DNA Oxidation Photoinduced by Norharmane Rhenium(I) Polypyridyl Complexes: Effect of the Bidentate N,N′-Ligands on the Damage Profile

Re(I)--polypyridyl complexes have interesting and distinctive photochemical and photosensitizing properties. This work describes the capability to induce (or photoinduce) DNA damage of three Re(I)-complexes with a naturally occurring alkaloid called norharmane (nHo) as ligand: [Re(CO)₃ (nHo)(L)]CF₃ SO₃ where L=2,2'-bipyridine (ReBpy), phenanthroline (RePhen) or dipyrido[3,2-a:2',3'-c]phenazine (ReDppz). The interaction of the complexes with DNA was investigated by steady-state and time-resolved spectroscopy. Comparison of the photoinductive properties of commercial, synthetic and soil-extracted humic substances

Abstract The photochemical behavior of a fulvic acid (FA) and several humic acids (HAs) were studied. These synthetic, commercial or soils-extracted humic substances were compared according to their abilities to sensitize the formation of singlet oxygen and to photoinduce the transformation of aromatic compounds such as 2,4,6-trimethylphenol (TMP) and 1,1-dimethyl-3-phenylurea (fenuron). Singlet oxygen was detected by ESR experiments, and quantum yields of singlet oxygen formation were measured by using 1,2-difuranyl-2-hydroxyethanone (furion) as a probe molecule. Distinct photodynamics of κ-N and κ-C pseudoisomeric iron(ii) complexes

Two closely related FeII complexes with 2,6-bis(1-ethyl-1H-1,2,3-triazol-4yl)pyridine and 2,6-bis(1,2,3-triazol-5-ylidene)pyridine ligands are presented to gain new insights into the photophysics of bis(tridentate) iron(II) complexes. The [Fe(N^N^N)2]2+ pseudoisomer sensitizes singlet oxygen through a MC state with nanosecond lifetime after MLCT excitation, while the bis(tridentate) [Fe(C^N^C)2]2+ pseudoisomer possesses a similar 3MLCT lifetime as the tris(bidentate) [Fe(C^C)2(N^N)]2+ complexes with four mesoionic carbenes. Publisher Summary This chapter discusses the use of repair endonucleases to assess DNA damage by peroxynitrite. Repair endonucleases allow a convenient quantification of various types of oxidative modifications induced by peroxynitrite, both in cultured cells and in cell-free DNA. Halogenated squaraine dyes 1 and 2 possess favorable photophysical and in vitro photobiological properties that make these new class of molecules interesting for photodynamic therapeutic applications. For a better understanding of the mechanism of their photobiological activity, we have analyzed the DNA damage and the cytotoxicity induced by these photosensitizers in mammalian cells and cell-free systems in the presence and absence of various additives and scavengers. Abstract The potential anti-tumor photosensitizer 3,3′-diethyloxadicarbocyanine iodide (DODCI) has been immobilized in dimyristoyl phosphatidylcholine (DMPC) vesicles. Binding of the dicarbocyanine to the lipid vesicles induces several changes in the electronic absorption, emission and excitation spectra of the dye. The monomeric dye molecules exist in at least two forms when immobilized in the DMPC liposomes and the non-luminescing H-aggregates of the dye show efficient excitation energy transfer to the DODCI monomers. Porphyrin and its derivatives as promising photosensitizers have been widely utilized in photodynamic therapy (PDT). However, the water-solubility and biocompatibility of porphyrins are the typical bottlenecks in clinical PDT processes. Photooxidations of Alkenes in Fluorinated Constrained Media:  Fluoro-organically Modified NaY as Improved Reactors for Singlet Oxygen “Ene” Reactions

Creating a stationary fluorinated environment inside the zeolite cavity can increase the reactivity observed for intrazeolite photooxidation of alkenes. Exchanging the zeolite with fluorinated organic cations is a much more effective strategy than simply using a fluorinated solvent for slurry irradiations. Use of cations containing C-F bonds is also more efficient than use of deuterated cations for creation of a singlet oxygen friendly environment where the quenching processes are slowed down. Doping the zeolite with fluoro-organic cation 4 resulted in an increase in the singlet oxygen lifetime to 12 micros.
