Spatial and temporal heterogeneity of ventilator-associated lung injury after surfactant depletion.
2008
Volutrauma and atelectrauma have been proposed as mechanisms of ventilator-associated lung injury, but few studies have compared their relative importance in mediating lung injury. The objective of our study was to compare the injury produced by stretch (volutrauma) vs. cyclical recruitment (atelectrauma) after surfactant depletion. In saline-lavaged rabbits, we used high tidal volume, low respiratory rate, and low positive end-expiratory pressure to produce stretch injury in nondependent lung regions and cyclical recruitment in dependent lung regions. Tidal changes in shunt fraction were assessed by measuring arterial Po2 oscillations. After ventilating for times ranging from 0 to 6 h, lu…
Calorimetric and Volumetric Investigations of the Effect of the Hydrophobicity of the Surfactant on the Binding between (Ethylene oxide)13-(propylene…
2004
The enthalpy and the volume of transfer (DeltaY(t)) of the unassociated (ethylene oxide)(13)-(propylene oxide)(30)-(ethylene oxide)(13) (L64) from water to the aqueous sodium alkanoate solutions as functions of the surfactant concentrations (m(S)) were determined at 298 K. The surfactants studied are sodium hexanoate, sodium heptanoate, sodium octanoate, sodium undecanoate, and sodium dodecanoate. As a general feature, for the short alkyl chain surfactants, DeltaY(t) describes an S-shaped curve in the range of m(S) analyzed whereas for the more hydrophobic surfactants the DeltaY(t) vs m(S) trends exhibit maxima which appear at ms values very close to the critical micellar concentration in w…
Survey of neonatal respiratory care and surfactant administration in very preterm infants in the Italian neonatal network
2013
INTRODUCTION: Variation of respiratory care is described between centers around the world. The Italian Neonatal Network (INN), as a national group of the Vermont-Oxford Network (VON) allows to perform a wide analysis of respiratory care in very low birth weight infants. METHODS: We analyzed the dataset of infants enrolled in the INN in 2009 and 2010 and, for surfactant administration only, from 2006 to 2010 from 83 participating centers. All definitions are those of the (VON). A questionnaire analysis was also performed with a questionnaire on centers practices. RESULTS: We report data for 8297 infants. Data on ventilator practices and outcomes are outlined. Variation for both practices and…
Humoral autoreactivity directed against surfactant protein-A (SP-A) in rheumatoid arthritis synovial fluids.
2000
SUMMARY SP-A is found principally in the lung, and has been associated with lamellar bodies also found in the synovial joint. Both SP-A and C1q contain collagen-like regions, and SP-A and C1q have some structural similarities, both having a globular head region and a collagen-like tail. Here we are able to show that (i) autoreactivity to SP-A, as expressed by IgG and IgM autoantibodies, is present in synovial fluid (SF) isolated from patients with rheumatoid arthritis (RA); (ii) in absorption experiments only a limited degree of cross-reactivity between autoantibodies reactive with C1q and SP-A is observed; (iii) there is no cross-reactivity between autoantibodies reactive with type II coll…
Kinetic evidence for the solubilization of pyridine-2-azo-p-dimethylaniline in alkanediyl-α,ω-bis(dimethylcetylammonium nitrate) surfactants. Role of…
2004
The incorporation of the bidentate ligand pyridine-2-azo-p-dimethylaniline (PADA) into micellar aggregates of the dimeric cationic surfactants propanediyl-, hexanediyl- and dodecanediyl-alpha,omega-bis(dimethylcetylammonium nitrate) (16-3-16,2NO(3)(-), 16-6-16,2NO(3)(-) and 16-12-16,2NO(3)(-), respectively) has been studied at 25 degreesC by examining the kinetics of the complexation reaction of the Ni(II) ion with this ligand. For comparison, cetyltrimethylammonium nitrate ( CTAN), which can be considered as the "monomeric'' surfactant of 16-3-16,2NO(3)(-), has also been used. The kinetic data have shown that, for 16-3-16,2NO(3)(-) and CTAN, at a surfactant concentration below the critical…
ABC A-subclass proteins: Gatekeepers of cellular phospho- and sphingolipid transport
2007
During the past years, available evidence suggests that members of a novel family of structurally highly related multispan proteins, designated ABC A-subclass transporters, exert critical functions in the control of cellular lipid transport processes. Loss-of-function scenarios, thus far, have revealed pivotal roles of individual ABC A-transporters in specialized lipid secretory pathways of the cell including HDL biogenesis (ABCA1), lung surfactant production (ABCA3), retinal integrity (ABCA4/ABCR) and skin lipid barrier formation (ABCA12). Although the specific transporter activities of many members of this novel protein family have not yet been established in detail, available evidence in…
Effect of Cholesterol on Electrostatics in Lipid−Protein Films of a Pulmonary Surfactant
2010
We report the changes in the electrical properties of the lipid-protein film of pulmonary surfactant produced by excess cholesterol. Pulmonary surfactant (PS) is a complex lipid-protein mixture that forms a molecular film at the interface of the lung's epithelia. The defined molecular arrangement of the lipids and proteins of the surfactant film gives rise to the locally highly variable electrical surface potential of the interface, which becomes considerably altered in the presence of cholesterol. With frequency modulation Kelvin probe force microscopy (FM-KPFM) and force measurements, complemented by theoretical analysis, we showed that excess cholesterol significantly changes the electri…
Surface Characteristics Control the Attachment and Functionality of (Chimeric) Avidin
2018
The physical adsorption (physisorption) of proteins to surfaces is an important but incompletely understood factor in many biological processes and is of increasing significance in bionanotechnology as well. Avidin is an important protein because of strong avidin–biotin binding, which has been exploited in numerous applications. We have undertaken thorough experimentation on the physisorption of avidin, to chemically different flat surfaces of Si and graphite and also to the curved version of the latter, on multiwalled carbon nanotubes (MWNTs) of different diameters. The difference in the behavior of avidin on Si versus graphite is drastic; on Si, avidin deposits as single globular tetramer…
2D dynamical arrest transition in a mixed nanoparticle-phospholipid layer studied in real and momentum spaces
2015
AbstractWe investigate the interfacial dynamics of a 2D self-organized mixed layer made of silica nanoparticles interacting with phospholipid (DPPC) monolayers at the air/water interface. This system has biological relevance, allowing investigation of toxicological effects of nanoparticles on model membranes and lung surfactants. It might also provide bio-inspired technological solutions, exploiting the self-organization of DPPC to produce a non-trivial 2D structuration of nanoparticles. The characterization of interfacial dynamics yields information on the effects of NPs on the mechanical properties, important to improve performances of systems such as colloidosomes, foams, creams. For thi…
Highly recyclable surfactant-based supramolecular eutectogels for iodine removal
2022
In this work, we screened several surfactants for their ability to harden Eutectic Mixtures. We found cetyldiethanolamine-N-oxide as the best candidate, which was able to form eutectogel in four eutectic mixtures. Characterization of the gels obtained in terms of thermal stability, rheology and gelation kinetics, showed a prominent influence of the nature of the hydrogen bonding accepting and donating components for the first two parameter and the other one, respectively. Then, we assessed the eutectogels for their ability to adsorb I2, as a model for its radioactive nuclides, from apolar solvent solutions. The best performing gel allowed fast and thorough removal of I2, over a broad range …
