Inhibition of Protein Isoprenylation Impairs Rho-Regulated Early Cellular Response to Genotoxic Stress

Activation of c-Jun N-terminal kinases (JNKs) and nuclear factor-kappaB (NF-kappaB) are early cellular responses to genotoxic stress involved in the regulation of gene expression. Pretreatment of cells with the hydroxymethyl glutaryl-CoA reductase inhibitor lovastatin blocked stimulation of JNK1 activity by UV irradiation and by treatment with the alkylating compound methyl methanesulfonate but did not affect activation of extracellular signal-regulated kinase 2 by UV light. The organization and dynamics of microtubules (MTs) and the actin cytoskeleton are critical for the correct development and functions of neurons, including intracellular traffic and signaling. In vitro ethanol exposure impairs endocytosis, exocytosis, and nucleocytoplasmic traffic in astrocytes and alters endocytosis in cultured neurons. In astrocytes, these effects relate to changes in the organization and/or function of MTs and the actin cytoskeleton. Lovastatin inhibits Rho-regulated expression of E-selectin by TNFalpha and attenuates tumor cell adhesion.

E-selectin mediated cell-cell adhesion plays an important role in inflammatory processes and extravasation of tumor cells. Tumor necrosis factor-alpha (TNF-alpha) induces E-selectin gene and protein expression in primary human endothelial cells (HUVEC) and in an endothelial cell line (EA.hy-926). As shown by ELISA and FACS analyses, HMG-CoA reductase inhibitors (e.g., lovastatin) impair the TNF-alpha stimulated increase in E-selectin protein expression. The structure of the Chapter is as follows. In Section 2 I discuss some of the factors that may have played a role in causing the crisis and emphasise that supporters of different economic theories will assign different weights to each factor in their analyses. As a consequence, suggested economic policies are highly sensitive to the economic theory employed in evaluating the set of causes. The goal for this workshop is to expose participants to an example of the Experience Workshop Math-Art Movement’s implementation of robot-design and construction in the mathematics classroom. While participants experiment with solving the problem of how to build an omnidirectional robot—a vehicle capable of driving in any direction in a 2D plane—several physical phenomena and properties can be discovered through geometrical measurements and mathematical calculations. This paper presents an experiential learning lab focused on building a digital storytelling using both robots with LEGO Mindstorms and virtual worlds with Kodu Game Lab. The experiential laboratory (32 hours) has involved 33 children of secondary schools in a collaborative learning setting in formal (school labs) and informal learning context (external cultural association). In a first phase of the activity children used Lego Mindstorm robotic kits, assembling them and creating a physical environment (pasterboard arenas) where robots were programmed for moving and taking actions.
