Sponges (Porifera) represent the lowest metazoan phylum; they have been shown to be provided with the characteristic metazoan structural and functional molecules. One autapomorphic character of sponges is the presence of high levels of telomerase activity in all cells (or almost all cells, including somatic cells). In spite of this fact previous attempts to cultivate sponge cells remained unsuccessful. It was found that dissociated sponge cells do not replicate DNA and lose their telomerase activity. ABSTRACT The Drosophila ventral midline cells generate a discrete set of CNS lineages, required for proper patterning of the ventral ectoderm. Here we provide the first evidence that the CNS midline cells also exert inductive effects on the mesoderm. Mesodermal progenitors adjacent to the midline progenitor cells give rise to ventral somatic mucles and a pair of unique cells that come to lie dorsomedially on top of the ventral nerve cord, the so-called DM cells. Cell ablation as well as cell transplantation experiments indicate that formation of the DM cells is induced by midline progenitors in the early embryo. We report the embryonic phenotype of muscleblind (mbl), a recently described Drosophila gene involved in terminal differentiation of adult ommatidia. mbl is a nuclear protein expressed late in the embryo in pharyngeal, visceral, and somatic muscles, the ventral nerve cord, and the larval photoreceptor system. All three mbl alleles studied exhibit a lethal phenotype and die as stage 17 embryos or first instar larvae. These larvae are partially paralyzed, show a characteristically contracted abdomen, and lack striation of muscles. Asynchronous replication dynamics of imprinted and non-imprinted chromosome regions in early mouse embryos.

We have used interphase FISH to analyze the replication behavior of four imprinted chromosome regions (Snrpn, Zim1-Peg3, Dlk1-Gtl2, and Igf2r) and five non-imprinted regions in mouse one-cell to morula-stage embryos and embryonic fibroblasts. In general, imprinted chromosome regions showed the expected asynchronous pattern of replication throughout all analyzed stages of preimplantation development and in differentiated cells. The Dlk1-Gtl2 locus which is not expressed and Igf2r which is biallelically expressed in early embryos showed a relaxation of replication asynchrony at the morula stage. Next generation sequencing (NGS) has enabled high throughput discovery of somatic mutations. Detection depends on experimental design, lab platforms, parameters and analysis algorithms. However, NGS-based somatic mutation detection is prone to erroneous calls, with reported validation rates near 54% and congruence between algorithms less than 50%. Here, we developed an algorithm to assign a single statistic, a false discovery rate (FDR), to each somatic mutation identified by NGS. This FDR confidence value accurately discriminates true mutations from erroneous calls. Summary The testes of the newly hatched larva of Oncopeltus fasciatus consist of three different cell types enclosed by the tunica propria: the germline stem cells, the apical cells, and the follicle border cells which are the predecessors of the cyst progenitor cells. There are usually twelve somatically derived apical cells per follicle that no longer divide during postembryonic development. The antigenic properties of somatic (SoP) and excretory-secretory products (ESP) of Echinostoma caproni were studied in experimentally infected rats. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis demonstrated a total of 15 and 9 major polypeptides, in the molecular weight range 10-165 kDa, in SoP and ESP products, respectively. Further analysis revealed some common as well as several unique polypeptides for each type of extract. A total of 2 polypeptides, weighing 58 and 115 kDa, were found in both types of antigen. Indirect enzyme-linked immunosorbent assay analysis using both types of antigens showed that rats developed a weak and slow response in infections with E. caproni. …
