Construction of a Trp commercial baker?s yeast strain by using food-safe-grade dominant drug resistance cassettes

We have designed a food-safe-grade module for gene disruptions in commercial baker's yeast strains, which contains the G418 resistance cassette, KanMX4, flanked by direct repeats from the MEL1 gene of Saccharomyces cerevisiae. This module was used to obtain a Trp(-) auxotrophic mutant of the polyploid HY strain by successive targeting to the TRP1 locus and later in vivo excision of the kan(r) marker. Southern blot analysis indicated that HY contains five copies of the TRP1 gene. However, after four disruption rounds, a strain named HYtrpM(4), unable to grow in the absence of tryptophan, was selected. Tirant: A new retrotransposon-like element inDrosophila melanogaster

In this paper we report a new retrotransposon-like element of Drosophila melanogaster called Tirant. This sequence is moderately repeated in the genome of this species and it has been found to be widely dispersed throughout its distribution area. From Southern blot and in situ analyses, this sequence appears to be mobile in D. melanogaster, since its chromosome location and the hybridization patterns vary among the different strains analyzed. In this way, partial sequencing of Tirant ends suggests that it is a retrotransposon, since it is flanked by two LTRs. Clinical and biochemical studies have revealed a great phenotypic variability in mucopolysaccharidosis type II (Hunter disease), probably due to different mutations in the IDS gene that has been localized in Xq28. Using a cDNA probe containing almost the entire coding region of the human IDS gene, we performed a molecular analysis on 7 patients with Hunter disease. In one patient, a complete deletion of the IDS coding sequences was found. Another patient had structural alterations of the IDS gene including a partial deletion. The primary aim of the insulin-dependent diabetes mellitus (IDDM) component of Genetic Analysis Workshop 5 (GAW5) was to collect and analyze new data on DNA polymorphisms closely linked to the HLA-D region and the insulin gene. The probes and restriction enzymes described here were used by all ten participating labs, and the data from Southern blotting were interpreted and reported according to conventions developed for the Workshop. These DNA data on members of 94 families with two or more IDDM sibs constitute the largest such sample available. The data were used in most of the analyses presented at the Workshop meeting, and are available on request.

A Southern analysis of genomic DNA using Drosophila melanogaster probes for the major heat shock protein genes (Hsp82, Hsp 70, Hsps encoding small proteins) was made to study the phylogenetic relationships between three Drosophila species belonging to the obscura group (D. subobscura, D. guanche, and D. madeirensis). The phylogenetic trees showed that D. madeirensis and D. subobscura are the most closely related species in the triad, while D. guanche is the most distantly related one. As in other Drosophila species, Hsp82 is a single copy gene in D. subobscura, D. guanche, and D. madeirensis, while Hsp 70 and Hsps, which encode small proteins, are genie families. Distribution of gypsy sequences in Drosophila species of the obscura subgroup.

Eight Drosophila species of the obscura subgroup were screened for sequences homologous to the gypsy retrotransposon of D. melanogaster. Molecular characterization of gypsy sequences was first approached through digesting genomic DNAs from these obscura species with appropriate restriction enzymes and subjecting them to Southern blot analysis. The results of this analysis indicate that gypsy-homologous sequences are well conserved among species of the obscura subgroup. With the exception of D. guanche, all other species bear a 7 kb Xho I fragment that represents the complete element in D. melanogaster. The precise excision of the transposon simultaneously rescued all these defects whereas its unprecise excision created new pros(V) alleles, including the null allele pros(V17). Here, we describe the relationship between the genetic structure of pros locus, larval locomotion, and larval gustatory response. The rodents of the Pirro Nord fauna (Foggia, Southern Italy)

The rodent fauna of various fissure fillings of the Pirro Nord karstic complex are examined here. They have been collected during several field caingns starting from the 70’s until 2006 and are here examined in a comprehensive matter. Hundreds of specimens of arvicolids, murids and, in much lower amount, Glirids have been recovered from several of these fissures. They belong to the species Allophaiomys ex gr. ruffoi, Myodes sp. Apodemus flavicollis and Muscardinus sp.
Intensity and spatial extent of droughts in southern Africa

International audience; The standardized precipitation index allows for monitoring the intensity and spatial extent of droughts at different time scales. We used it to do a retrospective analysis of the spatial extent of droughts in Southern Africa (South of 10°S), from 1901 to 1999. Accordingly, the 8 most severe droughts at the 6-month scale (October–April) for the summer rainfall region of Southern Africa ended in 1916, 1924, 1933, 1949, 1970, 1983, 1992 and 1995. At the 2-year scale, they ended in 1906, 1933, 1983, 1984, 1992, 1993, 1995 and 1996. Areas affected by those droughts ranged from 3.4 to 2 106 km2. Eight of those 12 years are El Niño years. We have cloned from a cDNA library of neuronal derivatives of retinoic-acid-induced embryonic carcinoma cells a nuclear receptor that may be involved in the control of late neurogenesis and early neuronal differentiation. The receptor which is practically identical in sequence with germ cell nuclear factor, has been designated neuronal cell nuclear factor (NCNF). NCNF is exclusively expressed in the neuronal derivatives of PCC7-Mz1 cells, with the expression beginning within hours of exposure to retinoic acid.
