systems

Abstract Star-shaped compounds, having a benzene ( 9a,b ) or a 1,3,5-triazine ( 11a,b ) core and stilbenoid arms were prepared. Hexyloxy chains, attached in the middle of the arms, provide nematic discotic phases ND, which are unusual for such systems. The position of the sidechains prevents the micro-segregation, which is valid for star-shaped discs of columnar phases. The stilbenoid character of 9a,b and 11a,b guarantees a high light sensitivity. Straightforward regio- and diastereoselective synthesis of bi-spirooxindole-engrafted rhodanine analogs 5a–d were achieved by one-pot multicomponent [3 + 2] cycloaddition (32CA) reaction of stabilized azomethine ylide (AYs 3a–d) generated in situ by condensation of L-thioproline and 6-chloro-isatin with (E)-2-(5-(4-chlorobenzylidene)-2,4-dioxothiazolidin-3-yl)-N-(2-morpholinoethyl)acetamide. The benzoxazol-2-yl- substituent was found to act as a removable activating and directing group in the Ir-catalyzed alkylation of C(sp(3))-H bonds adjacent to nitrogen in secondary amines. It can be easily introduced by oxidative coupling or by an SNAr reaction, and it can be removed by hydroxide or by hydride reduction. For 1,2,3,4-tetrahydroisoquinolines, activation exclusively takes place in the 3-position. A variety of activated as well as unactivated terminal olefins are suitable reaction partners.

Resume Dimeric zirconocene and hafnocene dihydrides [(η 5 -RC 5 H 4 ) 2 M(H)(μ-H)] 2 (M = Zr, Hf) react with grey selenium or tellurium to afford the known four-membered bimetallacyclic compounds [(η 5 -RC 5 H 4 ) 2 M(μ-E)] 2 (E = Se, Te) as the sole organometallic products. (R)-(+)-nor-Roefractine (1) was synthesized by the Bischler-Napieralski route, using asymmetric reduction of the 1, 2-didehydro precursor imine with sodium (S)-N-CBZ-prolinyloxyborohydride. Compound 1 was able to displace [3H]-raclopride (a D2 dopamine receptor-selective ligand) from its specific binding sites in rat striatum with selectivity vs [3H]-SCH23390 (D1 dopamine receptor-selective ligand).
Synthesis and monoamine uptake inhibition of conformationally constrained 2β-carbomethoxy-3β-phenyl tropanes

A series of 2beta-carbomethoxy-3beta-phenyl tropanes with conformationally constrained nitrogen substituents were synthesized as potential selective dopamine transporter ligands. These novel compounds were examined for their monoamine uptake inhibition potency at the human dopamine transporter (hDAT), the human serotonin transporter (hSERT) and the human noradrenalin transporter (hNET), stably expressed in human embryonic kidney cells (HEK). A SAR-study was conducted to determine the contribution of extended, 4-fluorinated, conformationally constrained C4 chains at the tropane nitrogen to human monoamine transporter affinity and selectivity.

The stereoselective synthesis of a 2-substituted tetrahydropyran with adjacent alkoxy-bearing stereogenic centre is described. The key steps of this synthesis were the stereoselective epoxidation of an allylic alcohol and the regioselective epoxide ring opening by lithium aluminum hydride. The regio and stereoselective synthesis of a trihydroxyselenide and a trihydroxysulfide is also described. The latter compounds are not suitable for cyclization to tetrahydrofuran ring.
