A new dual inhibitor of arachidonate metabolism isolated from Helichrysum italicum.

2003

Six acetophenones (1-6) and one gamma-pyrone (7), previously isolated from Helichrysum italicum, were tested for their ability to inhibit enzymatic and non-enzymatic lipid peroxidation, the stable 1,1-diphenyl-2-pycryl-hydrazyl free radical, superoxide scavenging and arachidonic acid metabolism. In addition, they were studied in different experimental models such as the chronic inflammation induced by 12-O-tetradecanoylphorbol 13-acetate (TPA), the phospholipase A(2)-induced mouse paw oedema test, the carrageenan-induced mouse paw oedema test, and the writhing induced by acetic acid in the mouse. Of the assayed compounds, only 1 inhibited enzymatic lipid peroxidation but had no effect on no…

Anti-inflammatory activity of erycristagallin, a pterocarpene from Erythrina mildbraedii.

2003

Erycristagallin, a pterocarpene isolated from Erythrina mildbraedii, was tested in vitro for its antioxidant properties on the stable 2,2-diphenyl-1-pycryl-hydrazyl (DPPH) free radical and on the arachidonic acid metabolism. In addition, erycristagallin was tested on different experimental models of inflammation, such as the acute and chronic inflammation induced by the application of 12-O-tetradecanoylphorbol 13-acetate (TPA) on mice and the phospholipase A(2)-induced mouse paw oedema test. In the carrageenan-induced mouse paw oedema test, the ethyl acetate extract obtained from E. mildbraedii showed anti-inflammatory activity, and erycristagallin was isolated as the active principle. In v…

Assessment of the anti-inflammatory activity and free radical scavenger activity of tiliroside

2003

Three flavonoids, gnaphaliin, pinocembrin and tiliroside, isolated from Helichrysum italicum, were studied in vitro for their antioxidant and/or scavenger properties and in vivo in different models of inflammation. In vitro tests included lipid peroxidation in rat liver microsomes, superoxide radical generation in the xanthine/xanthine oxidase system and the reduction of the stable radical 1,1-diphenyl-2-pycryl-hydrazyl (DPPH). Acute inflammation was induced by application of 12-O-tetradecanoylphorbol 13-acetate (TPA) to the mouse ear or by subcutaneous injection of phospholipase A(2) or serotonin in the mouse paw. Eczema provoked on the mouse ear by repeated administration of TPA was selec…

Opposite vascular activity of (R)-apomorphine and its oxidised derivatives. Endothelium-dependent vasoconstriction induced by the auto-oxidation meta…

2003

We have synthetised a series of oxidised apomorphine derivatives (orto and para quinones 2-5), in order to analyse their vascular activity. We have performed radioligand binding assays on rat cortical membranes and functional studies on rat aortic rings. Instead the relaxant activity exhibited by (R)-apomorphine, o-quinones 2, 4, show contractile activity dependent on endothelium in rat aortic rings. Compound 2, the main metabolite of (R)-apomorphine auto-oxidation, was the product which showed enhanced contractile activity by a complex mechanism related to activation of Ca(2+) channels through release and/or inhibition of endothelial factors. Moreover, this compound disrupts the endothelia…

Vascular Activity of (-)-Anonaine, (-)-Roemerine and (-)-Pukateine, Three Natural 6a(R)-1,2-Methylenedioxyaporphines with Different Affinities for α1…

2004

We have studied the mechanism of action of three 6a( R)-1,2-methylenedioxyaporphines as vasorelaxant compounds. The alkaloids assayed showed different affinities for the three human cloned alpha (1)-adrenoceptor (AR) subtypes stably expressed in rat-1 fibroblasts, showing lower affinity for alpha(1B)-AR with regard to the alpha(1A)- or alpha(1D)-subtypes. These three natural compounds are more potent inhibitors of [ (3)H]-prazosin binding than of [ (3)H]-diltiazem binding to rat cerebral cortical membranes. As all these alkaloids inhibited noradrenaline (NA)-induced [ (3)H]-inositol phosphate formation in cerebral cortex and rat tail artery, they may be safely viewed as alpha (1)-AR antagon…

A possible structural determinant of selectivity of boldine and derivatives for the alpha 1A-adrenoceptor subtype.

1996

1. The selectivity of action of boldine and the related aporphine alkaloids, predicentrine (9-O-methylboldine) and glaucine (2,9-O-dimethylboldine) and alpha 1-adrenoceptor subtypes was studied by examining [3H]-prazosin competition binding in rat cerebral cortex. WB 4101 and benoxathian were used as selective alpha 1A-adrenoceptor antagonists. 2. In the competition experiments [3H]-prazosin (0.2 nM) binding was inhibited by WB 4101 and benoxathian. The inhibition curves displayed shallow slopes which could be subdivided into high and low affinity components (pKi = 9.92 and 8.29 for WB 4101, 9.35 and 7.94 for benoxathian). The two antagonists recognized approximately 37% of the sites with h…

Ontogeny of Heterocypris bosniaca (Ostracoda: Cyprididae): description of postembryonic instars and rediscovery of the neglected A-9 stage

2013

Despite being the most common and widespread family of Ostracoda in fresh water, the Cyprididae are still poorly known with respect to species ontogeny. The full series of eight juvenile instars has been described in detail for just five of the approximately 1000 living species, and only in one case did this include the earliest A-9 stage. We aimed to fill this gap with a morphological study of the post-embryonic development of Heterocypris bosniaca Petkowski, Scharf and Keyser, 2000, a species with unusual morphological traits within the genus, including putatively paedomorphic characters such as valves without tubercles and the narrow calcified inner lamella on both valves. Inside the egg…

Figures of equilibrium in close binary systems

1992

The equilibrium configurations of close binary systems are analyzed. The autogravitational, centrifugal and tidal potentials are expanded in Clairaut's coordinates. From the set of the total potential angular terms an integral equations system is derived. The reduction of them to ordinary differential equations and the determination of the boundary conditions allow a formulation of the problem in terms of a single variable.

Influence of Physical Treatments on the Potato Starch Granules Micro- and Ultrastructure

2012

In this research, potato starch was subjected to ph ysical treatments in order to analyse the changes at the micro- and ultrastructural levels an d finally to identify the optimal conditions for disintegrating the starch granules. For the analysi s, 10% aqueous solutions of potato starch were treated with a) microwaves; b) heat and c) heat com bined with ultrasounds. For each treatment type, the duration was 5 minutes, excepting the microwave treatment, where the time was 1 and 5 minutes; heat power varied from 180W to 900W at the tests with microwave, temperature varied between 65 o C and 100 o C in the thermal experiments and at the thermal ana lysis combined with ultrasounds temperature …
