Cloning of Several Genes Coding for Retinoic Acid Nuclear Receptors in the Mouse Embryonal Carcinoma Cell Line PCC7–MZ1
1993
Mouse embryonal carcinoma cell line PCC7-Mz1 can be induced by retinoic acid (RA) to differentiate into several well defined phenotypes of neuroectodermal origin (Lang, E. et al. (1989) J. Cell. Biol. 109, 2481-2493). Several subclones of the cell line (clonal variants) differ from each other in their developmental potential. To test whether these differences in cellular fate are due to somatic mutations in specific genes of these cells, we have cloned full length cDNAs coding for the alpha 1 and beta 2 isoforms, and partial length cDNAs coding for the alpha 2, beta 1 and beta 3 isoforms of the retinoic acid nuclear receptor (RAR). The cloned cDNAs did not differ in sequence from those of n…
Somatic experience of emotion in emotionally focused couple therapy: Experienced trainer therapists' views and experiences.
2021
In general, arousal of emotions is often felt and expressed as a somatic experience in the body. In Emotionally Focused Couple Therapy (EFT), the deepening of emotional experiencing enables therapeutic change. This research explores the experiences of eight experienced EFT trainer therapists regarding their somatic experiences in their work with couples. Using interpretative phenomenological analysis of transcribed in-depth interviews, this research data yielded three core themes that captured therapists' experiences of using their own and couples' somatic experiences in therapy: (1) the importance and relevance of somatic experiences, (2) therapists' use of their own somatic experiences, a…
Interoception moderates the relation between alexithymia and risky-choices in a framing task: A proposal of two-stage model of decision-making.
2021
Decision-making depends on the context (frame) in which questions and alternatives are presented. Moreover, research has showed that the ability to detect bodily sensations (interoception) and being able to attribute these changes to emotions correctly (alexithymia) influence how we make decisions. The aim of the present research was to study how interoception and alexithymia might affect the Framing effect (FE), a cognitive bias closely related to emotional system. 42 healthy participants completed the Risky-choice Framing task and their interoception and alexithymia levels were measured. Results showed that the participants were more risk-taking under the negative frames in comparison to …
The influence of the physical education teacher on intrinsic motivation, self-confidence, anxiety, and pre-and post-competition mood states
2001
One of the fundamental problems facing teachers of physical education (PE) is how to increase pupils' motivation. From the point of view of goal achievement, guidelines need to be established so that information may be used to greater effect in classes. This study examined the relationship between the motivational climate created by the PE teacher and the intrinsic motivation of the preparatory sessions together with self-confidence and anxiety prior to competition and pre- and post-competition mood states. The sample was made up of school children (M age = 11.7) from a state school (N = 115), who, after an introduction to an athletics course of 12 sessions, took part in a sports competitio…
A highly dynamic hot hydrothermal system in the subduction environment: Geochemistry and geochronology of jadeitite and associated rocks of the Sierr…
2021
A U-Pb zircon date of ~113 Ma revealed that a variety of jadeitites and related omphacitite, chloritite and albite-rich rocks from the subduction-related Sierra del Convento block-in-serpentinite-matrix mélange (eastern Cuba) formed nearly synchronously with MORB metabasite-derived anatectic trondhjemitic liquids at high-temperature and pressure in a hot subduction environment. Field, petrologic and geochemical data indicate hydrothermal/metasomatic processes triggered by juvenile fluids likely evolved from the crystallizing hydrous trondhjemitic melts. These fluids, variably mixed with sediment-derived fluids and channelized along fractures in the supra-slab mantle, precipitated relatively…
Confidence-based Somatic Mutation Evaluation and Prioritization
2012
Next generation sequencing (NGS) has enabled high throughput discovery of somatic mutations. Detection depends on experimental design, lab platforms, parameters and analysis algorithms. However, NGS-based somatic mutation detection is prone to erroneous calls, with reported validation rates near 54% and congruence between algorithms less than 50%. Here, we developed an algorithm to assign a single statistic, a false discovery rate (FDR), to each somatic mutation identified by NGS. This FDR confidence value accurately discriminates true mutations from erroneous calls. Using sequencing data generated from triplicate exome profiling of C57BL/6 mice and B16-F10 melanoma cells, we used the exist…
Differentiation and ultrastructure of the spermatogonial cyst cells in the milkweed bug,Oncopeltus fasciatus
2002
Summary The testes of the newly hatched larva of Oncopeltus fasciatus consist of three different cell types enclosed by the tunica propria: the germline stem cells, the apical cells, and the follicle border cells which are the predecessors of the cyst progenitor cells. There are usually twelve somatically derived apical cells per follicle that no longer divide during postembryonic development. Their fine structural organization remains constant from hatching until death from old age. The apical complex cells are surrounded by at least four germline stem cells which all send cell projections toward the apical complex cells. A testicular follicle includes at least four border cells at the fir…
Immunological Characterization of Somatic and Excretory–Secretory Antigens of Echinostoma caproni (Trematoda: Echinostomatidae) in Experimentally Inf…
2004
The antigenic properties of somatic (SoP) and excretory-secretory products (ESP) of Echinostoma caproni were studied in experimentally infected rats. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis demonstrated a total of 15 and 9 major polypeptides, in the molecular weight range 10-165 kDa, in SoP and ESP products, respectively. Further analysis revealed some common as well as several unique polypeptides for each type of extract. A total of 2 polypeptides, weighing 58 and 115 kDa, were found in both types of antigen. Indirect enzyme-linked immunosorbent assay analysis using both types of antigens showed that rats developed a weak and slow response in infections with E. caproni. …

AbstractReprogramming of mouse somatic cells into induced pluripotent stem cells (iPSCs) often generates partially reprogrammed iPSCs (pre-iPSCs), low-grade chimera forming iPSCs (lg-iPSCs) and fully reprogrammed, high-grade chimera production competent iPSCs (hg-iPSCs). Lg-iPSC transcriptome analysis revealed misregulated Dlk1-Dio3 cluster gene expression and subsequently the imprinting defect at the Dlk1-Dio3 locus. Here, we show that germ-cell marker Dppa3 is present only in lg-iPSCs and hg-iPSCs, and that induction with exogenous Dppa3 enhances reprogramming kinetics, generating all hg-iPSCs, similar to vitamin C (Vc). Conversely, Dppa3-null fibroblasts show reprogramming block at pre-i…
Sponges (Porifera) model systems to study the shift from immortal to senescent somatic cells: the telomerase activity in somatic cells.
1998
Abstract Sponges (Porifera) represent the lowest metazoan phylum, characterized by a pronounced plasticity in the determination of cell lineages. In a first approach to elucidate the molecular mechanisms controlling the switch from the cell lineage with a putative indefinite growth capacity to senescent, somatic cells, the activity of the telomerase as an indicator for immortality has been determined. The studies were performed with the marine demosponges Suberites domuncula and Geodia cydonium . It was found that the activity for the telomerase in the tissue of both sponges is high; a quantitative analysis revealed that the extract from S. domuncula contained 10.3 TPG units per 5000 cell e…
