Amphetamine-type stimulants analysis in oral fluid based on molecularly imprinting extraction
2018
Abstract A methamphetamine-based molecularly imprinted polymer (MIP) has been prepared by bulk polymerization to recognize new psychoactive substances (NPS) of the amphetamine, cathinones and 2C families in oral fluid samples, being the first precedent of a synthetized MIP for the extraction and preconcentration 32 NPS including amphetamine type substances and synthetic cathinones from oral fluids. Pre-polymerization complex and resulting materials were appropriately characterized by infrared spectroscopy, scanning electron microscopy, and nitrogen adsorption-desorption isotherms. Appropriateness of the material for the specific recognition of the target analytes was also evaluated through …
Application of solid-phase microextraction combined with derivatization to the enantiomeric determination of amphetamines.
2005
Abstract The utility of combining chiral derivatization and solid-phase microextraction (SPME) for the enantiomeric analysis of primary amphetamines by liquid chromatography has been investigated. Different derivatization/extraction strategies have been evaluated and compared using the chiral reagent o -phthaldialdehyde (OPA)– N -acetyl- l -cysteine (NAC) and fibres with a Carbowax-templated resin coating. Amphetamine, norephedrine and 3,4-methylenedioxyamphetamine (MDA) were used as model compounds. On the basis of the results obtained, a new method is presented based on the derivatization of the analytes in solution followed by SPME of the OPA–NAC derivatives formed. The proposed conditio…
Chiral determination of amphetamine and related compounds using chloroformates for derivatization and high-performance liquid chromatography
1999
The enantiomeric determination of amphetamine and various amphetamine-type compounds by liquid chromatography after chiral derivatization with 9-fluorenylmethyl chloroformate-L-proline (FMOC-L-Pro) is reported. The results obtained were compared with those achieved after achiral derivatization with 9-fluorenylmethyl chloroformate and subsequent separation of the derivatives on a beta-cyclodextrin chiral stationary phase. Conditions for the derivatization of amphetamines with FMOC-L-Pro were investigated, including the effect of the derivatization reagent concentration, pH and reaction time, using amphetamine, ephedrine and pseudoephedrine as model compounds. On the basis of these studies, p…
The pharmacological and non-pharmacological treatment of attention deficit hyperactivity disorder in children and adolescents: A systematic review wi…
2017
Background Attention deficit hyperactivity disorder (ADHD) is one of the most commonly diagnosed psychiatric disorders in childhood. A wide variety of treatments have been used for the management of ADHD. We aimed to compare the efficacy and safety of pharmacological, psychological and complementary and alternative medicine interventions for the treatment of ADHD in children and adolescents. Methods and findings We performed a systematic review with network meta-analyses. Randomised controlled trials (≥ 3 weeks follow-up) were identified from published and unpublished sources through searches in PubMed and the Cochrane Library (up to April 7, 2016). Interventions of interest were pharmacolo…
'Up-regulation of histone acetylation induced by social defeat mediates the conditioned rewarding effects of cocaine
2016
Social defeat (SD) induces a long-lasting increase in the rewarding effects of psychostimulants measured using the self-administration and conditioned place procedures (CPP). However, little is known about the epigenetic changes induced by social stress and about their role in the increased response to the rewarding effects of psychostimulants. Considering that histone acetylation regulates transcriptional activity and contributes to drug-induced behavioral changes, we addressed the hypothesis that SD induces transcriptional changes by histone modifications associated with the acquisition of place conditioning. After a fourth defeat, H3(K9) acetylation was decreased in the hippocampus, whil…
Experiencing Effects of Cocaine and Speed with Self-Regulation Therapy.
2015
AbstractThis study demonstrates the efficacy of Self-Regulation Therapy (SRT) to induce effects of cocaine and speed in a single session. SRT is a suggestion procedure of sensorial recall exercises (salivation, feeling of weight, tension, etc.) that increases the capacity to reproduce all sensation types and those that drugs produce. The Self-Regulation Scale (SRS) measures this capacity. Four groups participated, formed according to drug use: Group 1 (uses no illegal drugs); Group 2 (experimentally uses cannabis only); Group 3 (moderate drug users); Group 4 (regular drug users, especially stimulants). All four groups participated in an SRT session to induce relaxation. No differences in th…
Influence of the Novelty-Seeking Endophenotype on the Rewarding Effects of Psychostimulant Drugs in Animal Models
2015
Novelty seeking (NS), defined as a tendency to pursue novel and intense emotional sensations and experiences, is one of the most relevant individual factors predicting drug use among humans. High novelty seeking (HNS) individuals present an increased risk of drug use compared to low novelty seekers. The NS endophenotype may explain some of the differences observed among individuals exposed to drugs of abuse in adolescence. However, there is little research about the particular response of adolescents to drugs of abuse in function of this endophenotype, and the data that do exist are inconclusive. The present work reviews the literature regarding the influence of NS on psychostimulant reward…
Coffee increases state anxiety in males but not in females.
2003
Coffee, reproducing the conditions under which caffeine is normally ingested, containing 3, 75, 150 or 300 mg of caffeine was given to healthy male and female volunteers. 25-30 min after drinking the beverage, they completed the Spanish version of the state-trait anxiety inventory (STAI). The beverage increased state anxiety, in a dose-dependent manner, in males but not in females. This could be due to a lesser sensitivity of females to coffee.
Behavioural and neurochemical effects after repeated administration of N-ethylpentylone (ephylone) in mice
2022
N-ethyl-pentylone (NEP), also known as "ephylone" and N-ethylnorpentylone, has been identified as one of the most recent novel psychostimulants to emerge into the illicit drug market and it has been associated with some intoxications and even fatalities. However, little is known about the consequences of its repeated consumption as well as the role of the monoaminergic system in such consequences. Thus, the aim of our study was to investigate the neurochemical profile and the behavioural effects after both acute and repeated NEP exposure. Male OF1 mice were acutely (1, 3, 10 mg/kg, i.p.) or repeatedly (1, 3, 10 mg/kg, i.p., 5 days, twice/day) exposed to NEP, and anxiety-like behaviour, aggr…
