SNAT2 silencing prevents the osmotic induction of transport system A and hinders cell recovery from hypertonic stress.

AbstractUnder hypertonic conditions the induction of SLC38A2/SNAT2 leads to the stimulation of transport system A and to the increase in the cell content of amino acids. In hypertonically stressed human fibroblasts transfection with two siRNAs for SNAT2 suppressed the increase in SNAT2 mRNA and the stimulation of system A transport activity. Under the same condition, the expansion of the intracellular amino acid pool was significantly lowered and cell volume recovery markedly delayed. It is concluded that the up-regulation of SNAT2 is essential for the rapid restoration of cell volume after hypertonic stress.

A growing demand for mobility and the deficiencies in the transport system have transformed urban centres in “unliveable” places. A prevalent use of private cars produces high levels of pollution, casualties and road congestion. The promotion of public transport is considered an effective strategy to make mobility more sustainable for people. When a public transport system serves areas with a weak transportation demand, as it happens in territories with dispersed settlements (urban outskirts, rural areas, mountains, …), the resort to unconventional forms of public transport, such as dial and ride bus services, is necessary.

Member 4 of human solute carrier family 7 (SLC7A4) exhibits significant sequence homology with the SLC7 subfamily of human cationic amino acid transporters (hCATs) [Sperandeo, Borsani, Incerti, Zollo, Rossi, Zuffardi, Castaldo, Taglialatela, Andria and Sebastio (1998) Genomics 49, 230–236]. It is therefore often referred to as hCAT-4 even though no convincing transport activity has been shown for this protein.
