Abstract The filamentous fungus Neurospora crassa, a model microbial eukaryote, has a life cycle with many features that make it suitable for studying experimental evolution. However, it has lacked a general tool for estimating relative fitness of different strains in competition experiments. To remedy this need, we constructed N. crassa strains that contain a modified csr-1 locus and developed an assay for detecting the proportion of the marked strain using a post PCR high resolution melting assay. DNA extraction from spore samples can be performed on 96-well plates, followed by a PCR step, which allows many samples to be processed with ease. Furthermore, we suggest a Bayesian approach for…

Marked Neurospora crassa strains for competition experiments and Bayesian methods for fitness estimates

Epigenetic control of phenotypic plasticity in a filamentous fungus Neurospora crassa

2016

AbstractPhenotypic plasticity is the ability of a genotype to produce different phenotypes under different environmental or developmental conditions. Phenotypic plasticity is an ubiquitous feature of living organisms, and is typically based on variable patterns of gene expression. However, the mechanisms by which gene expression is influenced and regulated during plastic responses are poorly understood in most organisms. While modifications to DNA and histone proteins have been implicated as likely candidates for generating and regulating phenotypic plasticity, specific details of each modification and its mode of operation have remained largely unknown. In this study, we investigated how e…

Triterpene glycosides from Blighia welwitschii and evaluation of their antibody recognition capacity in multiple sclerosis

2020

Multiple sclerosis (MS) in a multifactorial autoimmune disease in which reliable biomarkers are needed for therapeutic monitoring and diagnosis. Autoantibodies (autoAbs) are known biomarker candidates although their detection in biological fluids requires a thorough characterization of their associated antigens. Over the past twenty years, a reverse chemical-based approach aiming to screen putative autoantigens has underlined the role of glycans, in particular glucose, in MS. Despite the progress achieved, a lack of consensus regarding the nature of innate antigens as well as difficulties proposing new synthetic glucose-based structures have proved to be obstacles. Here is proposed a strate…

Innovative On-Site Adapted System for Fish Migration with Flow Divider and Glass Collector Basin

2020

Abstract The authors designed a new site-adapted fish passage system for upstream and downstream migration of small and large fish on the urban sector of the Bistriţa River. The longitudinal connectivity of this lotic system is interrupted by numerous transversal hydrotechnical works (weirs). This proposed system was designed to facilitate fish migration, promote fish recolonization of upstream and downstream habitats, allow the expansion of fish range, and increase spawning potential. All components of the proposed system are attached to a concrete girder located to the right stream bank, except a glass basin that is submerged in the riverbed. The positioning of the system in this girder e…

Role of lignin and sodium carbonate on the swelling behavior of black liquor droplets during combustion

2018

AbstractA partial removal of lignin from black liquor (BL) by carbonation and lignin precipitation was studied. In lignin-lean BL droplets during combustion in a laboratory furnace at 800°C in stagnant air, the maximum swelling was decreased. This observation was interpreted as showing that the lignin content decrement is due mainly to removal of higher molar mass (HMM) lignin and that the Na2CO3content of the BL is increased. Stepwise precipitation experiments with industrial softwood and hardwood kraft BLs by carbonation (resulting in pH decrement from 13 to 9) indicated that a fraction of HMM lignin (MM >10 kDa) with a higher amount of carbohydrates precipitated more prominently and e…

Seaport competitiveness research: the past, present and future

2019

This study presents a review of articles with a focus on seaport competitiveness from the maritime literature. We investigated how port competitiveness research has evolved during the last two decades using bibliometric citation analysis tools and techniques. Bibliography data, collected from the ISI Web of Science database, consisted of 267 research papers by 465 authors in 117 journals. Based on citation analysis, we identified the key universities, journals and articles and their impact on port competitiveness research. Also, seven key research streams with few sub-streams were revealed as a result of a mixed co-citation and in-depth content analysis of the most cited articles. A bibliom…

Volatile unsaturated hydrocarbons emitted by seedlings of Brassica species provide host location cues to Bagrada hilaris

2018

Bagrada hilaris Burmeister, is a stink bug native to Asia and Africa and invasive in the United States, Mexico, and more recently, South America. This species can cause serious damage to various vegetable crops in the genus Brassica, with seedlings being particularly susceptible to B. hilaris feeding activity. In this study, the role of volatile organic compounds (VOCs) emitted by seedlings of three Brassica species on the host preference of B. hilaris was evaluated. In dual choice arena and olfactometer bioassays, adult painted bugs preferred B. oleracea var. botrytis and B. napus over B. carinata. Volatiles from B. oleracea seedlings were collected and bioassayed with B. hilaris adults an…

Evidence for ontogenetically and morphologically distinct alternative reproductive tactics in the invasive Round Goby Neogobius melanostomus.

2017

Alternative reproductive tactics are characterized by the occurrence of discrete alternative morphs that differ in behavioural, morphological and physiological traits within the same sex. Although much effort has been made to describe the behaviour, morphology and physiology of such alternative morphs, less effort has been invested investigating how much overlap there is in the characteristics of such morphs in natural populations. We studied random population samples of the invasive Round Goby Neogobius melanostomus from five different localities in the river Rhine system in the Netherlands. We found two morphologically and physiologically distinct male morphs which likely represent altern…
