The fractal calibration method applied to the characterization of polymers in solvent mixtures and in mixed gel packings by SEC.

The size-exclusion chromatographic (SEC) behaviour of different solvent/polymer systems in three packing sets has been analysed from fractal considerations. The three-column sets studied are specifically formed by: (i) 'pure' micro-styragel, (ii) 'mixed' TSK Gel H(HR + XL + HR) and (iii) mixed TSK Gel H(XL + HR + XL). The experimental data reveals that in most of the systems assayed the classical universal calibration (UC) is not fulfilled, denoting the existence of secondary effects accompanying the main SEC mechanism. Formation of micro- and nano-spheric particles (filter dust) during the preparation of cellulose acetate membranes

Abstract Membranes were prepared from six samples of cellulose acetate (CA) differing in their average molecular weight (75–260 kg/mole) and molecular weight distribution using methyl acetate as solvent and 2-propanole as precipitant. The routes through the phase diagram and the evaporation times were varied in these experiments. Electron microscopy demonstrates that the amount of filter dust (CA particles deposited on the membrane surface) decreases as the fraction of low molecular weight material in the starting polymer becomes less. Acid Carboxypeptidases in Grains and Leaves of Wheat, Triticum aestivum L

Extracts of resting and germinating (3 days at 20 degrees C) wheat (Triticum aestivum L. cv Ruso) grains rapidly hydrolyzed various benzyloxycarbonyldipeptides (Z-dipeptides) at pH 4 to 6. Similar activities were present in extracts of mature flag leaves. Fractionation by chromatography on CM-cellulose and on Sephadex G-200 showed that the activities in germinating grains were due to five acid carboxypeptidases with different and complementary substrate specificities. The wheat enzymes appeared to correspond to the five acid carboxypeptidases present in germinating barley (L Mikola 1983 Biochim Biophys Acta 747: 241-252). Fabrication of soft-etched nanoporous polyimide membranes for ionic conduction and discrimination

Abstract Ionic selectivity in nanopores is usually based either on steric or charge exclusion mechanisms. By simultaneously incorporating both mechanisms into a functionalized membrane, an improved control over selectivity can be achieved. We describe the fabrication and experimental characterization of alkali metal cation-selective nanopores in heavy ion-tracked polyimide (PI) membranes using the soft-etching (SE) technique. The latent ion tracks in the PI membrane are selectively dissolved by an organic solvent to form tiny pores without affecting the bulk material. Capillary electroendoosmotic chromatography of peptides

This review focuses on the current state of peptide separation by capillary electroendoosmotic chromatography (CEC). When carried out under optimised conditions, peptide separation by CEC methods represents an orthogonal and complementary technique to micro-HPLC (micro-HPLC) and high-performance capillary zone electrophoresis (HPCZE). The origin of the selectivity differences that can be achieved with these three separation techniques (CEC, micro-HPLC and HPCZE), respectively are discussed, and the current limits of performance with CEC methods documented. Ion size effects on the current efficiency of narrow charged pores

Abstract The effects of ion size on the current efficiency (CE) of charged membranes with narrow pores are studied theoretically. The CE is a measure of the membrane permselectivity defined as the ratio between the counterion flux and the sum of the counterion and coion fluxes when an electric potential difference is applied between the two solutions bathing the membrane. It is studied here as a function of two relevant experimental parameters: the ratio between the ionic radius and the pore radius, and the ratio between the external salt concentration and the membrane fixed charge concentration. Localization and Activity of a Carboxypeptidase in Germinating Seeds of Scots Pine, Pinus sylvestris

Extracts prepared from the endosperm of germinating seeds of Scots pine, Pinus sylvestris L., hydrolysed two typical carboxypeptidase substrates, Z-Phe-Ala and Z-Phe-Phe, with pH optima at 4.2 and 5.0. The activities were completely destroyed by diisopropylfluorophosphate. Identical heat inactivation curves and elution patterns in gel chromatography on Sephadex G-200 suggest that the two activities are due to a single enzyme. In resting seeds very low carboxypeptidase activity was present in both the endosperm and the embryo. Use of reversed-phase C18 Sep-Pak cartridges for the purification and concentration of sepiapterin and other pteridines

Abstract Several pteridines have been tested for their ability to bind to C 18 Sep-Pak. Riboflavin, sepiapterin, deoxysepiapterin, 6-acetyl-7,8-dihydropterin, 6-acetyldihydrohomopterin and 3′-hydroxysepiapterin were strongly retained and all but 6-acetyldihydrohomopterin quantitatively recovered upon elution with 2 ml of methanol. The effect of the concentration and volume of the sample, pH and salt concentration on the retention of sepiapterin have been studied. The procedure was very useful for the purification, desalting, solvent exchange and concentration of pteridines having high affinity for the cartridge.
