We compared T2-weighted fast spin-echo (FSE) magnetic resonance (MR) imaging and conventional spin-echo (CSE) T2-weighted imaging in the detection of brain lesions of multiple sclerosis (MS). 20 patients with clinically definite MS underwent brain imaging with both FSE and CSE T2-weighted imaging. There was no significant difference in total number of lesions detected on FSE vs CSE (1354 vs 1378). Slightly more periventricular and subcortical lesions were detected using CSE than FSE, whereas more posterior cranial fossa lesions were detected by FSE. Neurodegeneration plays a major role in multiple sclerosis (MS), in which it is thought to be the main determinant of permanent disability. However, the relationship between the immune response and the onset of neurodegeneration is still a matter of debate. Neuroinflammation by cytotoxic T-lymphocytes impairs retrograde axonal transport in an oligodendrocyte mutant mouse.

Mice overexpressing proteolipid protein (PLP) develop a leukodystrophy-like disease involving cytotoxic, CD8+ T-lymphocytes. Here we show that these cytotoxic T-lymphocytes perturb retrograde axonal transport. Using fluorogold stereotactically injected into the colliculus superior, we found that PLP overexpression in oligodendrocytes led to significantly reduced retrograde axonal transport in retina ganglion cell axons. We also observed an accumulation of mitochondria in the juxtaparanodal axonal swellings, indicative for a disturbed axonal transport. A novel automated segmentation method for retinal layers in OCT images proves retinal degeneration after optic neuritis.
